THA improves word priming and clonidine enhances fluency and working memory in Alzheimer's disease. (1/2899)

We investigated the effects of a single administration of a cholinesterase inhibitor, tetrahydroaminoacridine (THA, 25 and 50 mg, orally), and an alpha 2-agonist, clonidine (0.5 and 2 micrograms/kg, orally), on neuropsychologic performance in two groups of patients with Alzheimer's disease (AD). Clonidine enhanced a spatial working memory and verbal fluency, but had no effect on spatial span or word priming. THA enhanced word priming, but had no effect on other performance measures. Our data suggests that degeneration of the LC noradrenergic system and the cholinergic cells of the basal forebrain have different functional consequences during the progression of AD. Finally, a combined treatment with noradrenergic and cholinergic drugs might produce a qualitatively broader effect on cognitive functions than either of the treatments alone, and more effectively attenuate clinical dementia.  (+info)

Exposure to trichloroethylene III. Psychological functions. (2/2899)

The effect of exposure to the solvent trichloroethylene (TRI) on the performance of tests of numerical ability, reaction time (simple and choice), and short-term memory was studied in 15 healthy male subjects. The subjects were tested individually on three different occasions during exposure to 540 and 1,080 mg/m3 of TRI in inspiratory air and under control conditions, respectively. At predetermined times during the three 70-min exposure periods, samples were taken of the subjects' alveolar air. Neither the reaction time tests nor the short-term memory test showed any signs of performance decrement during exposure to TRI as compared to those administered under control conditions. However, a statistically significant decrement in performance was obtained on the test of numerical ability during exposure to TRI. The results as a whole indicate that there should not be any risk of an acute effect on central nervous functions at concentrations which do not considerably exceed the Swedish threshold limit value for the solvent (160 mg/m3).  (+info)

Isodirectional tuning of adjacent interneurons and pyramidal cells during working memory: evidence for microcolumnar organization in PFC. (3/2899)

Studies on the cellular mechanisms of working memory demonstrated that neurons in dorsolateral prefrontal cortex (dPFC) exhibit directionally tuned activity during an oculomotor delayed response. To determine the particular contributions of pyramidal cells and interneurons to spatial tuning in dPFC, we examined both individually and in pairs the tuning properties of regular-spiking (RS) and fast-spiking (FS) units that represent putative pyramidal cells and interneurons, respectively. Our main finding is that FS units possess spatially tuned sensory, motor, and delay activity (i. e., "memory fields") similar to those found in RS units. Furthermore, when recorded simultaneously at the same site, the majority of neighboring neurons, whether FS or RS, displayed isodirectional tuning, i.e., they shared very similar tuning angles for the sensory and delay phases of the task. As the trial entered the response phase of the task, many FS units shifted their direction of tuning and became cross-directional to adjacent RS units by the end of the trial. These results establish that a large part of inhibition in prefrontal cortex is spatially oriented rather than being untuned and simply regulating the threshold response of pyramidal cell output. Moreover, the isodirectional tuning between adjacent neurons supports a functional microcolumnar organization in dPFC for spatial memory fields similar to that found in other areas of cortex for sensory receptive fields.  (+info)

Humour appreciation: a role of the right frontal lobe. (4/2899)

Humour occupies a special place in human social interactions. The brain regions and the potential psychological processes underlying humour appreciation were investigated by testing patients who had focal damage in various areas of the brain. A specific brain region, the right frontal lobe, most disrupted the ability to appreciate humour. The individuals with damage in this brain region also reacted less, with diminished physical or emotional responses (laughter, smiling). Performance on the humour appreciation tests used were correlated in a distinct pattern with tests assessing cognitive processes. The ability to hold information in mind (working memory) was related to both verbal (jokes) and non-verbal (cartoon) tests of humour appreciation. In addition, the demands of the specific type of humour test were related in a logical manner to cognitive processes, verbal humour being associated with verbal abstraction ability and mental shifting and cartoon humour being related to the abilities to focus attention to details and to visually search the environment. The ability of the right frontal lobe may be unique in integrating cognitive and affective information, an integration relevant for other complex human abilities, such as episodic memory and self-awareness.  (+info)

Memory consolidation for contextual and auditory fear conditioning is dependent on protein synthesis, PKA, and MAP kinase. (5/2899)

Fear conditioning has received extensive experimental attention. However, little is known about the molecular mechanisms that underlie fear memory consolidation. Previous studies have shown that long-term potentiation (LTP) exists in pathways known to be relevant to fear conditioning and that fear conditioning modifies neural processing in these pathways in a manner similar to LTP induction. The present experiments examined whether inhibition of protein synthesis, PKA, and MAP kinase activity, treatments that block LTP, also interfere with the consolidation of fear conditioning. Rats were injected intraventricularly with Anisomycin (100 or 300 microg), Rp-cAMPS (90 or 180 microg), or PD098059 (1 or 3 microg) prior to conditioning and assessed for retention of contextual and auditory fear memory both within an hour and 24 hr later. Results indicated that injection of these compounds selectively interfered with long-term memory for contextual and auditory fear, while leaving short-term memory intact. Additional control groups indicated that this effect was likely due to impaired memory consolidation rather than to nonspecific effects of the drugs on fear expression. Saccades require focal attention and are facilitated by a short-term memory system. (6/2899)

We performed two sets of experiments in which observers were instructed to make saccades to an odd colored target embedded in an array of distractors. First, we found that when the colors of the target and distractors switched unpredictably from trial to trial (the mixed condition), saccadic latencies decreased with increasing numbers of distractors. In contrast, saccadic latencies were independent of the number of distractors when the color of the target and distractors remained the same on each trial (the blocked condition). This pattern of results mirrors visual search tasks in which focal rather than distributed attention is required (Bravo, M.J., Nakayama, K. (1992). Second, we found that saccades to an odd target were made more quickly and accurately when the target was the same color as on previous trials than when it changed color. This priming of the target color accumulates across five to seven trials over a period of approximately 30 s. Rapid, labile, and protein synthesis-independent short-term memory in conditioned taste aversion. (7/2899)

Short-term memory is a rapid, labile, and protein-synthesis-independent phase of memory. The existence of short-term memory in conditioned taste aversion (CTA) learning has not been demonstrated formally. To determine the earliest time at which a CTA is expressed, we measured intraoral intake of sucrose at 15 min, 1 hr, 6 hr, or 48 h after contingent pairing of an intraoral infusion of 5% sucrose (6.6 ml over 6 min) and toxic lithium chloride injection (76 mg/kg). Rats were implanted with intraoral catheters to allow presentation of taste solutions at arbitrary times. Intraoral intake was measured under conditions of long-delay, single-trial learning typical of CTA. Rats decreased intraoral intake of sucrose at 15 min after contingent pairing of sucrose and LiCl, but not after noncontingent LiCl or sucrose. Thus CTA learning can be expressed rapidly. To determine if short-term CTA memory is labile and decays in the absence of long-term memory, we measured intraoral intake of sucrose after pairing sucrose with low doses of LiCl. Rats received an intraoral infusion of 5% sucrose (6 ml/6 min); 30 min later LiCl was injected at three different doses (19, 38, or 76 mg/kg). A second intraoral infusion of sucrose was administered 15 min, 1 hr, 3 hr, 4.5 hr, 6 hr, or 48 hr later. The formation of long-term CTA memory was dependent on the dose of LiCl paired with sucrose during acquisition. Low doses of LiCl induced a CTA that decayed within 6 hr after pairing. Central administration of the protein synthesis inhibitor cycloheximide prior to LiCl injection blocked long-term CTA expression at 6 and 48 hr, but not short-term CTA expression at 1 hr. Thus, short-term memory for CTA learning exists that is acquired rapidly and independent of protein synthesis, but labile in the absence of long-term memory formation.  (+info)

Callosal and cortical contribution to procedural learning. (8/2899)

Acallosal and callosotomized subjects usually show impairments on tasks requiring bilateral interdependent motor control. However, few studies have assessed the ability of these subjects to learn a skill that requires the simultaneous contribution of each hemisphere in its acquisition. The present study examined whether acallosal and callosotomized subjects could learn a visuomotor skill that involved a motor control from either both or a single hemisphere. Eleven adult patients, six acallosal and five callosotomized, participated in this study. Seven of these patients had epileptic foci located in the frontal and/or temporal areas and one of the acallosal patients showed bilateral prefrontal atrophy following surgical removal of an orbitofrontal cyst. The performance of the experimental subjects was compared with that of 11 matched control subjects, on a modified version of a serial reaction time task developed by Nissen and Bullemer (Cogn Psychol 1987; 19: 1-32). This skill acquisition task involved bimanual or unimanual key-pressing responses to a sequence of 10 visual stimuli that was repeated 160 times. A declarative memory task was then performed to assess explicit knowledge of the sequence. None of the experimental subjects learned the task in the bimanual condition. Patients with frontal epileptic foci or orbitofrontal damage also failed to learn the task in the unimanual condition when they were using the hand contralateral to the damaged hemisphere. All other subjects, including the acallosal and callosotomized patients with temporal foci, learned the visuomotor skill as well as their controls in the unimanual condition. In spite of the absence of transfer and interhemispheric integration of procedural learning, some of the acallosal and callosotomized patients were able to learn the sequence explicitly. These findings indicate that the corpus callosum and the frontal cortical areas are important for procedural learning of a visuomotor skill. They also confirm the dissociation described by Squire (Science 1986; 232: 1612-9 and J Cogn Neurosci 1992; 4: 232-43) between the declarative and procedural memory systems and extend this dissociation to processes involving simultaneous bihemispheric co-operation.  (+info)

However, age-related changes in cognitive abilities such as working memory (WM) have a profound influence on the products of language comprehension (e.g., problem solving, learning, following instructions). At the same time, the effects of age and working memory on the moment-to-moment processes underlying language comprehension are less well understood. Background: Cognitive impairment is a very frequent and severe nonmotor symptom of Parkinsons disease (PD). Early intervention in this at-risk group for cognitive decline may be crucial for long-term preservation of cognitive functions. Twenty-nine pediatric patients (age range, 10-16 years) with working memory (WM) deficits, including children with pediatric bipolar disorder (PBD) with and without attention-deficit hyperactivity disorder (ADHD) comorbidity and children with ADHD, underwent a Cogmed WM training program. For both patient groups, WM performance on Cogmed tasks and on the Digit Span test improved significantly after training. Moreover, the PBD group improved on Trails Making Test A and on the Inhibition Scale, the Behavior Regulation Index, and the Global Executive Composite of the Behavioral Rating Inventory of Executive Function. The ADHD group improved significantly on the Trails Making Test B, the Spatial Span Test, and the Reading Fluency Test of the Woodcock-Johnson III, as well as on depressive symptoms. The present findings suggest that working memory training is beneficial not only in youths with ADHD but also in youths with PBD. Objectives: Cerebral ischemia results from a variety of causes that cerebral blood flow is reduced due to a transient or permanent occlusion of cerebral arteries. Reactive astrocytes and microglial activation plays an important role in the neuronal cell death during ischemic insult. Acupunctural treatment is effective for symptom improvement in cerebrovascular accident, including cerebral ischemia. Methods: In the present study, the effects of acupuncture at the ST40 acupoint on short-term memory and apoptosis in the hippocampal CA1 region following transient global cerebral ischemia were investigated using gerbils. Transient global ischemia was induced by occlusion of both common carotid arteries with aneurysm clips for 5 min. Acupuncture stimulation was conducted once daily for 7 consecutive days, starting one day after surgery. Results: In the present results, ischemia induction deteriorated short term memory, increased apoptosis, and induced reactive astrocyte and microglial activation.

The ability of the nervous system to retain, manipulate and use visual information which is no longer present in the external environment contributes to intelligent behaviour. A new approach to studying visual working memory has led to re-evaluation of the nature of its limitations in keeping with a finite memory resource which is flexibly distributed across space according to attentional priority. Using a novel behavioural paradigm to study visual working memory precision for sequentially presented items, I demonstrate how the resolution with which healthy subjects recall simple objects changes dynamically with each new item in the sequence. Stochastic modelling of the distribution of responses suggested that memory for earlier objects in the sequence was especially prone to failure in integration of visual features, such as orientation and colour, into complete objects. Our ability to remember what we have seen is very limited. Most current views characterize this limit as a fixed number of items-only four objects-that can be held in visual working memory. We show that visual memory capacity is not fixed by the number of objects, but rather is a limited resource that is shared out dynamically between all items in the visual scene. This resource can be shifted flexibly between objects, with allocation biased by selective attention and toward targets of upcoming eye movements. The proportion of resources allocated to each item determines the precision with which it is remembered, a relation that we show is governed by a simple power law, allowing quantitative estimates of resource distribution in a scene.

Duration of Short-Term Memory. To study how long short-term memory lasts, rehearsal must not be allowed to refresh information in short-term memory or to transfer it to long-term memory. The Peterson and Peterson procedure (also known as the Brown-Peterson procedure) minimizes rehearsal and recoding by tying up active processes in short-term memory during the retention interval (time between getting items and recalling them). A current divisive issue in psycholinguistic research centres around the precise nature of this memory resource. Selective attention, the ability to focus our cognitive resources on information relevant to our goals, influences working memory (WM) performance. Indeed, attention and working memory are increasingly viewed as overlapping constructs. Here, we review recent evidence from human neurophysiological studies demonstrating that top-down modulation serves as a common neural mechanism underlying these two cognitive operations. The core features include activity modulation in stimulus-selective sensory cortices with concurrent engagement of prefrontal and parietal control regions that function as sources of top-down signals. Notably, top-down modulation is engaged during both stimulus-present and stimulus-absent stages of WM tasks; that is, expectation of an ensuing stimulus to be remembered, selection and encoding of stimuli, maintenance of relevant information in mind and memory retrieval.

Our patient D.M. showed a spatial bias in report from VSTM, being more accurate at reporting stimuli presented in her left than her right visual field (Experiment 1). The classic modal model of memory argues that short term memory (STM) serves as the primary gateway for the formation of long term memory (LTM) representations (Atkinson & Shiffrin, 1968). Over the years, though, this model has been disregarded by many because of various incompatible results. For example, one common interpretation of this model is that STM serves as an incubator that strengthens representations through repeated rehearsal so that they can be successfully transferred to LTM. A large corpus of research indicates that exposure to stress impairs cognitive abilities, specifically executive functioning dependent on the prefrontal cortex (PFC). We collected structural MRI scans (n = 61), well-validated assessments of executive functioning, and detailed interviews assessing stress exposure in humans to examine whether cumulative life stress affected brain morphometry and one type of executive functioning, spatial working memory, during adolescence-a critical time of brain development and reorganization. Analysis of variations in brain structure revealed that cumulative life stress and spatial working memory were related to smaller volumes in the PFC, specifically prefrontal gray and white matter between the anterior cingulate and the frontal poles. On the basis of a meta-analysis of pairwise correlations between working memory tasks and cognitive ability measures, P. L. Ackerman, M. E. Beier, and M. O. Boyle (2005; see record 2004-22408-002) claimed that working memory capacity (WMC) shares less than 25% of its variance with general intelligence (g) and with reasoning ability. Behavioral evidence indicates that working memory (WM) in schizophrenia is already impaired at the encoding stage. However, the neurophysiological basis of this primary deficit remains poorly understood. Using event-related fMRI, we assessed differences in brain activation and functional connectivity during the encoding, maintenance and retrieval stages of a visual WM task with 3 levels of memory load in 17 adolescents with early-onset schizophrenia (EOS) and 17 matched controls. The amount of information patients could store in WM was reduced at all memory load levels. During encoding, activation in left ventrolateral prefrontal cortex (VLPFC) and extrastriate visual cortex, which in controls positively correlated with the amount of stored information, was reduced in patients.
