Cylindrospermopsis raciborskii is a freshwater cyanobacterium. This bacterium is an aquatic photosynthetic bacteria belonging to the phylum Cyanobacteria. They are composed of chained filaments known as trichomes that can show variation in morphology, varying from about 50-300 micrometers in length. These bacteria can also produce a thick walled, cylindrical, spore like structure known as akinetes, which also demonstrate variation in morphology. Some strains of this species are able to produce several toxins which affect humans: cylindrospermopsin, anatoxin-a and saxitoxin. This first toxin, cylindrospermopsin, has the ability to affect the liver and kidneys in humans as well as cause mild skin reactions upon exposure. The last two toxins, anatoxin-a and saxitoxin, are thought to be shellfish neurotoxins. The germination of akinetes assists the dispersal and persistence of the species. The assessment of the akinete pool in lake sediments is essential to predict the bloom formation of the Nostocales population. Thus, the variation among global C. raciborskii populations was reflected differently in the nifH and cpcBA-IGS nucleotide sequences. Although nifH was the smaller of the two gene fragments sequenced, regional distinctions could be made based on these sequences. C. raciborskii isolates originating from Europe, Australia, or the Americas were separated phylogenetically into distinct clusters based upon the nifH sequence data. While there were distinctions between regions revealed by the nifH sequence data (variation of up to 2.5%), the amount of variation within each of these three geographic regions was very low (,1%). The EPA issued health advisories in June 2015 for two cyanotoxins (microcystins and cylindrospermopsin) and utilities have been struggling with how to manage bloom events and optimize treatment to remove cyanotoxins. This webcast will highlight the effectiveness of conventional and advanced treatment processes for managing intracellular and extracellular cyanotoxins while minimizing unintended consequences. It forms large blooms on the water surface and overwhelmingly dominates the ecosystem through the formation of colonies from single cells surrounded by mucilage; however, the mechanism of colony formation is poorly understood. Two mechanisms of Microcystis colony formation have been proposed: cell-division, where cells remain attached after binary fission; and cell-adhesion, where single cells stick together. We examined the seasonal and interannual variations in nutrient (nitrogen and phosphorus) concentrations between 2013 and 2014 to describe the relations between these factors and the growth dynamics of Aphanizomenon and toxigenic Microcystis as described by DNA sequencing and qPCR. Although total nutrients and chlorophyll a concentrations were similar between years, qPCR results showed the cyanobacterial populations to be 40 times larger in 2014 and indicated a large shift from an Aphanizomenon-dominant, low microcystin-level regime in 2013 to one dominated later in the season by

The Lake Champlain Basin Program provides funding to support two aspects of cyanobacteria monitoring on Lake Champlain: through the Lake Champlain Long-Term Monitoring Program, in partnership with VT Agency of Natural Resources, NYS Department of Environmental Conservation, and SUNY Plattsburgh. Trained staff collect samples from relatively deep locations from a dozen sites around Lake Champlain. If a site has a full bloom or is developing a bloom, samples are collected and sent to the VT Department of Health Laboratory in Burlington for toxin analysis, and the cyanobacteria species is identified. All sample results, including negative (or clean results) are posted on the VT Department of Health Cyanobacteria tracker website.. Cyanobacteria are found almost everywhere, but particularly in lakes and in the ocean where, under certain conditions, they reproduce exponentially to form blooms. Cyanotoxins are equally toxic whether they are within the algal cell (intracellular) or have been released into the water column (extracellular). However, cyanotoxins contained within the algal cell are more easily removed by water treatment plants using conventional treatment processes of coagulation and sedimentation, and this is the reason a lot of attention is paid to whether the cyanotoxin is intracellular or extracellular.. Preliminary data from impartial 3rd party labs indicate that EarthTec is effective at halting growth of cyanobacteria and production of cyanotoxins without a significant increase in the extracellular fraction of toxins. I am interested in understanding the microbial composition and interaction in the environment. Our group focuses mainly on soil and aquatic environments. We are particularly interested in understanding how eukaryotic microbes respond to changes in their environments and how these changes impact on the trophic links of that environment. The projects involve collaborations with national and international researchers with complementary expertise in these areas.. In the soil, we are particularly interested in understanding how filamentous and zoosporic fungi grow in soil under different soil conditions. Our group has developed a new technique for quantifying fungal growth in three dimensions using computer-aided tomography. We are keen to develop this approach by applying this technique to other biological specimens..
