Acute-phase proteins (APPs) are a class of proteins whose plasma concentrations increase (positive acute-phase proteins) or decrease (negative acute-phase proteins) in response to inflammation. This response is called the acute-phase reaction (also called acute-phase response). The terms acute-phase protein and acute-phase reactant (APR) are often used synonymously, although some APRs are (strictly speaking) polypeptides rather than proteins. In response to injury, local inflammatory cells (neutrophil granulocytes and macrophages) secrete a number of cytokines into the bloodstream, most notable of which are the interleukins IL1, IL6 and IL8, and TNFα. The liver responds by producing a large number of acute-phase reactants. At the same time, the production of a number of other proteins is reduced; these proteins are, therefore, referred to as negative acute-phase reactants. Increased acute-phase proteins from the liver may also contribute to the promotion of sepsis. Positive acute-phase ...
Lipopolysaccharide (LPS) binding protein (LBP) is an acute phase protein with the ability to bind and transfer LPS of Gram-negative bacteria. This soluble pattern recognition molecule represents an important defense principle of the host. Regulation of the hepatic acute phase response and its termination are important mechanisms for limiting systemic inflammatory activity of the host. Here were analyze the cooperation of Interleukin (IL)-1, IL-6, and Dexamethasone (Dex) at LBP expression in the hepatoma cell lines HuH-7 and Hep G2. The major inducer of LBP expression is IL-6. Within the LBP promoter numerously highly consensus binding sites such as AP-1, C/EBP-beta? and STAT3 are present, that confer transcriptional activity as shown by truncation and mutation experiments. Additionally, activate NF-kappaB sites activated by IL-1 were detected at the LBP promoter. By mutation experiments of the promoter furthermore were found differentially active glucocorticoid response elements (GREs). The ...
The glomerular basement membrane (GBM) and extra-cellular matrix (ECM) are essential to maintain a functional interaction between the glomerular podocytes and the fenestrated endothelial cells in the formation of the slit diaphragm for the filtration of blood. Dysregulation of ECM homeostasis can cause Focal segmental glomerulosclerosis (FSGS). Despite this central role, alterations in ECM composition during FSGS have not been analyzed in detail yet. Here, we characterized the ECM proteome changes in miR-193a-overexpressing mice, which suffer from FSGS due to suppression of Wilms’ tumor 1 (WT1). By mass spectrometry we identified a massive activation of the acute phase response, especially the complement and fibrinogen pathways. Several protease inhibitors (ITIH1, SERPINA1, SERPINA3) were also strongly increased. Complementary analysis of RNA expression data from both miR-193a mice and human FSGS patients identified additional candidate genes also mainly involved in the acute phase response. In

Most tissue-damaging processes such as infections, inflammatory diseases and malignant neoplasms are associated with a major acute phase response of the C-reactive protein (CRP) and other acute reactants e.g. AAT, AAGP, C3C, C4 HAPT. The CRP response frequently precedes clinical symptoms, including fever. In normal healthy individuals CRP is a trace protein with a range up to 5 mg/L. After onset of an acute phase response the serum CRP concentration rises rapidly and extensively. Levels of up to thousand times the normal value are associated with severe stimuli such as myocardial infarction, major trauma, surgery or malignant neoplasm. CRP has a half-life of only a few hours, making it an ideal tool for clinical monitoring ...
Interleukin 1 (IL-1) plays an important role in host defense mechanisms by increasing body temperature, inducing the synthesis of a variety of lymphokines and hepatic acute phase proteins and acting as a chemoattractant for lymphocytes. However, in some microenvironments such as injured tissue or joint spaces, elevated IL-1 levels may contribute to pathologic processes, for example, proliferation and fibrosis of tissue involved in pannus formation as well as degradation of matrix and abnormal tissue architecture. To investigate potential mechanisms that may lead to excessive production of IL-1, we have examined the ability of IL-1 to participate in an amplification event by inducing its own gene expression leading to synthesis of biologically active IL-1. When injected into rabbits, recombinant human IL-1-alpha induced biphasic fevers, and during the second temperature elevation 3 hr later, a circulating pyrogenic material was detected by passive transfer of plasma to other rabbits. Induction of ...

Turpentine in the Nose:. Use with caution and learn from my experience. I have used the turpentine in my sinus for sometime now. I slightly dilute it with water in the palm of my hand and standing up I snuff it into my sinus. It clears out any mucus and I believe it is even caring for the brain because evidently it penetrates the blood brain barrier; Therefore the biofilm melting property is quite health promoting.. Caution: I suspected Cytomegelo Virus (a herpes which can lead to Kaposis Sarcoma ( it is considered incurable due to the biofilm coating)) I am interested in mechanizing a cure.. While lying down on my side I chose to snuff a single drop of the diamond G turpentine into my nostril. If it had not been so painful, it would have been comical. The turpentine entered my sinus but also the sinus above my eye.. Instantly; The mucus drainage was profound! Sneezing violent. Entire area around my eye was bright red as well as the eye. The deep cleansing produced sinus pain and some swelling ...
BENESHT Trading Company started its commercial activity in 1392 regarding the purchase and sale of raw turpentine juice. The company is headquartered in Tehran and has offices in the western provinces of Iran to purchase the harvested crop. The company is the largest trading company currently with 300 employees in various sectors including purchasing, sales, accounting, warehousing, transportation … and having extensive relationships with more than 10,000 workers in the field of raw turpentine juice extraction. They are working and buying and exporting a huge amount of harvested raw turpentine juice (about 60%). From the very beginning of the activity, in order to eliminate intermediaries and access to the harvesters of the crop, he put extensive cooperation with the workers who harvested raw turpentine juice at the top of his agenda. For centuries, local workers have struggled to harvest. Prior to 1392, due to the lack of a reliable and well-known shopping center, these workers offered their ...
When an immune response occurs from challenge by a pathogen, a systemic acute phase or inflammatory response results that is considered to be the foundation of non-specific immunity. Antibody titers that arise as a result of the humoral response are not a direct burden on the animals pool of nutrients and are therefore nutritionally inconsequential. It is therefore the systemic acute phase response that has nutritional consequences for the animal. It is this response that is associated with a shift in nutrients use away from skeletal muscle accretion and towards hepatic secretion of acute phase proteins as well as cellular and macromolecule turnover (Klasing, 1998).. There is no question that a well-nourished bird is more immunologically competent and better able to cope with disease challenges than a poorly nourished bird. A change in feed intake leads to a change in nutrient and energy intake. The development and maturation of the immune system of young poultry fed nutritionally deficient ...

This work is focused on determining the rate controlling mechanisms of decarburisation of iron-carbon droplets with gas mixtures containing up to 10% oxygen at 1723 K. The results indicate that the decarburisation rate is limited by the transport of oxygen in the bulk gas to the melt surface. The work also shows the absence of any significant resistance to decarburisation from interfacial chemical kinetics or gas phase reactions. Phosphorus in the melt shows no influence on the rate of decarburisation. Chromium has no significant effect on the decarburisation rate ...

Low serum selenium has been associated with lower CD4 counts and greater mortality among HIV-1-seropositive individuals, but most studies have not controlled for serum albumin and the presence of an acute phase response. A cross-sectional study was conducted to evaluate relationships between serum selenium concentrations and CD4 count, plasma viral load, serum albumin, and acute phase response markers among 400 HIV-1-seropositive women. In univariate analyses, lower CD4 count, higher plasma viral load, lower albumin, and the presence of an acute phase response were each significantly associated with lower serum selenium concentrations. In multivariate analyses including all four of these covariates, only albumin remained significantly associated with serum selenium. For each 0.1 g/dl increase in serum albumin, serum selenium increased by 0.8 μg/l (p < 0.001). Women with an acute phase response also had lower serum selenium (by 5.6 μg/l, p = 0.06). Serum selenium was independently associated with serum
Results There were 42 female and 31 male patients with an age range of 18-54 years. In the comparison of laboratory parameters of the patients with or without the components of BDCAF, no significant relation could be found between headache, erythema nodosum, superficial thrombophlebitis, skin pustules, GIS and eye involvement and acute phase response parameters. However, we found significantly higher levels of ferritin in patients with arthritis and higher levels of fibrinogen in patients both with arthralgia and arthritis versus without joint involvement (p=0,01; p=0,030 and p=0,000; respectively). Also Hp levels in patients with genital ulceration and arthritis (p=0,005 and p=0,002) and Cp levels in patients with oral ulceration and arthritis were found to be higher compared to patients without these involvements (p=0,01 and p=0,001). Prealbumin as a negative acute phase reactant was determined as significantly lower in patients with joint involvement than the patients without it (p=0,023). ...

The serum protein 1-acid glycoprotein (AGP), also known as orosomucoid, is normally referred to as an archetypical positive acute phase protein. as analyzed by 2-D electrophoresis. This MAb was utilized to build up an immunoassay (ELISA) for quantification of AGP in pig serum samples. The adult serum concentrations of pig AGP had been in the number of 1C3 mg/ml in several regular pig breeds although it was low in G?ttingen and Ossabaw minipigs (in the 0.3 to 0.6 mg/ml vary) and higher in young (2C5 times old) regular pigs (mean: 6.6 mg/ml). Amazingly, pig AGP was discovered to Betanin work as a poor acute phase proteins during a selection of experimental infections and aseptic irritation with significant decreases in serum focus and in hepatic ORM1 expression through the acute stage response. To your understanding this is actually the first explanation in virtually any species of AGP being truly a negative severe phase protein. The host inflammatory response against infections is characterized by the release of pro-inflammatory cytokines and acute-phase proteins, driving both innate and adaptive arms of the immune response. Distinct patterns of circulating cytokines and acute-phase responses have proven indispensable for guiding the diagnosis and management of infectious diseases. This review discusses the profiles of acute-phase proteins and circulating cytokines encountered in viral and bacterial infections. We also propose a model in which the inflammatory response to viral (IL-18/ferritin) and bacterial (IL-6/CRP) infections presents with specific plasma patterns of immune biomarkers ...
C-reactive protein is a serum acute-phase reactant that increases several thousand-fold in concentration during inflammation in most mammals. However, mouse C-reactive protein is considered to be a minor acute-phase reactant, since its blood level increases only from approx. 0.1 to 1-2 micrograms/ml. A mouse genomic clone of approximately 5 kb was obtained to determine the molecular basis for the regulation of the expression of mouse C-reactive protein. Several cis-acting elements in the 5′ flanking region that potentially regulate transcription were identified: two glucocorticoid-responsive elements, two CCAAT-enhancer-binding protein C (C/EBP) consensus elements that are required for the interleukin-1 responsiveness of some acute-phase reactant genes, an interleukin-6-responsive element, two hepatocyte nuclear factor-1 (HNF-1) elements and a single heat-shock element. Transfection of the hepatoma cell line Hep 3B.2 with a pCAT expression vector containing the 5′ flanking sequence from ...

CRP is an acute-phase plasma protein that increases during systemic inflammation. CRP is produced primarily in the liver and is regulated by proinflammatory cytokines, especially IL-6. In turn, we have shown here that CRP induces the release of IL-6 by PBMC and purified CD14+ monocytes, envisioning a feed-forward loop between CRP and IL-6. CRP levels in blood are normally very low and difficult to detect in healthy individuals, but increase rapidly with inflammation. Increased CRP concentrations have been reported in many diseases, including cardiovascular diseases, diabetes, inflammatory bowel diseases, arthritis, and many cancers. With respect to the CRP association with cancer (45), the robust association described between circulating levels of CRP and cancer risk suggest alternative possibilities: (i) elevated CRP might have a causal role in carcinogenesis, (ii) elevated CRP levels might be the result of an underlying cancer, and (iii) a third factor, such as chronic inflammation, might ...
Figure 1- The components of the innate immune system. Sentinel cells such as the macrophage detect potential environmental threats from infection, chemicals, and foods by PRRs that activate signaling pathways and release proinflammatory cytokines (IL-6 and TNF-α). Known PRRs include TLR-4, which senses bacterial LPS and the receptor for AGEs. Cytokines stimulate acute-phase protein production from the liver and also act on the brain to release adrenocorticotrophic hormone (and thereby cortisol from the adrenal gland) and activate the sympathetic nervous system with the release of catecholamines. Psychological stress can cause an acute-phase response via innervation of cytokine-producing cells and via activation of the sympathetic nervous system and adrenergic receptors on macrophages. Central cytokine-induced sickness behavior includes lethargy, sleep changes, and depression. The innate immune system also controls the adaptive (acquired) immune system via costimulatory molecule expression ...

The NFκB pathway is complex and regulates many downstream effects depending on the type of stimulating ligand and cell context. [13] TLR9 can be activated by mycobaterial DNA. Further, intracellular viral ssRNA and dsRNA activate the inflammation that induces cleavage of the pro-form of IL-1β into its mature active form.95 In fact, mice lacking the TLR3, which recognizes dsRNA and activates cytokine synthesis, if infected with mouse-adapted influenzavirus have attenuated increases in NREM sleep, hypothermia, and body weight loss compared to the prototypical responses of normal mice infected with influenza.96 Further, intracellular dsRNA that is produced during viral replication is also recognized by cytoplasmic PAMPs.97 Thus, PAMPs play a crucial role in the recognition of pathogens that lead to the production of sleep regulatory cytokines and the acute phase response to infections. Potentially, mitochondria simply act as a physical scaffold that promotes inflammasome assembly. By continuing ...

The NF-κB family of transcription factors are conserved from flies to humans and regulate the expression of a wide variety of cellular and viral genes (reviewed in reference (1)). Biochemical and molecular characteristics of NF-κB and their activation pathways have been extensively studied, especially in terms of immune and acute phase responses. The mammalian NF-κB proteins, RelA (p65), c-Rel, RelB, p50/p105, and p52/p100, share the rel homology domain which facilitates dimerization, nuclear translocation and DNA binding. Transactivation domains are also present at the COOH termini of RelA, c-Rel, and RelB. These NF-κB proteins form multiple interchangeable heterodimers and homodimers and their activity is regulated by binding of IκB inhibitory factors which determine the localization of NF-κB dimers, either in the cytoplasm or in the nucleus. Upon activation, NF-κB dimers dissociate from IκB and translocate to the nucleus and then bind to the κB sites in promoters and enhancers of ...
