Miniature Bull Terriers are prone to many health problems (physiologic and psychologic) anyone considering adopting a bull terrier must be aware of these issues! Deafness occurs in both coloured and white Miniature Bull Terriers. Puppies can be born unilaterally deaf (deaf in one ear) or bilaterally deaf (deaf in both ears). Deaf dogs should not be bred due to deafness being hereditary.[1] BEAP (or BAER) testing is done on puppies prior to sale to discover which puppies have hearing problems. LAD or LETHAL ACRODERMATITIS in bull terriers. Is a LETHAL genetic degenerative disease. It starts appearing in puppies around the age of 4 weeks. The usual black color of affected puppies will start to appear diluted and turn liverish, due to a zinc deficiency which result in terrible skin condition, dysfunction of their overall system and eventually premature death. A new DNA test was released to test breeding stock and ensure LAD CLEAR puppies. ...

The association between serum zinc level and preeclampsia (PE) remains controversial. A systematic literature search was performed in PubMed, Web of Science and Embase for relevant available articles. The articles were limited to those in English from January 1990 to April 2015. Observational studies evaluating the association between serum zinc level and PE were included. The I2 was used to assess heterogeneity and the random effect model (REM) was adopted as the pooling method. The pooled standard mean difference (SMD) with 95% confidence interval (CI) was used to estimate the association between serum zinc level and PE. Seventeen observational studies were included. Compared with healthy pregnancy controls, PE patients have lower serum zinc level in 14 studies about total PE (SMD (95% CI): −0.587 (−0.963, −0.212), Z = 3.06, p for Z = 0.002; I2 = 88.4%, p for I2 < 0.0001). In subgroup analysis, a lower serum zinc level in PE patients compared with healthy pregnancy controls was observed in
Given the structural essential, catalytic and co-catalytic played by zinc in many sections of protein metabolism, carbohydrate and lipid (zinc is involved in the function of more than 300 metalloenzymes and metalloproteins), one can imagine the impact of a deficiency or even a sub-chronic zinc deficiency on the health of the individual. Studies multiply that show that, long-term, marginal zinc deficiency is a risk factor for the development of cancer or neurodegenerative complex diseases (eg Alzheimers disease). In addition, the short-term zinc deficiencies foster the development of skin conditions and susceptibility to viral and bacterial infections. The aim of this project is to identify, in the population of patients with pseudo-acrodermatitis enteropathica (AE) tested in the investigators laboratory, rare variants (mutations real epimutations or polymorphisms) located in solute carrier family 39 member 4 (SLC39A4) gene or in 55 other genes chosen for their role in zinc homeostasis ...
In dermatology, there are typical examples of disorders related to dietary factors. The pathogenic, links between nutritional factor(s) and skin disease may be different.. Nutrient deficiency and nutrient excess are the simplest causes of specific diet-related cutaneous changes: scurvy (vitamin C deficiency) and acrodermantis enteropathica (zinc deficiency) are examples of the first type, and caronenoderma (carotene excess) is an example of the second type. Genetic metabolic defects or enzyme deficiencies, although subtle, may pave the way for the onset of diet-related skin disorders, where a toxic effect is exerted by the dietary factor(s): alcohol intake is responsible for porphyria curanea tarda, and the ingestion of choline- and lecithin-containing foods is the cause of eccnine bromhidrosis, with the typical fishy odor that features trimethylaminuria. More often, an immune (and complex) mechanism is involved in the pathogenesis of strictly diet-dependent skin disorders, e.g. atopic ...
Mechanisms through which gene expression is regulated by zinc are central to cellular zinc homoeostasis. In this context, evidence for the involvement of zinc dyshomoeostasis in the aetiology of diseases, including Type 2 diabetes, Alzheimers disease and cancer, highlights the importance of zinc-regulated gene expression. Mechanisms elucidated in bacteria and yeast provide examples of different possible modes of zinc-sensitive gene regulation, involving the zinc-regulated binding of transcriptional activators and repressors to gene promoter regions. A mammalian transcriptional regulatory mechanism that mediates zinc-induced transcriptional up-regulation, involving the transcription factor MTF1 (metal-response element-binding transcription factor 1), has been studied extensively. Gene responses in the opposite direction (reduced mRNA levels in response to increased zinc availability) have been observed in mammalian cells, but a specific transcriptional regulatory process responsible for such a ...
Isolation of cells differentially expressing lacZ activity: After development of the X-gal color for 2 wk, the membranes were photographed using Ektapan (Kodak, 4162) film and the resulting pairs of negatives were superimposed with a slight offset and examined by eye against a clear incandescent lightbulb. By applying strips of removable tape, the negatives were readily scanned for colonies putatively exhibiting differential lacZ expression. Colonies on the master plate were located by comparison with prints from the negatives. To purify the clones and document zinc-regulated lacZ expression, clones of interest were dispersed into 1 ml of low-zinc, Leuselective medium and 1 μl of this suspension spread as sectors on another low-zinc, Leu-selective plate. After colonies reached full size, the replica-plating procedure was repeated. About 70% of the clones exhibited perceptible differences in X-gal color in the two growth conditions and were kept for further study.. Identification of sequences ...

Poikiloderma vasculare atrophicans, or PVA, indicates that extra or altered skin pigmentation (poikiloderma)[10] is occurring, associated with heightened visibity of capillaries (vasculare, referring to telangiectasia) under the skin, related to thinning and wasting away (atrophicans) of the skin and its tissue. Telangiectasia is an enlargement of capillaries underneath the skin.[10] PVA also has common names that include parapsoriasis-related terminology (i.e. parapsoriasis variagata, or variegated parapsoriasis).[5] Parapsoriasis is a term first used by Brocq in 1902,[11] intended to represent a group comprising a number of uncommon skin disorders, under a once used, now antiquated classification scheme for all inflammatory dermatoses (skin diseases known to be associated with or cause inflammation).[5] Brocq chose the term parapsoriasis to illustrate that the dermatoses placed in this group had or would have commonalities with psoriasiasis, including appearance and chronicity ...

The zinc transporter ZIP13, also known as SLC39A13, is a member of a family of divalent ion transporters. Zinc is an essential ion for cells and plays significant roles in the growth, development, and differentiation. The zinc transporter family is divided into four subfamilies (I, II, LIV-1 and gufA). ZIP13 is a multipass membrane protein that belongs to the ZIP transporter subfamily LIV-1. Mutations in ZIP13 have recently been shown to cause a spondylocheiro dysplastic form of Ehlers-Danlos syndrome (SCD-EDS), a generalized skeletal dysplasia involving mainly the spine with clinical abnormalities of the hands in addition to EDS-like features. Other experiments have shown that ZIP13 is required for proper connective tissue development and is involved in BMP/TGF- signaling pathways ...
complexation with the protein transferrin (Tf). Upon entering the blood, Mn is distributed diffusely throughout the body, with the brain, bone, kidneys and liver acquiring the highest accumulation. A variety of transporters regulate Mn import, including DMT1 or Tf-Tf receptor internalization, choline transporter, citrate transporter, voltage-gated and store-operated calcium channels, and the zinc transporters ZIP8/14. Recent findings suggest a role for the magnesium transporter HIP14 and the P-type transmembrane ATPase ATP13A2 in Mn uptake across cellular membranes. Mn serves as an important cofactor for many enzymes that are key in regulating general cellular function. Intracellular buffering mechanisms also include preferential sequestration within mitochondria. Finally, Mn export has yet to be fully understood, but new evidence points to the iron exporter ferroportin as a potential exporter. This chapter will address the various processes associated with maintenance of optimal Mn levels and ...
Thirty-four HIV/AIDS patients at various stages of disease progression volunteered to manage their health using a nutritional supplement that contained several micronutrients that included a 15 nig daily dose of elemental zinc. This initial publication only focuses on trends in the serum zinc levels and the observed biochemical changes following intervention, considering the critical role this trace element plays in human immunity. At baseline and after 30 months of follow-up, the patients serum zinc levels were determined as was their clinical status. Four women who were found to be HIV negative at baseline and who had lost their husbands to HIV/AIDS, yet they had regularly had un-protected sex with them, had a mean serum zinc level of 116.2 ± 32.7 meg/100 ml. The serum zinc levels of asymptomatic, moderately symptomatic and severely symptomatic HIV/AIDS patients in the cohort reduced from baseline to post intervention levels of 92.5+12.1 to 78.0 + 8.2 meg/100 ml (P = 0.056); 81.9+ 17.6 to ...

Thirty-nine years ago, my great-uncle, Reverend Dr. Martin Luther King Jr., delivered to mindful citizens of America a prophetic dream which envisaged all of this great nations people living in the full exercise of all rights granted to them by the U.S. Constitution. My forebears dream was deeply rooted in the American dream wherein the Founders of our Union saw every lawful citizen standing in dignity outfitted by unalienable rights from God to life, liberty and the pursuit of happiness free from the tyrannies of personal whim and ideology handed down by kings, dictators, political groups, and yes, some institutions of democratic government.... Yet, there is a larger body of science represented even by the above scientists that agrees that mature behavior patterns rely much more on social shaping and choices than genetic predisposition, such that these behaviors may be successfully modified by a variety of means. Zinc deficiency is defined either as insufficient zinc to meet the needs of the body, or as a serum zinc level below the normal range. However, since a decrease in the serum concentration is only detectable after long-term or severe depletion, serum zinc is not a reliable biomarker for zinc status. Common symptoms include increased rates of diarrhea, pneumonia, and malaria.[citation needed] Zinc deficiency affects the skin and gastrointestinal tract; and the central nervous, immune, skeletal, and reproductive systems. Zinc deficiency in humans is caused by reduced dietary intake, inadequate absorption, increased loss, or increased body system utilization. The most common cause is reduced dietary intake. In the U.S., the Recommended Dietary Allowance (RDA) is 8 mg/day for women and 11 mg/day for men. Zinc plays an essential role in numerous biochemical pathways. The highest concentration of dietary zinc is found in oysters, meat, beans, and nuts. Increasing the amount of zinc in the soil and thus ...

In this randomized controlled trial, we did not find any significant difference in the need for antibiotics, intravenous antibiotics, hospitalization or colonization with Pseudomonas among children with cystic fibrosis receiving zinc supplementation of 30 mg daily as compared with those receiving placebo.. There is a paucity of data on the role of zinc in cystic fibrosis. It is thought that steatorrhea causes loss of zinc in the stool and should lead to low zinc levels among children with cystic fibrosis. However, no such relation was found in the study on 36 children with cystic fibrosis by Van Biervliet et al.14 Another study reported that serum zinc levels in children with cystic fibrosis were similar to those of normal controls but that children with lower serum zinc levels had significantly worse results on the pulmonary function tests.15 An increase has been observed in the sputum zinc levels in pulmonary exacerbations, and a decrease has been observed in the levels after treatment in ...

A mouse cDNA clone, M96, encoding a metal-regulating-element (MRE)-binding protein, was analysed for its ability to act as a metal-regulated transcription factor. The metal depletion of a glutathione S-transferase (GST)-M96 fusion protein showed that Zn2+ ions modulate the MRE-binding activity, suggesting that the M96-encoded protein is a Zn2+-regulated factor (ZiRF1). The methylation interference assay showed the specific interactions of ZiRF1 with the MRE, MREd/c, present on the mouse metallothionein Ia promoter. Point mutations of the MREd/c nullified the metal-regulatory properties of this region. In mouse L-cell nuclear extracts, mobility-shift assays revealed a Zn2+-dependent MRE-binding complex (MBC) with DNA-recognition properties similar to those of ZiRF1. Antibodies raised against purified GST-ZiRF1 were able to specifically recognize MBC in Western-blot analyses. Competition analysis of MRE-binding proteins from mouse NIH3T3 cells with oligonucleotide matching the binding sites for ...

Zinc is one of the essential micronutrients, and plays an important role in human nutrition and health. In 1961, Prasad first recognized zinc deficiency as the cause of dwarfism and hypogonadism among iron-deficient adolescent Iranian village boys.1,2. Zinc deficiency occurs in individuals and populations with diets low in sources of readily bioavailable zinc, such as red meat, and high in unrefined cereals that are rich in phytate. The elderly population is potentially vulnerable to zinc deficiency because of decreased intake of food energy, protein, vitamins and minerals, and increased intake of carbohydrates.3,4 In this review, we discuss the importance of zinc to humans, as well as the causes, clinical features and management of zinc deficiency in the elderly population.. Among the first-line defense genes, ZRT1, ZRT2, and FET4 are induced by mild zinc deficiency to increase the ability of the cell to accumulate zinc from its environment. ZPS1 is also induced under these mild conditions and we have previously proposed that its product, a secreted protein related to metalloproteases, may also be involved in zinc acquisition by degrading extracellular proteins and releasing any bound metals [3]. Similarly, ZRT3 is up-regulated by mild zinc deficiency and also responds rapidly to zinc withdrawal. Thus, mobilization of zinc stores from the vacuole is also a first-line response. Induction of the YOR387C gene in response to mild zinc deficiency suggests that this gene is also involved in the first-line defense against zinc limitation. The function of this protein is not yet known but its pattern of regulation by zinc suggests that it may play a role in zinc uptake or vacuolar zinc export. TIS11 ( also known as CTH2) is also induced in LZM + 300 μM ZnCl2. Tis11 binds ...
