The environmental information an organism receives during early development is integrated through epigenetic mechanisms, such as DNA methylation. The gene expression or the ability to respond to environmental factors experienced later in life may be altered by this process. We have focused our studies on two types of environmental factors that confer changes in DNA methylation: temperature and farming conditions. First, we studied temperature effects on DNA methylation of the aromatase promoter, the enzyme that converts androgens into estrogens and is essential for ovarian development, in 1-year old sea bass. We showed that males had higher DNA methylation levels than females and that high temperature led to increased DNA methylation levels in females, leading a portion of them to differentiate into males. To our knowledge, studies on the heritability of cortisol response to stress in fish have been limited to salmonids [27-29] and cyprinids [21], which limits generalizations. In addition, lines with high and low cortisol response to stress have been selected in rainbow trout [29]. However, the heritability of cortisol response to stress appears to be variable even among related species: 0.27-0.50 in rainbow trout, Oncorhynchus mykiss[27, 29, 30] and 0.60 in carp [21], but only 0.05 in Atlantic salmon Salmo salar[27, 31]. The in vitro approach encompasses the development, characterization, and validation of a LMB primary hepatocyte model. Once this tool is developed, I plan to characterize dose and time response for Vtg and ER mRNA induced by E2, EE2, and MXC. This will be followed by Vtg protein analysis. Similar experiments examining dose and time response also will be conducted in vivo to compare cell-specific responses to that as seen in the whole animal. Scale loss is a common occurrence in both wild fish and those in aquaculture production systems, and regeneration of scales has been described in several freshwater species. Relatively little information exists about this process in marine fish, and in the present study, the chronology of scale regeneration was characterized in juvenile sea bass Dicentrarchus labrax, maintained in full seawater (SW; 36‰, 11.2 mM Ca(2+)) or brackish water (BW; 3.5‰, 1.1 mM Ca(2+)). Despite the significant differences in plasma osmolality, plasma calcium (Ca) and phosphorus (P) were similar between SW and BW. Intersex occurrence in freshwater fishes was evaluated for nine river basins in the United States. Testicular oocytes (predominantly male testes containing female germ cells) were the most pervasive form of intersex observed, even though similar numbers of male (n = 1477) and female (n = 1633) fish were examined. Intersex was found in 3% of the fish collected. The intersex condition was observed in four of the 16 species examined (25%) and in fish from 34 of 111 sites (31%). Intersex was not found in multiple species from the same site but was most prevalent in largemouth bass (Micropterus salmoides; 18% of males) and smallmouth bass (M. dolomieu; 33% of males). The percentage of intersex fish per site was 8-91% for largemouth bass and 14-73% for smallmouth bass. The aim of the study was to validate a dummy regression approach for predictive modelling the fillet fatty acid (FA) composition of cultured European sea bass with dietary FA composition and lipid fillet content as independent variables. The model used our own data on gilthead sea bream as reference subgroup dataset and data from turbot, sole and European sea bass as dummy variables. Most of the observed variance within and among species was explained by the regression model without statistical significant interactions on blocks between diet composition and fish species subgroups. The effect of gutting and filleting on the formation of biogenic amines and its relation to microbiological and sensory attributes in ice-stored sea bass (Dicentrarchus labrax) was studied. Pseudomonads, H2S-producing bacteria, Brochothrix thermosphacta and to a lesser extent Enterobacteriaceae were the dominant micro-organisms in whole, gutted and filleted sea bass. Higher populations (P,0.05) of these micro-organisms were present in filleted sea bass as compared to gutted and whole fish samples. Seven biogenic amines namely: putrescine, cadaverine, spermidine, tyramine, tryptamine, spermine and histamine were determined in fish samples. Putrescine was the main biogenic amine formed. Its highest value (23.86 mg/kg) was recorded in filleted sea bass on day 16 of storage. Respective putrescine values for gutted and whole sea bass were 12.64 and 3.12 mg/kg (day 16). Sea bass is and always has been a very important fish for recreational sea anglers. However, since commercial fishing for bass took off in the 1970s the stock has become overexploited, with fever and smaller bass left in our waters as the result. The European Anglers Alliance (EAA) and the European Fishing Tackle Trade Association (EFTTA) pushed for more than a decade that the EU should curb overfishing of sea bass, and increase the minimum landing size. The Member States were well aware that the bass was in trouble but delayed the adoption of any management measures for years. Finally, the EU Commission by itself put in place emergency measures in 2015 and increased the minimum landing size from 36 cm to 42 cm for bass fisheries in North Atlantic waters. Analysis of population genetic relationships reveals the signatures of ongoing processes such as spawning behavior and migration, and historical events such as vicariance and climate change. This study provides a dual genome analysis of population genetic patterns of spawning smallmouth bass across 13 locations-including all Great Lakes and the Mississippi, Ohio, St. Lawrence, and Hudson rivers -using eight nuclear DNA microsatellite loci and mtDNA cytochrome b sequences. Null hypotheses are (1) genetic variation is not significantly partitioned among watersheds, lakes, rivers, and basins and (2) there is not a significant difference between mitochondrial and nuclear DNA patterns. Results discern genetic demarcations across the range, and pronounced differences even among closely spaced riverine sites.
