Here we report a two-component optical silencer system comprising photoactivated adenylyl cyclases (PACs) and the small cyclic nucleotide-gated potassium channel SthK. Activation of this PAC-K silencer by brief pulses of low-intensity blue light causes robust and reversible silencing of cardiomyocyte excitation and neuronal firing. In vivo expression of PAC-K in mouse and zebrafish neurons is well tolerated, where blue light inhibits neuronal activity and blocks motor responses. Following the bpacnz campaign Laboratory Testing in Diabetes, we were asked why fasting glucose was the best initial test for diagnosing diabetes, and the role of the post prandial glucose. We contacted Rick Cutfield for a comparison and explanation of the role of these tests.. Screening guidelines throughout the world recommend fasting glucose as the best test, mainly because of standardisation, reproducibility and reasonable correlation with the oral glucose tolerance test. However there are some problems with this. The specificity of a fasting glucose greater than 7.0 mmol/L is over 95% but sensitivity is only about 50%. It is less reliable and sensitive in the elderly and not reliable in all ethnic groups. As a result some people with fasting glucose of 5.5 - 6.0 mmol/L will have diabetes on further testing.. Natural hydrocarbon seeps represent a habitat for microbial communities that might provide the molecular tool kit for sustainable strategies to reduce the negative impact of oil spills. They also are a persistent source of methane (CH4) [16], a greenhouse gas whose climate warming potential is 25 times greater than that of CO2 [48]. Biological CH4 oxidation in the marine ecosystem has been well documented and identified as a CH4 sink of global significance [49-51]. Anaerobic oxidation of methane (AOM), mediated by microbiomes associated with ocean sediments and deposits, has been proposed as the dominant biological process responsible for the removal of ,300 Tg CH4 per year from the ocean [52,53]. This presentation summarizes the first study of hyperbaric oxygen chemo-radiation sensitization for locally advanced squamous cell carcinomas of the oropharynx. It took the form of a Stage I dose escalation trial, designed to determine safety, feasibility and tolerability when hyperbaric oxygen was added to standard care, namely intensity modulated radiation therapy and cisplatinum chemotherapy. The presentation will describe the biological plausibility and physiologic basis for pre-radiation hyperbaric hyperoxia, and the rationale for selection of this tumor type and tumor grade. The hyperbaric dosing regimen, one based upon previous human tumor oxygen response curves, is discussed, and the critical time window for radiation therapy beam on from exiting the chamber discussed.
