Objective: A field experiment was conducted in order to study the effect of phosphorus and manure application on agronomic performance and seed yield of groundnut. Methods: Factorial combination of two groundnut varieties (Werer 962 and Oldhale), Three rates of phosphorus (0, 90 and 180 kg P2O5 ha-1) and three rates of manure (0, 5 and 10 ton ha-1) were laid out in randomized complete block design with three replication. Results: The combined application of 180 kg P2O5 ha-1 and 10 ton manure ha-1 resulted in the highest number of pods per plant and seed yield. The increase in number of pods per plant and seed yield at the highest combined application rates of the two fertilizers were 40 and 60%, compared to the control of the two fertilizers. Generally, the present study has shown that the integrated use of manure (10 ton ha-1) and inorganic phosphorus (180 kg P2O5 ha-1) fertilizer resulted in highest seed yield of groundnut compared to the application of either fertilizer alone.

To achieve this, one of the important strategies is to genetically engineer the ruling peanut varieties using transcription factor regulating the expression of several downstream, abiotic-stress responsive gene(s). AhAREB1 (Arachis hypogaea Abscisic-acid Response Element Binding Protein 1) is a member of the basic domain leucine zipper (bZIP)-type transcription factor in peanut. Previously, we found that expression of AhAREB1 was specifically induced by abscisic acid (ABA), dehydration and drought. Constitutive expression of AhAREB1 confers water stress tolerance and is highly sensitive to exogenous ABA. Microarray and further real-time PCR analysis revealed that drought stress, reactive oxygen species (ROS) scavenging, ABA synthesis/metabolism-related genes and others were regulated in transgenic Arabidopsis overexpressing AhAREB1. Near infrared reflectance spectra (NIRS) was collected from Arachis hypogaea seed samples and used in predictive models to rapidly identify varieties with high oleic acid. The method was developed for shelled peanut seeds with intact testa. Spectra was evaluated initially by principal component analysis (PCA) followed by partial least squares (PLS). PCA performed with full spectra and reduced spectra with one principal component accounted for 97% to 99% variability, respectively. The PLS model generated from first derivative spectra provided a standard error of prediction (SEP) of 7.7204808. This technique provides a non-destructive method to rapidly identify high oleic peanut seeds to support the selection and cultivation of high oleic acid peanut varieties. The method can also be useful at peanut processing facilities for screening and quality assessments.

The other thing with MaraNatha is the price, I had to take out a loan at the check out counter to pay for it~ and after my down payment on the 6.49 bottle of creamed nuts they let me take it home ... Presented in this dissertation are comprehensive studies of the structures of the peanut allergen protein Ara h 2 and the effect of food processing (roasting) on it. A detailed elucidation of the primary structure and PTM of Ara h 2 from the raw peanuts has been described. Ara h 2 isoforms were purified and cleaved via microwave accelerated trypsin digestion. The peptide mixtures were analyzed by LC-MS/MS and targeted CID. De novo sequencing of the MS/MS spectra revealed the protein sequence of each Ara h 2 isoform. Several hydroxyproline sites have been discovered while disulfide bond structures have been partially determined. Using anti-Ara h 2 antibodies, Western blotting of 1-D gels of the raw and dark roasted peanuts was carried out in order to characterize the changes of Ara h 2 between these two samples.
