A process is provided for isolating and purifying biologically active RNA from a biological sources containing RNA, DNA and other cellular materials. The process involves contacting the RNA-containing source with particles comprising siliceous material, such as finely-divided glass, in the presence of a binding solution comprising concentrated, acidified chaotropic salt. Under these conditions, RNA, but not DNA, binds selectively to the siliceous material, and can be separated easily from the other components of the sample. Preferably, the selective binding process is applied to biological cells containing RNA of interest. Intact cells are disrupted by exposing them to a lysing solution comprising, as its main component, concentrated, acidified chaotropic salt. The RNA is then isolated and purified from the lysate using the selective binding process of the invention. The process of the invention may be conveniently provided as a kit, which may include the siliceous material, reagents and instructions

One version of the Steppe Hypothesis of Indo-European language origins suggests that Proto-Indo European languages developed in the steppe north of the Black and Caspian seas, and that the earliest known diverging branch - Anatolian - was spread into Asia Minor by movements of steppe peoples through the Balkan peninsula during the Copper Age around 4000 BCE, as part of the same incursions from the steppe that coincided with the decline of the tell settlements. [51] If this were correct, then one way to detect evidence of it would be the appearance of large amounts of characteristic steppe ancestry first in the Balkan Peninsula, and then in Anatolia. However, our genetic data do not support this scenario. While we find steppe ancestry in Balkan Copper Age and Bronze Age individuals, this ancestry is sporadic across individuals in the Copper Age, and at low levels in the Bronze Age. Moreover, while Bronze Age Anatolian individuals have CHG/Iran Neolithic related ancestry, they have neither the EHG ...

Referencing the Councils conclusion that dementia, and people with dementia, should become an accepted and visible part of society, the first of these elements concerns combating stigma which - the WHO report suggests - will involve implementing strategies to improve public awareness and understanding of dementia, and ensuring that the work of caregivers is properly valued. In relation to this aim, the WHO report also finds (in its second element) that "people with dementia have the right to live in the community and to have access to health, social and other support services that enable them to lead full and meaningful lives within society. Societies should enable them to take part in activities, have equitable access to facilities and be involved in communal life." The Council very much concurs with this conclusion, suggesting in its own report that services should be flexible and appropriate to the individual and enable them to live well with dementia - an approach based on respect for the ...
Barbara McAneny, MD, and Joshua Stoneking, PharmD, directed this discussion. The #1 source of revenue for oncology practices is drugs from infusion therapy, whereas, in comparison, revenue from evaluation and management is quite a bit lower. The impact of MIPS on oncology practices was discussed, including mandates for PQRS reporting, advancing care information, and clinical practice improvement activities. FDA guidelines on biosimilars development were discussed; physicians are concerned that biosimilar use will be mandated by payers, but the American Medical Association and other physician groups are developing policy statements that physicians should remain in control of whether to use biosimilars for specific patients. The evolving role of specialty pharmacy (SP) was also discussed, including how to find the right SP for a practice. The range of services that SPs provide was reviewed, including 24-hour access, adherence management, financial assistance, and other support services, such as ...
April is Autism Awareness Month, and April 2 is World Autism Day. These observances offer the opportunity to highlight the increasing number of children identified with autism spectrum disorder (ASD) and the substantial burden on families and health, educational and other support services, as well as an opportunity to celebrate the unique perspectives of those living with ASD.. ASD is a developmental disability that can cause major social, communication, and behavioral challenges. Signs of ASD begin during early childhood and usually last throughout a persons life (1). The cause of most cases of ASD is unknown, and there is currently no cure. CDCs most recent surveillance data indicate that about one in 68 children has been identified with ASD (2), which represents an almost 30% increase since the previous estimate in 2012. CDC has been active in documenting changes in the number and characteristics of children with ASD over the past decade. However, there remains an urgent need to continue ...

Background: Epigenetics is one of the most important mechanisms for inactivating tumor suppressor genes or tumor-related genes in various human cancers. Hepatocellular carcinoma (HCC) is one type of human cancer in which epigenetic gene suppression is frequently found. To effectively detect a tumor-related, we performed triple combination array analysis, which consists of an expression array, a single nucleotide polymorphism (SNP) array, and a methylation array of the same surgical HCC specimen.. Methods: Triple array analysis of one HCC sample from a 68-year-old female patient was performed. Gene expression profiles were determined using Affymetrix HGU133A and B Gene-Chips using RNA. The SNP chip array was conducted using the standard protocol for Affymetrix Gene Chip Mapping 500K arrays. Methylation array analysis used an Illumina Infinium HumanMethylation27 Beadchip Kit. The gene encoding Collagen type1 alpha1 (COL1A1) was identified and verified using HCC cell lines and 48 primary HCC ...

It was charged by the local German command post immediately after German advance into Feodosia first to care for the Russian wounded remaining behind in the Russian field hospital. During the following time period, he then also made an effort to take care of the German wounded as well. After the surrender of the city of Feodosia by the Germans on 29 December 1941, Russian marines and later infantry men from the Red Army came as well. The marines ran amok throughout the city, completely drunk. Still on the first day, they went to the field hospital opposite the castle and shot the German wounded lying there, according to his estimate about 30 to 35 German soldiers. On the third day of the occupation, i.e. on 1 January 1942, the general order was issued to shoot all Germans found in the reoccupied area. Commandos were then created by the commissars and N.K.V.D. who also shot all the German wounded lying in the other field hospitals and casualty collection centers. About 60 German soldiers lay in ...
During the time I was in Gross-Rosen these were the only two occasions on which I had any social contact or a kind gesture was made towards me. These were shrouded, in a necessary veil of secrecy. If this contact had been recognized as an act of kindness either I or the German soldier or my Polish workmate would have been shot or severely punished. It was mandatory for prisoners to take off their hats when passing any man in a German uniform. When passing this German soldier I removed my hat as I said. Dinke Shane. He acknowledged this with an expression of gratitude, pierced with sorrow. In contrast to this kindness, during the lunch breaks some of the civilians treated me as if I were an animal, hurling vegetable peels different distances to watch me scurry and scramble to retrieve them. I have wondered whether they did this as a foil to protect themselves in the event that they were caught or if they enjoyed this heartless play. The man I worked with threw vegetable scraps that were thicker ...
Drug abuse, incarceration, unexplained illnesses, chronic fatigue syndrome, gastrointestinal symptoms, skin diseases, fibromyalgia, and chronic pain may also be associated with the stresses of being in a war, but the evidence to support these links is weaker. For other health problems and adverse effects that the committee reviewed, the data are lacking or contradictory; the committee could not determine whether links between these ailments and deployment-related stress exist.. Although the report cannot offer definitive answers about the connections between many health problems and the stresses of war, it is clear that veterans who were deployed to war zones self-report more medical conditions and poorer health than veterans who were not deployed. Those who were deployed and have PTSD in particular tend to report more symptoms and poorer health, the committee found. PTSD often occurs in conjunction with other anxiety disorders, depression, and substance abuse; its prevalence and severity is ...

Ever since the ancient time, dogs have always been important to man. The friendship between humans and this animal was formed for untold thousands of years. Dogs have generally helped ancient man survive by performing various tasks. Because dogs are highly valued, maltreatment of these animals is considered a serious crime that results to severe punishments. The ancient tribes in the Balkan Peninsula have always kept herding dogs. These dogs have accompanied the nomadic tribes in their seasonal migrations. This resulted to the development of several types of herding dogs. The Karakachan Dog, a breed whose ancestors have started to develop in the 3rd millennium BC is considered to be one of the oldest breeds of dogs in Europe. Similar to other ancient breeds, the precise origin of the Karakachan Dog is unknown. This herding breed, similar to other Turkish and Balkan molosser breeds is believed to have descended from ancient Central Asian and Tibetan mastiffs. This ancestry made the Karakachan Dog ...

To provide full service in the field of biologics, PPS offers an all-encompassing range of analytical capabilities from drug discovery and development to release testing and regulatory CMC Services under ICH Q6B.. Biologics have revolutionized the treatment and prevention of many disabling and life-threatening diseases. These drugs are mostly derived from biological sources such as animal cells, bacterium and yeast. Biopharmaceutcial drugs can either be small molecules such as human insulin, erythropoietin (EPO), human growth hormone G-CSF or complex large molecules such as monoclonal antibodies.. As proteins are complex molecular entities derived from biological processes, they are almost always not fully purified and homogeneous molecules, but instead show a certain degree of structural heterogeneity. This means that the characterization of a protein always has three aspects:. ...
A simple ecosystem model is coupled to a 3-dimensional general circulation model for the North Atlantic. The physical model is based on the Los Alamos Parallel Ocean Program (POP) and forced by climatological monthly mean data. Four biological components(phytoplankton, zooplankton, nutrients and detritus) are incorporated into POP as additional tracers with biological sources and sinks. The model solutions, obtained with different physical and biological parameterizations are compared against monthly mean SeaWiFS colour data averaged over the period 1997-2003 and Levituss climatological nitrate data. A reference model solution, with constant biological model parameters over the whole basin, underestimates both the average chlorophyll level and its regionalvariability at mid- to high-latitudes. Experiments with a different parameterization of heat and freshwater fluxes, which affects upper ocean mixing, indicate a strong impact of such parameterizations on nutrient supply to the surface layer at ...
Press Release issued Mar 15, 2017: Biologics are substance or products extracted or manufactured from biological sources such as living organisms, proteins, nucleic acid, or a complex combination of these substances. It includes a wide range of products such as gene therapy, somatic cells, tissues, recombinant therapeutic proteins, and blood components. Virus contamination of commercially valued cell culture may pose a health risk to the population and a financial burden to the manufacturing and biopharmaceuticalcompanies. In addition, safety testing for biologics is difficult and requires specialized evaluation.

Berman and his cohorts are flatly wrong. The FDA does regulate what are called "biologics" - treatments that are not drugs but they are proteins, blood products, cells, and other material prepared from biological sources. So taking adipose tissue from a patient, treating it, and then injecting it as a stem cell treatment is legitimately considered a biologic and under FDA purview.. A core part of the stem cell clinic model, which we have seen before from charlatan innovators like Stanislaw Burzynski, is to claim that you are doing research. The treatments are therefore experimental, which nicely obviates you from having to provide evidence for safety and efficacy. They call it "patient-funded research" because the patients are still paying out-of-pocket thousands of dollars for the treatment. This, of course, raising its own problems.. There is a carefully designed system, evolved over decades, for protecting the public from unethical medical research. The rules are not arbitrary, have been ...

This chapter explores the modeling of biological sources in regard to electronic neuron models. The bioelectric behavior of neural cells and the electric concepts used in this discussion are further exemplified with electronic circuits. The discussion on electronic neuron models may also serve as a basic introduction to neurocomputers, which are a fascinating example of applying biological principles to technological systems. ...
Human saliva has been used as an auxiliary biological fluid for disease diagnosis (1-3), because the biomolecular composition of saliva changes in a disease state. Total RNA extracted from saliva is useful in determining gene expression profiles in cancer, as well as in genetic studies (4, 5). Salivary RNA is prone to degradation by RNases and other nucleases present in saliva, and therefore care should be taken to alleviate or minimize sample degradation before downstream transcriptomic analysis. Although the number of methods for isolating RNA from saliva has markedly increased over the years, most of the protocols remain relatively costly, owing to the predominant use of commercial kits (6) and, more importantly, the relatively low RNA yields. Furthermore, a large body of literature on transcriptomic studies of saliva has considered only the cell-free salivary supernatant as a biological source for isolating RNA, as opposed to the salivary cellular pellet (7-10). New high-throughput ...
Methods and compounds for the preparation of nucleic acid samples. Polymerase inhibitors are treated so as to allow for amplification of nucleic acid from biological sources, including whole blood.
The present invention describes an RNA isolation process which utilizes low pH reagents. In addition, the reagents are less hazardous and are more stable than those used in prior art methods. This rapid method may be used to obtain purified RNA from a variety of biological sources including human whole blood, plant and animal tissues, cultured cells, body fluids, yeast, and bacteria.
Dr. McKinzie began her career at NCTR developing a specialized polymerase chain reaction (PCR) method for detecting and quantifying cancer-related mutations at the sensitivity of 1 mutant out of 100,000 normal copies of the gene. This method, called allele-specific competitive blocker PCR (ACB-PCR) has proven useful over the years for collecting data on cancer-gene mutations. However, with the advent of next generation sequencing (NGS), the quantification of these mutations can be expanded to cover many mutations simultaneously. This will increase the throughput of cancer-gene mutation data from various biological sources.

Four main classes of proteolyic enzymes have been routinely utilized to describe proteases. The serine proteases are probably the best characterized. This class of proteases includes trypsin, chymotrypsin, and elastase. The cysteine protease class includes papain, calpain, and lysozomal cathepsins. Aspartic proteases include pepsin and rennin. Metalloproteinases include thermolysin and carboxypeptidase A.. During isolation and characterization one or all four classes of proteases may pose a threat to the fate of a protein. Broad-spectrum protease inhibitors and mixtures (or cocktails) have been developed to protect the integrity of isolated proteins. Sigma® offers and manufactures the broadest range of protease inhibitors and inhibitor cocktails of any supplier. Sigma inhibitor cocktails have been specifically formulated for particular applications as they relate to the biological source or method of expression.. Protease inhibitors can be added during cell growth and protein expression or can ...
