Amino acid decarboxylation activity in dispersed rat pancreas acinar cells and fractions derived by differential centrifugation of homogenate of these cells was studied. The rate of decarboxylation was measured by determining the rate of production of the [3H]-amine from [3H]-amino acid or the rate of production of 14CO2 from the [14C]-carboxy-labelled amino acid. Only the hydroxylated amino acids L-dopa and 5-hydroxytryptophan are decarboxylated by intact dispersed pancreas acinar cells or cell homogenates at all pH values and amino acid concentrations tested. The decarboxylase activity is located exclusively in the cell cytosol. Each substrate competitively inhibits the decarboxylation of the other and the decarboxylation of each is inhibited by NSD-1055. The estimated Km and Vmax are, for L-dopa, 4.8 X 10(-5) M and 2.5 nmol/mg protein/min and for 5-hydroxytryptophan, 2.9 X 10(-5) M and 0.3 nmol/mg protein/min. The pH optimum for 5-hydroxytryptophan decarboxylation is from 7.0-8.5 while that for L

Enhances brain serotonin 5-Hydroxytryptophan (5-HTP) is an intermediate in the natural synthesis of the essential amino acid, tryptophan, to serotonin. The enzyme tryptophan hydroxylase adds a hydroxyl group (OH) to tryptophan, forming the 5-HTP intermediate. In the body, it converts to serotonin with the removal of a carboxyl group (COOH) by a second enzyme. Serotonin is an important neurotransmitter involved in the regulation of endocrine and brain activity responsible for emotion, appetite and sleep/wake cycles. In clinical studies, administration of 5-HTP supported serotonin production. The 5-HTP supplied in this supplement is derived from the Griffonia simplicifolia plant. Supplementation with 5-HTP encourages brain serotonin levels that can lead to positive effects on emotional well-being, appetite control, and wake/sleep cycles. each vegetable capsule contains: 5-hydroxytryptophan (Griffonia simplicifolia) 100 mg. BACKGROUND Autism is accompanied by peripheral and central disorders in the metabolism of serotonin (5-HT). The present study examines plasma dehydroepiandrosterone-sulphate (DHEA-S) and the cortisol/DHEA-S ratio following administration of L-5-hydroxytryptophan (5-HTP), the direct precursor of 5-HT, to autistic patients. METHODS Plasma DHEA-S levels were determined both before and after administration of 5-HTP or placebo, on two consecutive days in a single blind order in 18 male autistic patients and 22 matched healthy controls. RESULTS The 5-HTP-induced DHEA-S responses were significantly higher in autistic patients than in controls. In baseline conditions, the cortisol/DHEA-S ratio was significantly higher in autistic patients than in controls. DISCUSSION The results suggest that autism is accompanied by a major disequilibrium in the serotonergic system. The increased Cortisol (neurotoxic) versus DHEA-S (neuroprotective) ratio suggests that an increased neurotoxic potential occurs in autism.
5-Hydroxytryptophan (5-HTP) is the intermediate metabolite produced when the body initiates the process of converting the essential amino acid L-tryptophan to serotonin. Serotonin levels in the body are associated with the regulation of sleep, mood, aggression, appetite, temperature, sexual behavior, and pain sensation. Supplementing with 5-HTP to facilitate serotonin production is preferable to supplementing with L-tryptophan because, unlike L-tryptophan, 5-HTP cannot be shunted into niacin or protein production. Supplementation with 5-HTP bypasses the conversion of L-tryptophan into 5-HTP by the enzyme tryptophan hydroxylase, the rate-limiting step in the synthesis of serotonin and a step that can be inhibited by a number of factors, including stress, insulin resistance, increasing age, and deficiencies of vitamin C, folate, and iron. 5-HTP is well absorbed when taken orally, with about 70 percent ending up in the bloodstream. Intestinal absorption of 5-HTP does not require the presence of a
5-Hydroxytryptophan (5-HTP) is the intermediate metabolite the body produces when it initiates the process of converting the essential amino acid L-tryptophan to serotonin. Therefore, supplementation with 5-HTP will bypass the conversion of L-tryptophan into 5-HTP by the enzyme tryptophan hydroxylase - which is the rate-limiting step in the synthesis of serotonin - and is a conversion that can be inhibited by a number of factors, including stress, insulin resistance, increasing age, and deficiencies of vitamin C, folate, and iron.* 5-HTP is well absorbed when taken orally, with about 70 percent going into the bloodstream.* Intestinal absorption of 5-HTP does not require the presence of a transport molecule, and its absorption is not affected by the presence of other amino acids; therefore, 5-HTP can be taken with meals without reducing its effectiveness. 5-HTP has been shown to have a beneficial impact on mood, sleep, carbohydrate cravings, and discomfort associated with fatigue.* Numerous ...

5-hydorxytryptophan (5-HTP) is a compound made from the amino acid (protein) tryptophan and is primarily found in the brain. 5-HTP is one of the basic building blocks for serotonin, a neurotransmitter (brain chemical) thought to be responsible for mood regulation.
Depression: Much of the published research on 5-HTP has to do with its use in the treatment of depression. Since the early 1970s, at least 15 studies have evaluated the clinical effects of 5-HTP on depression. These are summarized in. Taken together, these studies examined a total of 511 patients with different types of depression. Of these 511 subjects, 285 (56%) showed significant improvement while taking 5-HTP.. In addition, biochemical studies show 5-HTP is closely involved in depressive disorders. In a study employing positron-emission tomography (PET) scanning, eight healthy volunteers and six people diagnosed with major depression received infusions of radiolabelled 5-HTP. The researchers found significantly less 5-HTP crossed the blood-brain barrier into the brains of the depressed subjects than into the brains of the normal controls. The authors suggested the transport of 5-HTP across the blood-brain barrier may be compromised in major depression, which might make the brain dependent on ...
Interpretive Summary: Serotonin regulates maternal calcium homeostasis during the perinatal period of sheep. By: Anne Wallace. Serotonin is a neurotransmitter produced from the amino acid tryptophan, which is first converted to the intermediary molecule 5-Hydroxytryptophan (5-HTP). In this December 2019 study published in the Journal of Animal Science, researchers supplemented pregnant sheep with serotonin precursors L-Tryptophan and 5-HTP in order to evaluate how calcium homeostasis and gene expression was affected by this neurotransmitter.. Sheep infused with IV saline, saline with L-tryptophan, or saline with 5-HTP for seven days prepartum. Mammary tissue, femur tissue, postpartum milk and serum (pre- and post-partum) was collected and analyzed. The 5-HTP infusions increased total serum calcium and total milk calcium of sheep, compared to the control. Several transport proteins involved in calcium homeostasis e.g. the parathyroid hormone-related protein (PTHrP) sodium/calcium exchanger, were ...

Chromium is present to support healthy glucose metabolism and support food in-take regulation.. 5-Hydroxytryptophan (5-HTP) is a naturally-occurring amino acid precursor to serotonin. Numerous studies during the 90s, including those randomized, double-blind, and placebo-controlled, confirmed the safety and efficacy of 5-HTP in reducing appetite and food intake in obese healthy and non-insulin-dependent diabetic individuals. A 2006 study in mice concluded, 5-HTP-induced anorexia may be mediated by facilitation of leptin secretion. Vitamins B6 and C are important cofactors in the 5-HTP to serotonin pathway. Among the several serotonin receptors thus identified, the 5HT2C receptors are suspected in control of food intake. Mice without this receptor exhibit increased food intake ...

Positron emission tomography (PET) performed with various radiolabelled compounds facilitates the study of tumor biochemistry. If the tumor uptake of an administered tracer is greater than that of surrounding normal tissue, it is also possible to localize the tumor. In initial studies, 18F-labeled deoxyglucose (FDG) was attempted to visualize the tumors, since this tracer had been successfully used in oncology, reflecting increased glucose metabolism in cancerous tissue. However, this tracer was not to any significant degree taken up by the neuroendocrine tumors. Instead, the serotonin precursor 5-hydroxytryptophan (5-HTP) labeled with 11C was used and showed an increased uptake and irreversible trapping of this tracer in carcinoid tumors. The uptake was selective and the resolution so high that we could detect more liver and lymph node metastases with PET than with CT or octreotide scintigraphy. One problem was, however, the high renal excretion of the tracer producing streaky artifacts in the ...

The dispute, which is better with supplements, has been going on for some time. Some users say that 5-HTP guarantees better results in terms of improving mood. Others, however, believe that L-tryptophan has a stronger effect on sleep regulation. Below are a few facts in that matter.. One of the things to consider when deciding L-Tryptophan or 5-HTP supplementation is that 5-HTP is so effective that your body can start to build tolerance after some time. This has not yet been proven by researchers, but theoretically can occur after long-term use of 5-HTP. L-tryptophan on the other hand, although more subtle in its effects, may be safer and healthier for long-term use. L-tryptophan supplements are also less likely to interact with drugs than 5-HTP.. ...
