Purpose : Mechanisms used by epidemic keratoconjunctivitis (EKC)-causing adenoviruses to overcome the mucosal barrier of the ocular surface epithelium and initiate infection remain unknown. At the 2015 ARVO General Meeting, we reported that an EKC-causing adenovirus, but not a non-EKC-causing one, induces release of the ectodomain of MUC16 - a membrane-associated mucin (MAM) with barrier functions - from human corneal epithelial cells. The purpose of this study is to determine if MUC16 ectodomain release facilitates adenoviral infection of ocular surface epithelial cells. Methods : Telomerase-immortalized human corneal-limbal epithelial (HCLE) cells, grown to confluence and allowed to differentiate for optimal mucin expression, were exposed to human adenoviruses HAdV-D37 (EKC-causing) and HAdV-D19p (non-EKC-causing) at an MOI of 3. As a measure of epithelial barrier function, rose bengal dye penetrance assays were performed on HCLE cells exposed to HAdV-D37 and HAdV-D19p for 2 h. To compare ...
Infection of mice or rats with adenoviruses can alter their normal immune response and thereby skew experimental data. For example, infection with MAD-1 can produce extensive persistent lesions in the kidneys of adult mice and render them more susceptible to experimental Escherichia coli-induced pyelonephritis. Mouse adenovirus infection has also been shown to accelerate experimental scrapie infection in mice. Although mouse adenoviral infection is usually subclinical in immunocompetent mice, wasting may result in nude mice. ...
Adenoviruses (HAdV) can cause serious life threatening infections, especially in pediatric patients after allogeneic stem cell transplantation (SCT). To improve the morbidity and mortality rate of HAdV infected patients after SCT three issues have been explored: which patients are at risk for HAdV infection, what is the best treatment for ... read more HAdV infected patients and when do we start treatment. We found that in pediatric patients HAdV-DNA positivity in nasopharyngeal aspirate before SCT is a very important risk factor for developing invasive HAdV infection after SCT. As determined with multiplex immuno-assay we also found that children with a localized HAdV infection have a different cytokine profile compared with patients with an invasive HAdV infection after SCT. Cytokine signatures can also be helpful to differentiate invasive HAdV infection from other SCT related complications as Graft versus Host Disease (GvHD) and Epstein-barr virus infection. Immune profiling could possibly ...

Mouse adenovirus causes a persistent infection in the mouse kidney that produces extensive mononuclear cell infiltrates in cortex and medulla. Tubular necrosis, dilatation, and occasional collapse occur but no glomerular changes or periglomerular fibrosis have been observed. Acute and chronic adenovirus infection of the kidney predispose the kidney to develop acute pyelonephritis when the mouse is challenged by the intravenous or retrograde route with Escherichia coli. ...
Adenovirus vectors have significant potential for long- or short-term gene transfer. Preclinical and clinical studies using human derived adenoviruses (HAd) have demonstrated the feasibility of flexible hybrid vector designs, robust expression and induction of protective immunity. However, clinical use of HAd vectors can, under some conditions, be limited by pre-existing vector immunity. Pre-existing humoral and cellular anti-capsid immunity limits the efficacy and duration of transgene expression and is poorly circumvented by injections of larger doses and immuno-suppressing drugs. This review updates canine adenovirus serotype 2 (CAV-2, also known as CAdV-2) biology and gives an overview of the generation of early region 1 (E1)-deleted to helper-dependent (HD) CAV-2 vectors. We also summarize the essential characteristics concerning their interaction with the anti-HAd memory immune responses in humans, the preferential transduction of neurons, and its high level of retrograde axonal transport in the
Doctors Ask: Adenovirus infection is exposed to all segments of the population. Basic manifestations of adenovirus infection The latent (incubation period) is usually 5-7 days. The disease most often begins acutely, from the phenomena of intoxication: slight increase in temperature, rashes in the body, chills, lethargy, headache, loss of appetite, etc.
Adenovirus infection generally may lead to common cold, pneumonia and pharyngitis. There are few types of adenoviruses. Specifically, adenoviruses types 8,19 and 37 will lead to keratoconjunctivitis mostly in adult.
In one mode of adenovirus infection (called lytic infection because it destroys large numbers of cells), adenoviruses kill healthy cells and replicate up to 1 million new viruses per cell killed, of which 1 to 5 percent are infectious. People with this kind of infection feel sick. In chronic or latent infection, a much smaller number of viruses are released, and healthy cells can multiply more rapidly than they are destroyed. People who have this kind of infection do not exhibit symptoms. Children who have normal immune systems usually experience only minor symptoms when infected with adenovirus. The course of infection tends to be more serious in children who are immunocompromised, such as those undergoing chemotherapy or those who have a disease that disrupts normal immune response (e.g. human immunodeficiency syndrome [HIV]). In such children, the virus more often affects organs such as the lungs, liver, and kidneys, and the risk of fatality increases. Symptoms common to respiratory illnesses ...

The inner-city infants had lower rates of viral detection overall. This may suggest that other factors, such as bacteria or allergic reactions to air pollutions or toxic exposures, contribute significantly to respiratory illness. Sick urban infants had lower rates of two kinds of viruses, HRV and RSV, and higher rates of adenovirus infections, compared to suburban infants. In the urban babies, 4.8 percent of nasal washes tested positive for only adenovirus, while just 0.7 percent of samples from suburban babies were positive for only adenovirus. "Adenovirus infections, either as a single pathogen or when detected in concert with other viruses, were significantly more common in the urban population, and this held true for each of the four urban locations where our study was conducted," the authors wrote ...

Inclusion body hepatitis caused by different fowl aviadenovirus (FAdV) serotypes has been described in several countries in recent years. In Spain, from the spring of 2011 to 2013, an increased number of outbreaks in broiler and broiler breeder flocks from different regions occurred. The objectives of the present work were to carry out the molecular characterization of FAdV strains from Spanish inclusion body hepatitis cases and to study the pathogenicity and viral dynamics of these strains in specific pathogen-free (SPF) chickens. A total of 52 inclusion body hepatitis clinical cases, including 45 from broiler farms and seven from broiler breeder farms, were analysed by conventional polymerase chain reaction and sequencing targeting the FAdV hexon gene. From these, 37 strains were classified as FAdV type 8b, while the remaining 15 were classified as FAdV types 11 (n = 10), 2 (n = 4) and 8a (n = 1). In addition, two different FAdVs belonging to the genotypes 8b and 11 were used for experimental ...

As combining therapeutic agents with different action mechanisms may enhance efficacy, YSC-02, an oncolytic adenovirus with multi targets was loaded with five different genes, which were designed with the expectation that different action mechanisms would cooperate. In spite of concerns regarding many APIs, YSC-02 was constructed to be an adenovirus-based anti-cancer drug. By using our own established mouse model system suitable for efficient adenoviral infection and replication, immune activity as well as survival potential could be precisely estimated for anti-cancer drug efficacy. YSC-02 was designed to decrease tumor cell survival, metastasis and to increase tumor cell death, and immune activation. It is composed of five different target genes, including one fused form and two shRNAs, but each of these genes functions is closely linked to produce the maximal antitumor effect. YSC-02 is like an organic complex designed to be applied primarily to heterogeneous liver cancer and melanoma. The ...

Adenoviruses are ubiquitous. Weve all been infected with lots of them (gets to be a problem when making gene therapy vectors). The older you are, the more likely you have been exposed to the different kinds of adenovirus, to the point where if you are an adult reading this, you probably have antibodies to serotypes 1, 2, 5, and/or 6 (depending on where you live).. Thus older children are more likely to have been exposed to any adenovirus, including Ad-36. If your obese group of children is 63% kids aged 15-18, and your healthy weight group is only 17% 15-18, I am not shocked at all that antibodies to Ad-36 is correlated with obesity. I bet you could have picked any other adenovirus serotype and found the same correlation. But they didnt look.. William M. Briggs gets it. I dunno the dude. Statistician. But he gets it. *thumbs-up*. From a virologists perspective, there are putative, plausible mechanisms for how Ad-36 could actually play a role in obesity. Those reasonings require ongoing ...

Human adenovirus, of the Adenoviridae family, is a nonenveloped icosahedral particle containing a single linear dsDNA genome. Human adenovirus comprises 6 species (A through F) consisting of serotypes 1 - 51, and associated with a variety of clinical illnesses. Serotypes 1-39, 42-51 are associated with a variety of respiratory disease, generally in children and immunocompromised persons; whereas serotypes 40 and 41 is associated with enteric disease (enteric adenovirus) particularly in children - these enteric adenoviruses will not be detected with the assay described here. Serotypes 12, 18, 31 have a high oncogenic potential and serotypes 4 and 7 are associated with acute respiratory disease (ARD) frequently in military recruits and typically occur in the winter and spring. Smaller outbreaks of serotypes 3, 4, and 7 occur in the summertime and are associated with contaminated swimming pool water.. Adenovirus infections of the eye may lead to pharyngo-conjunctival fever, follicular ...
Corneal subepithelial infiltrates are a known complication of adenoviral conjunctivitis.1 These lesions usually become apparent within 10-14 days after onset of symptoms and in some cases may persist for months or even years after the acute phase of the infection. Although the opacities gradually fade with time, those associated with reduced visual acuity may require a course of topical corticosteroids. However, return of the opacities can be seen with discontinuation of the corticosteroids.2 In cases of prolonged follicular conjunctivitis, equivocal ocular signs, or suspected superimposed infections, specimen culture is an important tool to aid diagnosis.3,4. Although small star-shaped ulcers (herpetic stellates) have been documented as a clinical manifestation seen in herpes simplex eye infections,5 these lesions have been confined to the corneal epithelium. To our knowledge, no such lesions have been documented in the tarsoconjunctiva, either in adenoviral or herpes simplex viral ...

The present study describes the development of a SYBR Green based real-time polymerase chain reaction (real-time PCR) for detection and quantitation of all fowl adenovirus (FAdV) species. Primers were designed based on conserved nucleotide sequences within the 52K gene. Ten-fold serial dilutions of a vector DNA were used as standard for quantitation. The real-time PCR had an efficiency of 98%, a regression squared value of 0.999 and showed a range of 6.73-6.73×10(8) copies of FAdV DNA per reaction. The assay was highly specific for FAdVs and an exact quantitation of all 5 FAdV species (FAdV-A to FAdV-E) could be demonstrated. It was shown, that twelve FAdV serotypes (FAdV-1 to 8a, and 8b to 11) were detectable and quantifiable. Other viral genomes as well as uninfected chicken embryo liver (CEL) cells did not produce positive signal. Cloacal swabs were taken during the animal experiment, which was performed with all FAdV species. Shedding of FAdVs was investigated in cell culture, by ...

The oncolytic potency of CRAds is critically linked to effective adenoviral infection and replication. First, there must be efficient infection in target cells. To this end, we have shown gains in infectivity via CAR-independent infection to overcome the hurdle that most tumor cells are deficient with respect to the level of the CAR receptor. These gains provide a logical framework for addressing other biological factors related to CRAd potency, such as efficacy of replication, as studied here. Furthermore, elucidation of distinct mechanisms will allow us to develop a strategy to circumvent other barriers and improve the potency of CRAds. In the present study, we endeavored to define the replicative blocks that may be limiting the full potential of oncolytic CRAds.. We showed that immortalized cell lines support wild-type adenoviral replication to a much greater degree than primary patient tumors. Because fiber and hexon are both late proteins involved in the assembly of virions, it was expected ...
