Geophagy has been associated with iron deficiency and anaemia, but no causal relationship has been established. To clarify this, we conducted a two-by-two factorial randomised, controlled trial on the effect of iron and multimicronutrient supplementation on geophagy in Zambian schoolchildren in Lusaka, from February to December 2001. Of the 406 children, 212 (52.2%) were girls and the mean (range) age was 10.2 (7-15) years. Geophagy was reported by 302 (74.4%) and more often in girls than in boys (80.2 vs. 67.7%, P = 0.007). The mean (range) daily earth intake was 25.2 (1-200) g. Geophageous children had more often geophageous relatives than non-geophageous children (79.5 vs. 1.9%, P , 0.001). Rats were immunized with crude Ascaris extracts together with Bordetella pertussis organisms and the homocytotropic antibody titers were estimated by homologous PCA. Rats injected with 1 mg of crude extract produced homocytotropic antibodies giving and average PCA titer of 1:80. A procedure for the isolation of malic enzyme from muscle tissue of the roundworm Ascaris suum is described. The fractionation method yields relatively large quantities of the enzyme,with a specific activity of fifteen moles of malate converted to pyruvate and carbon dioxide per min per mg at 25º. Homogeneity was established with analytical ultracentrifugation, zone electrophoresis, isoelectric focusing, and rechromatography. The molecular weight of the enzyme was 250,000, and it is dissociated under several conditions into four identical monomers of 64,000 daltons. The enzyme exists as a single electrophoretic form and prefers manganous and NAD over other cations and NADP. Ammonium sulfate competes with manganous for the active site and titration with DTNB yields eight thiol groups per mole. Titration of the first four thiol groups is accompanied by a complete loss in enzyme activity. Abstract: Ascaris suum extract (Asc) down-regulates the immune response to ovalbumin (OA) as well as preferentially stimulates a Th2 response to its own components, which is responsible for the suppression of the OA-specific Th1 response. IL-4 plus IL-10-specific monoclonal antibody treatment of mice before immunization with OA + Asc abolishes this suppression. The immunosuppressive activity of Asc is due to high MW proteins (PI) that are obtained by gel filtration chromatography. On ultrasound, it is seen as linear non-shadowing echogenic structures with target appearance in cross section, and the live worm may show writhing movements in real time.
