Purpose: : To evaluate the efficacy of early sutureless amniotic membrane transplantation in the management of severe bacterial keratitis to reduce pain, inflammation, and haze, and to promote healing. Methods: : A non-comparative case series including 3 eyes of 3 consecutive patients with severe bacterial keratitis exhibiting persistent epithelial defect/ulcer were retrospectively reviewed following early sutureless amniotic membrane transplantation via ProKeraTM together with selective topical antibiotics and preservative-free steroid. Pain relief, inflammation, haze, and corneal epithelial healing were monitored. Results: : ProKeraTM was inserted once in one eye, twice in the other two eyes. Pain was significantly relieved, and inflammation was markedly reduced in all cases. Objective: To evaluate the use of hypothermically stored human amniotic membrane for cartilage repair in adult sheep. Studies show that human amniotic membrane contains pluripotent mesenchymal stem cells that can be influenced to produce chondrocytes. It is unknown if human amniotic cells can produce hyaline-like cartilage. This study evaluates the use of hypothermically stored amniotic membrane (HSAM) to fill chondral defects in a sheep model. We hypothesized HSAM would fill defects with hyaline-like cartilage with chondrocytes populating the matrix. One sheep was used as a control, and four sheep received amniotic membrane. Two of these sheep were used as a normal control comparison. A 1 cm2 defect was created on the trochlear grove in all specimens. A reconstituted collagen membrane from human amnion has been investigated as a source of collagen matrix, which could be used as a substratum for culturing human fibroblasts. The suitability of pepsin-solubilized reconstituted human amniotic membrane, before and after cross-linking with chitosan, as a dermal matrix for culturing fibroblast was assessed by morphologic, physicochemical, cytotoxic and histochemical methods. Measurement of thermodynamic behaviour, by differential scanning calorimetric (DSC) and thermogravimetric analysis (TGA), and tensile strength suggested that the cross-linked membrane had sufficient elasticity to serve as an efficient dermal substrate for in vitro culture of fibroblasts. Increasing evidence has shown that mesenchymal stem cells (MSCs) yield a favorable therapeutic benefit for thermal burn skin wounds. Human amniotic MSCs (hAMSCs) derived from amniotic membrane have multilineage differentiation, immunosuppressive, and anti-inflammatory potential which makes them suitable for treating skin wounds. Wound healing is a complex pathophysiological process that involves a variety of cells and cytokines. In this study, we found that local injection of human amnion mesenchymal stem cells into wounds in rats could promote wound healing. Therefore, we hypothesized that the exosomes of human amnion mesenchymal stem cells contain substances that regulate the migration of epidermal cells. It has been reported that miR-135a is involved in cell migration and transformation. However, there have been no reports of its function in skin wound healing. To test this hypothesis, we injected exosomes overexpressing miR-135a directly into the wound margin. In addition, we tested the migration of BJ cells with overexpression or knockdown of miR-135a in vitro. Additionally, Western blot analysis was used to detect the expression of fibroblast migration-associated proteins after treatment with miR-135a overexpression or knockdown. Maternal immunoglobulin-G (IgG) is known to be transported across the placental syncytiotrophoblast during the period when the human fetus is incapable of manufacturing these defensive molecules. In this study we investigated the possible role of the amniochorion, that surrounds the amniotic cavity in which the fetus lies, in the transfer of immunoglobulin. Endogenous IgG was localised in the amniochorion by confocal immunofluorescence microscopy and by ultrastructural labelling of ultrathin frozen tissue sections using the protein A-gold technique. Immunoreactivity was identified in the extracellular matrix tissues and necrotic amniotic epithelial cells. The basic helix-loop-helix (bHLH) transcription factor, Hand1, plays an important role in the development of the murine extra-embryonic trophoblast cell lineage. In the present study, we have analysed the expression of Hand1 in human extra-embryonic cell types and determined its binding specificity and transcriptional activity upon interaction with different class A bHLH factors. Abstract: The purpose of this study was to investigate the relationship between cell apoptosis and wide-band electromagnetic pulse (WEMP) irradiation parameters and to provide experimental data to establish the biophysics damage model for WEMP. Cell counting kit-8 was used to detect the cell viability changes after WEMP irradiation and flow cytometry assay was used to detect cell apoptosis after irradiation under different WEMP irradiation parameters. Under experimental conditions, field strength and number of pulses were observed to be the main factors that increased the apoptotic rate of human amniotic epithelial cells. Specifically, electric field intensity and number of pulses were determined to be the primary and secondary factors, respectively. One of the most established roles of oxytocin (OT) is in inducing uterine contractions and labour. Apart from contractions, our recent study have shown that OT can also activate pro-inflammatory pathways in both human myometrial and amnion cells which suggests the pro-inflammatory role of OT should be taken into account when developing tocolytics targeting the OT/OT receptor (OT/OTR) system. The OTR antagonist, Atosiban, is currently used therapeutically for the treatment of PTL. We have previously shown that Atosiban fails to inhibit the pro-inflammatory effects of OT in human amnion, and Atosiban alone activates NF-κB and MAPKs thus upregulating downstream pro-labour genes. In this study we aimed to set up an in vitro culture of the rabbit amnion in order to support in vivo fetal membrane healing capacity following fetoscopy. Fetal membranes were collected from a mid- gestational rabbit, and cultured on collagen support material for 14 days. 34 rabbits at 22 - 23 days gestational age ( GA) underwent fetoscopy. Methods This retrospective study included 200 patients, above 8 years of age, with clinically diagnosed unilateral total limbal stem cell deficiency due to ocular surface burns treated between 2001 and 2010. Our purpose was to evaluate the diagnostic value of three-dimensional (3 D) CT surface reconstruction in spinal fractures in comparison with axial and reformatted images. A total of 50 patients with different CT-proven spinal fractures were analysed retrospectively. Based on axial scans and reformatted images, the spinal fractures were classified according to several classifications as Magerl for the thoraco-lumbar and lower cervical spine by one radiologist. Another radiologist performed 3 D CT surface reconstructions with the aim of characterizing the different types of spinal fractures. A third radiologist classified the 3 D CT surface reconstruction according to the Magerl classification. The results of the blinded reading process were compared. It was checked to see in which type and subgroup 3 D surface reconstructions were helpful. Readers one and two obtained the same results in the classification. The human tissue inhibitor of metalloproteinases (TIMP) is a glycoprotein with a molecular weight of 28,000. A human heteroploid epithelial-like cell line, designated as C-SPT, was recovered from one of several lots of human WI-38 fibroblast-like diploid cells that lacked evidence of Regan isozyme in early passages.
