The fungus C. albicans uses adhesins to interact with human epithelial surfaces in the processes of colonization and pathogenesis. The C. albicans ALS (agglutinin-like sequence) gene family encodes eight large cell-surface glycoproteins (Als1-Als7 and Als9) that have adhesive function. This study utilized C. albicans Δals mutant strains to investigate the role of the Als family in oral epithelial cell adhesion and damage, cytokine induction and activation of a MAPK-based (MKP1/c-Fos) signaling pathway that discriminates between yeast and hyphae. Of the eight Δals mutants tested, only the Δals3 strain showed significant reductions in oral epithelial cell adhesion and damage, and cytokine production. High fungal:epithelial cell multiplicities of infection were able to rescue the cell damage and cytokine production phenotypes, demonstrating the importance of fungal burden in mucosal infections. Despite its adhesion, damage and cytokine induction phenotypes, the Δals3 strain induced MKP1 ...

Deleted in Malignant Brain Tumors-1 protein (DMBT1), salivary agglutinin (DMBT1SAG), and lung glycoprotein-340 (DMBT1GP340) are three names for glycoproteins encoded by the same DMBT1 gene. All these proteins belong to the scavenger receptor cysteine-rich (SRCR) superfamily of proteins: a superfamily of secreted or membrane-bound proteins with SRCR domains that are highly conserved down to sponges, the most ancient metazoa. In addition to SRCR domains, all DMBT1s contain two CUB domains and one zona pellucida domain. The SRCR domains play a role in the function of DMBT1s, which is the binding of a broad range of pathogens including cariogenic streptococci, Helicobacter pylori and HIV. Mucosal defense proteins like IgA, surfactant proteins and lactoferrin also bind to DMBT1s through their SRCR domains. The binding motif on the SRCR domains comprises an 11-mer peptide in which a few amino acids are essential for binding (GRVEVLYRGSW). Adjacent to each individual SRCR domain are glycosylation domains,

(Standard deviation of RBC. volume/mean MCV) X 100. The normal value for RDW is 11.5% to 14.5%. The standard deviation of red cell volume is derived from size histogram data that plot red cell size after a large number of red cells has been analyzed by the instrument. The usefulness of the RDW is that in many cases the RDW will become abnormal earlier in the anemia process than the MCV Because many anemias (like iron deficiency anemias) develop over a period of time, this parameter may provide a sensitive indicator of red blood size change16 before the red cell indices become overtly abnormal.. The final item in the CBC is a platelet count. Information regarding ...
Treatment:. Patients with few clinical symptoms and mild anemia may not require treatment but only avoidance of cold. Keeping the patient warm may be sufficient treatment. Treat any underlying infection e.g. Clarithromycin or Azithromycin for Mycoplasma pneumoniae.. Anemia is generally mild. Only patients who have serious symptoms related to anemia or have a Raynaud-type syndrome that constitutes a threat to life or quality of life require active therapy.. Folic acid supplements for prevention and treatment of folate deficiency should be considered.. The disease is usually refractory to corticosteroids.. Rituximab may be an option for treatment in some cases.. In the presence of underlying lymphoma, chlorambucil or oral cyclophosphamide may be helpful.. Splenectomy is usually ineffective for the treatment of cold agglutinin disease, because the liver is the predominant site of sequestration. However, if the patient has splenomegaly, then the disease may respond to splenectomy. More importantly, ...
We retrospectively studied 232 patients with cold agglutinin disease (CAD) at 24 centers in five countries. In Norway and a northern region of Italy, the study was close to being population-based. For the first time, we demonstrate 4-fold differences between cold and warmer climates regarding prevalence (20 versus 5 cases/million) and incidence (1.9 versus 0.48 cases/million/year). Mean baseline hemoglobin level was 9.3 g/dL, but 27% had hemoglobin , 8 g/dL. Identification of typical features of CAD-associated lymphoproliferative disorder in the bone marrow was greatly increased by centralized biopsy assessment. CAD seems to be associated with a slightly increased risk of venous thrombosis. This work included a follow-up study of therapies, focusing on the long-term outcomes of the rituximab plus bendamustine and rituximab plus fludarabine regimens. Rituximab plus bendamustine therapy resulted in responses in 35 (78%) of 45 patients; 24 (53%) achieved complete response. Interestingly, these ...
Cold agglutinin disease (CAD) is a rare form of autoimmune hemolytic anemia with limited epidemiological and clinical data. We used the Danish National Patient Registries to examine CAD occurrence and risk of thromboembolic events (TEs) and mortality in CAD patients compared with a matched cohort from the general population in Denmark. We identified 72 patients diagnosed with CAD and 720 matched controls between 1999 and 2013. For 2013, the most recent year of study, crude incidence of CAD was 0.18 per 100 000 inhabitants per year and prevalence was 1.26 per 100 000 inhabitants. Risk of TEs was higher in the CAD patient cohort than in the comparison cohort at 1 year (7.2% of CAD patients had TEs vs 1.9% of comparisons), 3 years (9.0% vs 5.3%), and 5 years (11.5% vs 7.8%) after the index date. The median survival was 8.5 years. CAD patients had increased mortality compared with the general population cohort (adjusted hazard ratio [aHR], 1.84; 95% confidence interval [CI], 1.10-3.06; P 5 .020), ...
WALTHAM, Mass.--(BUSINESS WIRE)-- Bioverativ Inc., a Sanofi company dedicated to transforming the lives of people with rare blood disorders, today announced results from a new, population-based retrospective study that found a greater than twofold increased risk of mortality in people with cold agglutinin disease (CAD) within the first five years after diagnosis. The study, which is the first to compare overall survival among people with CAD with that of the general population, also showed that the increased risk of mortality and life-threatening thromboembolic events (TEs) such as stroke and heart attack was evident starting in the first year after diagnosis. These data were presented at the 23rd Annual Congress of the European Hematology Association (EHA). "The results of our survival analysis contribute to the growing body of evidence that indicate that CAD is a more severe disease than previously thought," said Sigbjørn Berentsen, MD, PhD, Consultant Hematologist, Department of Research and ...

Serological researches on serum and mucus in Shigellosis, Salmonellosis and other bacterial enteric infectious diseases during the period of 1 October 1962 through 31 December 1962 are summarized as follows: The cross Latex agglutination test Shigella flexneri v-x and Shigella flexneri v-y infected cases using both standard strains showed that the Latex agglutinin titers against Shigella flexneri v-y strain were higher than against Shigella flexneri v-x strain. The agglutinin titers of serum and mucus against Escherichia coli, E. aerogenes, Citrobacter, A. liquefaciens isolated from the feces of the gastroenteritis patients were mostly low except the agglutinin titer against A. liquefaciencs. The Latex agglutinin titers of serum and mucus against pathogenic halophilic vibrio-like bacilli were generally low in both serum and mucus. These strains were isolated from the feces of patients suffering from food-poisoning. (Author)

Background: CAD is a complement-driven autoimmune hemolytic anemia in which IgM antibodies bind to red blood cells (RBC) at lower temperatures. Although exposure to cold may precipitate RBC agglutination, we recently reported a lack of seasonal variability in anemia, hemolysis, and healthcare utilization. Thromboembolic events (TE) are a newly reported but consequential complication of CAD. The effect of colder weather on TE risk is unknown. We therefore compared TE rates in CAD patients during all seasons in the US.. Methods: CAD patients diagnosed between 2009 and 2016 were retrospectively identified within the Optum clinical-claims database. TE were identified using diagnostic codes. TE risk was compared among seasons using logistic regression adjusted for age, sex, race, region, year, Charlson Comorbidity Index, and clustering within subjects.. Results: 808 CAD patients (63% female; 66% ≥65 years old) were identified. 204 CAD patients experienced 287 TE; 56 (20%) were in summer, 57 (20%) ...

Cold Agglutinin hemolytic anemia due to cold autoantibodies associated with Mycoplasma pneumoniae is rare. Optimal treatment includes antibiotics and maintenance of a constant warm environment for the patient. This treatment, however, may result in a prolonged, costly, and confining hospitalization.. We report the use of an environmental suit for a patient with severe hemolysis due to M. pneumoniae pneumonia. This suit provides a constant temperature and has enabled the patient to journey outside his room, despite winter temperatures, yet avoid further hemolytic episodes. Early discharge was possible and has facilitated the patients return to home and work as well as ...

Since its reactivity with human erythrocytes but not with dog erythrocytes is reduced by treatment of the cells with ficin and its reactivity with both is abolished by treatment of the cells with neuraminidase, it has by definition Pr2 specificity [Roelcke, D. Peanut agglutinin (PNA) is plant lectin protein derived from the fruits of Arachis hypogaea. Peanut agglutinin may also be referred to as Arachis hypogaea lectin. Lectins recognise and bind particular sugar sequences in carbohydrates; peanut agglutinin binds the carbohydrate sequence Gal-β(1-3)-GalNAc. The name "peanut agglutinin" originates from its ability to stick together (agglutinate) cells, such as neuramidase-treated erythrocytes, which have glycoproteins or glycolipids on their surface which include the Gal-β(1-3)-GalNAc carbohydrate sequence. The protein is 273 amino acids in length with the first 23 residues acting and a signal peptide which is subsequently cleaved. It has a Uniprot accession of P02872. There are over 20 structures of this protein in the PDB which reveal and all beta-sheet protein with a tetrameric quaternary structure. It is a member of the Lectin_legB PFAM family. The ALS gene family encodes large cell-surface glycoproteins associated with C. albicans pathogenesis. Als proteins are thought to act as adhesin molecules binding to host tissues. Wide variation in expression levels among the ALS genes exists and is related to cell morphology and environmental conditions. ALS1, ALS3, and ALS4 are three of the four most highly expressed ALS genes. This work describes the production and use of specific high affinity monoclonal antibodies against Als1, Als3, and Als4 to evaluate protein production and localization at both the individual cell and population levels. Production and localization of these proteins is tightly regulated and occurs under specific growth conditions. When examined at a population level, it is clear that cells from the same culture can have different profiles of Als proteins on the cell surface. These results provide a very different view of the Als family than the view held by those who suggest that the Als family functions in antigenic ...
16 May 2003 This report represents my first set of observations with the SSP-4 (1mm detector). What I have tried to accomplish with this set of observations is to refine my observation technique so that I can make accurate and repeatable measurements. I feel this goal has been accomplished, that is, the SSP-4 photometer is capable of repeatable measurements with a predictable accuracy. I plan to add to this report as I gather more observations. In my next phase of observations I will concentrate on determination of the color transformation coefficients, airmass extinction terms, and the signal-to-noise (SNR) dependence with magnitude. Table 1 presents the results of differential photometry measurements taken with the Optec SSP-4 (1mm detector). The telescope used was a Meade 0.25m SCT in alt-az configuration. The column labeled "Comp SNR" and "Target SNR" represents the SNR of the comparison and target stars, respectively. The SNR is calculated from the ratio (average counts for n ...

In a previous paper (1) we described rather paradoxical serologic interrelationships between four variant strains of an alkaligenes bacillus that had been derived experimentally from a common ancestor. Three of these strains, namely, K-97, K-84 and K-160, morphologically diverse, were serologically reciprocal and a fourth, K-134 showed a typical group and specific agglutinin interrelationship with strains K-97 and K-84. This is the equivalent of saying that the anti-serum of K-134 apparently did not contain agglutinins for K-97 and K-84 although their antisera contained group agglutinins for K-134.. Inasmuch as such relationships are quite well known they would not have been questioned had not the strain K-160 agglutinated in the supposedly specific K-134 antiserum, but since K-160 was serologically identical with K-97 and K-84 we sought the reason why these latter two strains did not also agglutinate in the specific anti-serum of K-134.. ...

Plug it into DNAworks online software with your preferred excluded sequences (restriction sites, barcodes, screening primer sequences, etc.) and rough parameters - a rule of thumb that has worked in the past is 70 nt oligo length and ~63C annealing temp. You will get a set of primers out of the software. http://helixweb.nih.gov/dnaworks/ PAN oligo synthesis works for ordering all of these primers with enough fidelity to get a working full synthesis. This set will be an even number of oligos 1 - n, oligo number #1 and #n are the full length amplification oligos. Homologous recombination between DNA sequences residing in the chromosome and newly introduced, cloned DNA sequences (gene targeting) allows the transfer of any modification of the cloned gene into the genome of a living cell. This article discusses the current status of gene targeting with particular emphasis on germ line modification of the mouse genome, and describes the different methods so far employed to identify those rare embryonic stem cells in which the desired targeting event has occurred. ...
Methods are described for the preparation of bacterial immunoabsorbents consisting of organisms dispersed in agar beads 50 to 200 mum in diameter. The concentration of organisms could be made extremely high and columns prepared had sufficient capacity to enable them to be used for a number of purposes, but particularly for the removal of cross-reacting agglutinins from production batches of type-specific antisera. The absorptive capacity of these columns was regenerated after absorption by the elution of agglutinins with low pH buffers. Results obtained from the use of these immunoabsorbents are presented. ...
