To test the significance of the purine nucleotide cycle in renal ammoniagenesis, studies were conducted with rat kidney cortical slices using glutamate or glutamine labelled in the alpha-amino group with 15N. Glucose production by normal kidney slices with 2 mM-glutamine was equal to that with 3 mM-glutamate. With L-[15N]glutamate as sole substrate, one-third of the total ammonia produced by kidney slices was labelled, indicating significant deamination of glutamate or other amino acids from the cellular pool. Ammonia produced from the amino group of L-[alpha-15N]glutamine was 4-fold higher than from glutamate at similar glucose production rates. Glucose and ammonia formation from glutamine by kidney slices obtained from rats with chronic metabolic acidosis was found to be 70% higher than by normal kidney slices. Background and aim: In acute liver failure (ALF), the brain is exposed to high levels of ammonia. Human studies defining the clinical significance of ammonia in ALF are lacking. This prospective study evaluated the relationship of arterial ammonia levels at admission to complications and survival among patients with ALF. Methods: Eighty consecutive ALF patients admitted from March 2001 to December 2003 were followed up until death or complete recovery. All had arterial ammonia estimation at admission (enzymatic method). Logistic regression analysis was performed to identify independent predictors of mortality. Results: Forty two (52.5%) patients died. Non-survivors had significantly higher median ammonia levels than survivors (174.7 v 105.0 μ mol/l; p,0.001). An arterial ammonia level of = 124 μ mol/l was found to predict mortality with 78.6% sensitivity and 76.3% specificity, and had 77.5% diagnostic accuracy. An ammonia converter was constructed using a commercially available platinum catalyst, with the aim of applying it for the measurement of ammonia concentrations. The dependence of the ammonia converter efficiency on the reaction temperature and the ammonia concentration was investigated by measuring the concentration of the converted nitric oxide. The results showed that the catalyst almost completely oxidized ammonia into nitric oxide at temperatures above 600℃ in the tested range of ammonia concentrations. In addition, by combining the ammonia converter with a chemiluminescent NO_x analyzer we were able to measure the ammonia concentration in an ammonia-nitric oxide mixture to an accuracy of within 3% by making a correction due to the oxygen concentration.

Glutamine synthetase (GS) catalyzes the recycling of NH4+ with glutamate to form glutamine. Glutamine synthetase is highly expressed in the renal proximal tubule, suggesting ammonia recycling via glutamine synthetase could decrease net ammoniagenesis and thereby limit ammonia available for net acid excretion. This studys purpose was to determine the role of proximal tubule glutamine synthetase in ammonia metabolism under basal conditions and during metabolic acidosis. We generated mice with proximal tubule-specific glutamine synthetase deletion (PT-GS-KO) using Cre-loxP techniques. Under basal conditions, PT-GS-KO increased urinary ammonia excretion significantly. The optical properties and density of ammonia have not been exhaustively studied despite the relevance of these characteristics in astrophysical scenarios. This work presents new data pertaining to the real part of the refractive index of ammonia at 632.8. nm and the density at different temperatures of deposition ranging from 13. K to ammonias desorption temperature of approximately 110. K. The results indicate a significant variability for both parameters versus temperature: an increase of 50% for the density and of 10% for the refractive index as the temperature increases in the range of 13-60. K; at temperatures greater than 60. K, a constant value is reached for both parameters. Addition of nitrite to the medium increased the ammonia production as cells used the nitrite reductase pathway to form ammonia. When reactors were placed in anaerobiosis by N2 bubbling, ammonia production was sustained several days and the total ammonia formed was about two fold higher than in aerobiosis. Urbanization in the world increased the pollution levels in the environment making it for worsen life. General awareness about pollution is increasing today. To control pollution levels, biological methods are eco-safe and economical. In this study a wild strain of Bacillus species was isolated and optimized for bioremediation studies of ammonia in flask cultures. Strategy was tried to develop for ammonia bioremediation in a single stage aerobic nitrification and denitrification. The results reveal that the optimum pH for the isolated Bacillus species was 7 and optimum temperature was 30°C. At optimum pH and temperature this organism was able to remove 99.2% of ammonia in flask cultures supplemented with 5g/L of ammonium sulphate. The biomass obtained after bioremediation studies was 0.3g/L which is very low. The study on ammonia toxicity revealed that toxicity was due to pH and osmolarity.
