Background: Der f 7 is the group 7 allergen from the dust mite Dermatophagoides farinae, homologous to the major allergen Der p 7 from D. pteronyssinus. Monoclonal antibody that bind to residues Leu48 and Phe50 was found to inhibit IgE binding to residue Asp159, which is important for the cross-reactivity between Der f 7 and Der p 7. Methodology/Principal Findings: Here, we report the crystal structure of Der f 7 that shows an elongated and curved molecule consisting of two anti-parallel β-sheets - one 4-stranded and the other 5-stranded - that wrap around a long C-terminal helix. The overall fold of Der f 7 is similar to Der p 7 but key difference was found in the β1-β2 loop region. Antagonists of the type 1 cysteinyl leukotriene receptor (CysLT1R) are widely used to treat asthma and allergic rhinitis, with variable response rates. Alveolar macrophages express UDP-specific P2Y6 receptors that can be blocked by off-target effects of CysLT1R antagonists. Sensitization to house dust mites (HDM) is highly prevalent among the young atopic population in Singapore. Previously published data suggest that individuals with skin allergies show preferred sensitization to Dermatophagoides pteronyssinus while individuals with pure respiratory allergies show preferred sensitization to Blomia tropicalis. The aim of our study was to compare the sensitization profiles between children with asthma and those with eczema to D. pteronyssinus and B. tropicalis and their specific allergens. A total of 60 children, 30 with asthma and 30 with eczema were recruited. IgE levels specific for a panel of HDM allergens from the two mite species were measured using enzyme-linked immunosorbent assay. A cDNA clone coding for a mite allergen of mol wt approximately 14,000 has been isolated and its DNA sequence determined. The native component from mite extracts encoded by this DNA was identified by immunoprobing blots of mite body extract with human IgE eluted from the electroblotted cloned fusion polypeptides derived from the expressed cDNA clone. The clone encodes a polypeptide with a deduced mol wt of 17,460. The deduced amino acid sequence was not homologous to any known protein sequences and it contains no cysteine or tryptophan. Hydrogen water (HW) produced by electrolysis of water has characteristics of extremely low oxidation-reduction potential (ORP) value and high dissolved hydrogen (DH). It has been proved to have various beneficial effects including antioxidant and anti-inflammatory effects; however, HW effect on atopic dermatitis (AD), an inflammatory skin disorder, is poorly documented. In the present study, we examined the immunological effect of drinking HW on |i|Dermatophagoides farinae|/i|-induced AD-like skin in NC/Nga mice. Mice were administered with HW and purified water (PW) for 25 days. We evaluated the serum concentration of pro-inflammatory (TNF-|i|α|/i|), Th1 (IFN-|i|γ|/i|, IL-2, and IL-12p70), Th2 (IL-4, IL-5, and IL-10), and cytokine expressed by both subsets (GM-CSF) to assess their possible relationship to the severity of AD. The serum levels of cytokines such as IL-10, TNF-|i|α|/i|, IL-12p70, and GM-CSF of mice administered with HW was significantly reduced as compared to PW group.
In December 1995 we took dust samples from nine pairs of pillows and analysed them for Der p I. Each pair consisted of one feather filled pillow and one filled with polyester fibre; these had been used together on the same adult bed for more than six months to ensure that the environmental exposures of the pillows in each pair were similar. The pillow fillings were encased in closely woven cotton fabric. Two of the subjects in our study slept with the polyester filled pillow on top, four with the feather filled pillow on top, and three had no preference. We took dust samples with a portable Hitachi CV-2500 vacuum cleaner with a sock attachment, vacuuming each pillow for three minutes on each side, a total of six minutes per pillow. We sieved the dust collected to remove fluff and large particles and weighed the resulting fine dust. The bronchial airway epithelial cell (BAEC) is the site for initial encounters between inhaled environmental factors and the lower respiratory system. Our hypothesis was that release of pro inflammatory interleukins (IL)-6 and IL-8 from primary BAEC cultured from children will be increased after in vitro exposure to common environmental factors. Primary BAEC were obtained from children undergoing clinically indicated routine general anaesthetic procedures. Cells were exposed to three different concentrations of lipopolysaccharide (LPS) or house dust mite allergen (HDM) or particulates extracted from side stream cigarette smoke (SSCS). BAEC were obtained from 24 children (mean age 7.0 years) and exposed to stimuli. Compared with the negative control, there was an increase in IL-6 and IL-8 release after exposure to HDM (p ≤ 0.001 for both comparisons). There was reduced IL-6 after higher compared to lower SSCS exposure (p = 0.023). There was no change in BAEC release of IL-6 or IL-8 after LPS exposure.

Es wird deshalb auch erwartet, dass die Betriebe in Abhängigkeit vom regionalen Kontext unterschiedlich reagieren. Die Gewinnerwartung der Betriebe wird mithilfe des Testbetriebsnetzes des BMELV geschätzt. Die Betriebsaufgabewahrscheinlichkeit wird in einem zweiten Schritt anhand der flächendeckenden Daten der Agrarstrukturerhebung in einem logistischen Modell für verschiedene Betriebstypen bestimmt. Cyclooxygenasen regulieren die Produktion von Prostaglandinen und spielen eine Rolle bei der Entstehung und Progression maligner Tumore. Versuche mit COX Inhibitoren (NSAIDs) zeigten im Tiermodell eine deutliche Reduktion von Inzidenz und Größe der Tumoren in einer dosisabhängigen Weise. Das Ziel der vorliegenden Arbeit war, das Expressionsmuster der induzierbaren Isoform COX-2 und der konstitutiven Isoform COX-1 im Mammakarzinom zu untersuchen. Die Grundlage der Untersuchung bildeten Tumorproben von 221 Patientinnen mit primärem Mammakarzinom. Diese wurden mittels immunhistochemischer Methoden auf beide COX-Isoenzyme untersucht.
