During the last decade Bacillus megaterium was systemically developed for the gram per liter production of recombinant proteins. Fungal laccases have great potential as biocatalysts oxidizing a variety of aromatic compounds using oxygen as co-substrate. Here, the crystal structure of 7D5 laccase (PDB 6H5Y), developed in |em|Saccharomyces cerevisiae|/em| and overproduced in |em|Aspergillus oryzae|/em|, is compared with that of the wild type produced by basidiomycete PM1 (|em|Coriolopsis|/em| sp.), PDB 5ANH. SAXS showed both enzymes form monomers in solution, 7D5 laccase with a more oblate geometric structure due to heavier and more heterogeneous glycosylation. The enzyme presents superior catalytic constants towards all tested substrates, with no significant change in optimal pH or redox potential. It shows noticeable high catalytic efficiency with ABTS and dimethyl-4-phenylenediamine, 7 and 32 times better than the wild type, respectively. What are we trying to do while making koji?. OK, so what exactly are we trying to do when making koji? Well, to examine this we need to consider the role koji plays in Sake Brewing. In sake brewing we use koji to provide a wide variety of products. These include products that provide flavor and aroma elements as well as enzymes which degrade proteins and starches into smaller component parts. For example proteins are disassembled into peptides and amino acids while starches are converted into smaller starches, dextrins and sugars.. Rice starts out with 7% to 8% protein, but the higher polished the rice the less protein will be left. However, even with lower levels of milling we do not focus on kojis production of enzymes to break down this protein. IPU is important for the production of isopanose and is used in the structural analysis of oligosaccharides with α-1,4 and α-1,6 glucosidic linkages. We have isolated the ipuA gene encoding IPU from the filamentous fungi A. niger A.T.C.C. 9642. The ipuA gene encodes an open reading frame of 1695 bp (564 amino acids). We investigated the effect of proteases, widely used for neuron isolation in electrophysiological studies, on the amplitude and kinetic characteristics of persistent sodium current (INaP) in hippocampal CA1 pyramidal neurons. Properties of INaP were studied on neurons isolated by mechanical treatment (control group) and by mechanical and enzymatic treatment using pronase E (from Streptomyces griseus) or protease type XXIII (from Aspergillus oryzae). We show that in neurons isolated with pronase E kinetic of activation and density of INaP was unaltered. Enzymatic treatment with protease type XXIII did not alter INaP activation but result in significant decrease in INaP density. To this end, we will study transcription activator-like effector (TALE) function using the economically important model system Xanthomonas oryzae and rice (Oryza sativa). We follow an interdisciplinary approach combining biological and bioinformatics work packages. In the biological part we will identify TALEs from different, presently uncharacterized X. oryzae strains. Transcriptomics (RNA-seq) will reveal which target boxes of TALEs are functional in vivo and will also allow us to identify TALE target genes. Systematic reporter assays of target boxes in their promoter context will allow us to dissect different biological determinants of TALE function. In this study, primers designed from unique regions within the rDNA internal transcribed spacers have been used to develop a rapid PCR-based diagnostic test to provide an accurate identification of the species on rice. Detekcijom proteinskih vrpci nakon primjenjenih elektroforetskih metoda, te glikoproteina prenešenih na PVDF membranu nakon SDS-PAGE uoceno je da sve vrste meda pokazuju vrlo slicne proteinske i glikoproteinske elektroforetske profile koji se razlikuju samo po intenzitetu proteinskih vrpci.
