You may have fried the triggers--most of these cheap eBay type triggers have a voltage limit of around 300V, and the Vivitar 283s are known to go higher. So, maybe a voltmeter would be worth the investment. I would also go to the standby advice of check the batteries and the channel settings.. Theoretically, those triggers (which look like theyre for Nikon from the pass-through shoes contact pattern) should work. But if they rely upon a TTL pin signal of some kind, that could be another issue. Its unlikely that this is the case, since those triggers have separate Tx/Rx units, and Ive only see this type of misfunction with the Yongnuo RF-603s (which used a TTL signal to auto switch between Tx/Rx modes, so with non-Canon/Nikon hotshoes, the transmitter unit never went into Tx mode--which is why Yongnuo made an RF-603 II).. I will discuss two models: (1) We model RNA polymerase (pol-II) dynamics using data from pol-II ChIP-Seq time course datasets (wa Maina et al. 2014). Our model is used to estimate the elongation speed and promoter activation kinetics for early targets of ER-alpha in MCF7 cells. By clustering the promoter activation profiles we can associate differences in the kinetics with transcription factor binding, e.g. the earliest clusters are all associated with nearby FOXA1 binding. (2) We model the relationship between pol-II dynamics and mRNA production by combining 3 pol-II ChIP-Seq data with RNA -Seq data. Spatial/temporal control of Cas9 guide RNA expression could considerably expand the utility of CRISPR-based technologies. Current approaches based on tRNA processing offer a promising strategy but suffer from high background. Here, to address this limitation, we present a screening platform which allows simultaneous measurements of the promoter strength, 5, and 3 processing efficiencies across a library of tRNA variants. This analysis reveals that the sequence determinants underlying these activities, while overlapping, are dissociable. Rational design based on the ensuing principles allowed us to engineer an improved tRNA scaffold that enables highly specific guide RNA production from a Pol-II promoter. When benchmarked against other reported systems this tRNA scaffold is superior to most alternatives, and is equivalent in function to an optimized version of the Csy4-based guide RNA release system. Chelatable, mobile forms of divalent zinc, Zn(II), play essential signaling roles in mammalian biology. A complex network of zinc import and transport proteins has evolved to control zinc concentration and distribution on a subcellular level. Understanding the action of mobile zinc requires tools that can detect changes in Zn(II) concentrations at discrete cellular locales. We present here a zinc-responsive, reaction-based, targetable probe based on the diacetyled form of Zinpyr-1. The compound, (6-amidoethyl)triphenylphosphonium Zinpyr-1 diacetate (DA-ZP1-TPP), is essentially nonfluorescent in the metal-free state; however, exposure to Zn(II) triggers metal-mediated hydrolysis of the acetyl groups to afford a large, rapid, and zinc-induced fluorescence response. This region, located about 4 kilobases upstream from the genes, was able to direct transient expression of the bacterial Chloramphenicol Acetyl Transferase (CAT) gene in both bloodstream and procyclic forms of the parasite. The cyclic octapeptide amanitin is a relatively selective inhibitor of eukaryotic RNA polymerase II (RNAP II) and is commonly used to study RNAP II transcription. Therefore, we measured the sensitivity of G. lamblia RNAP II transcription to alpha-amanitin and found that unlike most other eukaryotes, RNAP II transcription in Giardia is resistant to 1 mg/ml amanitin. Greetings, Damage to the meristematic apex could do it - something that kills the chloroplasts. I dont know whether my plant is genetically variegated or not. Unlike wild plants I have seen with random white streaks in their leaves (which suggests viral infection to me), this plant always has white-edged leaves (variegata marginata). Chlorogalum is related to agave & manfreda and I dont know hoe genetics influences variegation in those plants (which are also frequently variegata marginata). I have selfed two years of seeds and none have come up variegated.
