Orally exhaled nitric oxide levels are related to the degree of blood eosinophilia in atopic children with mild-intermittent asthma. (1/3761)

Increased levels of nitric oxide have been found in expired air of patients with asthma, and these are thought to be related to the airway inflammatory events that characterize this disorder. Since, in adults, bronchial inflammatory changes are present even in mild disease, the present study was designed to evaluate whether a significant proportion of children with mild-intermittent asthma could have increased exhaled air NO concentrations. Twenty-two atopic children (aged 11.1+/-0.8 yrs) with mild-intermittent asthma, treated only with inhaled beta2-adrenoreceptor agonists on demand and 22 age-matched controls were studied. NO concentrations in orally exhaled air, measured by chemiluminescence, were significantly higher in asthmatics, as compared to controls (19.4+/-3.3 parts per billion (ppb) and 4.0+/-0.5 ppb, respectively; p<0.01). Interestingly, 14 out of 22 asthmatic children had NO levels >8.8 ppb (i.e. >2 standard deviations of the mean in controls). In asthmatic patients, but not in control subjects, statistically significant correlations were found between exhaled NO levels and absolute number or percentage of blood eosinophils (r=0.63 and 0.56, respectively; p<0.01, each comparison). In contrast, exhaled NO levels were not correlated with forced expiratory volume in one second (FEV1) or forced expiratory flows at 25-75% of vital capacity (FEF25-75%) or forced vital capacity (FVC), either in control subjects, or in asthmatic patients (p>0.1, each correlation). These results suggest that a significant proportion of children with mild-intermittent asthma may have airway inflammation, as shown by the presence of elevated levels of nitric oxide in the exhaled air. The clinical relevance of this observation remains to be established.  (+info)

Increased exhaled nitric oxide on days with high outdoor air pollution is of endogenous origin. (2/3761)

The aim of this study was to assess the effect of outdoor air pollution on exhaled levels of endogenously released nitric oxide. To exclude bias from exogenous NO in the recovered exhaled air (residual NO or NO in dead volume) an experimental design was used that sampled NO of endogenous origin only. The validity of the presented experimental design was established in experiments where subjects were exposed to high levels of exogenous NO (cigarette smoke or 480 microg x m(-3) synthetic NO). Subsequent 1 min breathing and a final inhalation of NO-free air proved to be sufficient to attain pre-exposure values. Using the presented method detecting only endogenous NO in exhaled air, 18 subjects were sampled on 4 separate days with different levels of outdoor air pollution (read as an ambient NO level of 4, 30, 138 and 246 microg x m(-3)). On the 2 days with highest outdoor air pollution, exhaled NO was significantly (p<0.001) increased (67-78%) above the mean baseline value assessed on 4 days with virtually no outdoor air pollution. In conclusion, the level of endogenous nitric oxide in exhaled air is increased on days with high outdoor air pollution. The physiological implications of this findings need to be investigated further.  (+info)

Induction of reactive oxygen intermediates in human monocytes by tumour cells and their role in spontaneous monocyte cytotoxicity. (3/3761)

The present study examined the ability of human monocytes to produce reactive oxygen intermediates after a contact with tumour cells. Monocytes generated oxygen radicals, as measured by luminol-enhanced chemiluminescence and superoxide anion production, after stimulation with the tumour, but not with untransformed, cells. The use of specific oxygen radical scavengers and inhibitors, superoxide dismutase, catalase, dimethyl sulphoxide and deferoxamine as well as the myeloperoxidase inhibitor 4-aminobenzoic acid hydrazide, indicated that chemiluminescence was dependent on the production of superoxide anion and hydroxyl radical and the presence of myeloperoxidase. The tumour cell-induced chemiluminescent response of monocytes showed different kinetics from that seen after activation of monocytes with phorbol ester. These results indicate that human monocytes can be directly stimulated by tumour cells for reactive oxygen intermediate production. Spontaneous monocyte-mediated cytotoxicity towards cancer cells was inhibited by superoxide dismutase, catalase, deferoxamine and hydrazide, implicating the role of superoxide anion, hydrogen peroxide, hydroxyl radical and hypohalite. We wish to suggest that so-called 'spontaneous' tumoricidal capacity of freshly isolated human monocytes may in fact be an inducible event associated with generation of reactive oxygen intermediates and perhaps other toxic mediators, resulting from a contact of monocytes with tumour cells.  (+info)

Rapid film-based determination of antibiotic susceptibilities of Mycobacterium tuberculosis strains by using a luciferase reporter phage and the Bronx Box. (4/3761)

Detecting antibiotic resistance in Mycobacterium tuberculosis is becoming increasingly important with the global recognition of drug-resistant strains and their adverse impact on clinical outcomes. Current methods of susceptibility testing are either time-consuming or costly; rapid, reliable, simple, and inexpensive methods would be highly desirable, especially in the developing world where most tuberculosis is found. The luciferase reporter phage is a unique reagent well-suited for this purpose: upon infection with viable mycobacteria, it produces quantifiable light which is not observed in mycobacterial cells treated with active antimicrobials. In this report, we describe a modification of our original assay, which allows detection of the emitted light with a Polaroid film box designated the Bronx Box. The technique has been applied to 25 M. tuberculosis reference and clinical strains, and criteria are presented which allow rapid and simple discrimination among strains susceptible or resistant to isoniazid and rifampin, the major antituberculosis agents.  (+info)

Evidence that halogenated furanones from Delisea pulchra inhibit acylated homoserine lactone (AHL)-mediated gene expression by displacing the AHL signal from its receptor protein. (5/3761)

Acylated homoserine lactone (AHL)-mediated gene expression controls phenotypes involved in colonization, often specifically of higher organisms, in both marine and terrestrial environments. The marine red alga Delisea pulchra produces halogenated furanones which resemble AHLs structurally and show inhibitory activity at ecologically realistic concentrations in AHL bioassays. Evidence is presented that halogenated furanones displace tritiated OHHL [N-3-(oxohexanoyl)-L-homoserine lactone] from Escherichia coli cells overproducing LuxR with potencies corresponding to their respective inhibitory activities in an AHL-regulated bioluminescence assay, indicating that this is the mechanism by which furanones inhibit AHL-dependent phenotypes. Alternative mechanisms for this phenomenon are also addressed. General metabolic disruption was assessed with two-dimensional PAGE, revealing limited non-AHL-related effects. A direct chemical interaction between the algal compounds and AHLs, as monitored by 1H NMR spectroscopy, was shown not to occur in vitro. These results support the contention that furanones, at the concentrations produced by the alga, can control bacterial colonization of surfaces by specifically interfering with AHL-mediated gene expression at the level of the LuxR protein.  (+info)

Reduction of serum cholesterol and hypercholesterolemic atherosclerosis in rabbits by secoisolariciresinol diglucoside isolated from flaxseed. (6/3761)

BACKGROUND: Secoisolariciresinol diglucoside (SDG) is a plant lignan isolated from flaxseed. Lignans are platelet-activating factor-receptor antagonists that would inhibit the production of oxygen radicals by polymorphonuclear leukocytes. SDG is an antioxidant. Antioxidants studied thus far are known to reduce hypercholesterolemic atherosclerosis. The objective of this study was to determine the effect of SDG on various blood lipid and aortic tissue oxidative stress parameters and on the development of atherosclerosis in rabbits fed a high-cholesterol diet. METHODS AND RESULTS: Rabbits were assigned to 4 groups: group 1, control; group 2, SDG control (15 mg. kg body wt-1. d-1 PO); group 3, 1% cholesterol diet; and group 4, same as group 3 but with added SDG (15 mg. kg body wt-1. d-1 PO). Blood samples were collected before (time 0) and after 4 and 8 weeks of experimental diets for measurement of serum triglycerides, total cholesterol (TC), and LDL, HDL, and VLDL cholesterol (LDL-C, HDL-C, and VLDL-C). The aorta was removed at the end of the protocol for assessment of atherosclerotic plaques; malondialdehyde, an aortic tissue lipid peroxidation product; and aortic tissue chemiluminescence, a marker for antioxidant reserve. Serum TC, LDL-C, and the ratios LDL-C/HDL-C and TC/HDL-C increased in groups 3 and 4 compared with time 0, the increase being smaller in group 4 than in group 3. Serum HDL-C decreased in group 3 and increased in group 4 compared with time 0, but changes were lower in group 3 than in group 4. SDG reduced TC and LDL-C by 33% and 35%, respectively, at week 8 but increased HDL-C significantly, by>140%, as early as week 4. It also decreased TC/LDL-C and LDL-C/HDL-C ratios by approximately 64%. There was an increase in aortic malondialdehyde and chemiluminescence in group 3, and they were lower in group 4 than in group 3. SDG reduced hypercholesterolemic atherosclerosis by 73%. CONCLUSIONS: These results suggest that SDG reduced hypercholesterolemic atherosclerosis and that this effect was associated with a decrease in serum cholesterol, LDL-C, and lipid peroxidation product and an increase in HDL-C and antioxidant reserve.  (+info)

Expression of T lymphocyte p56(lck), a zinc-finger signal transduction protein, is elevated by dietary zinc deficiency and diet restriction in mice. (7/3761)

Compromised immune function is common to Zn deficiency, protein and energy malnutrition; however, the causative mechanisms at the molecular level have not been elucidated. The T lymphocyte signal transduction pathway contains several Zn-finger proteins, and it is possible that the in vivo functioning of these proteins could be affected by dietary deficiency of Zn and amino acids. Thus, the objective was to investigate the effects, on expression of the T lymphocyte signal transduction proteins p56(lck), phospholipase Cgamma1 (PLCgamma1) and protein kinase C (PKCalpha), of dietary Zn deficiency (ZnDF, < 1 mg Zn/kg diet) and protein-energy malnutrition syndromes [2% protein deficiency (LP), combined Zn and 2% protein deficiency (ZnDF+LP), and diet restriction (DR, body weight equal to ZnDF)] compared with control (C) mice. Indices of nutritional status and splenocyte counts were also determined. Based on serum albumin and liver lipid concentrations, the ZnDF+LP and LP groups had protein-type malnutrition, whereas the ZnDF and DR groups had energy-type malnutrition. For Western immunoblotting of the signal transduction proteins, mouse splenic T lymphocytes were isolated by immunocolumns. The expression of T lymphocyte p56(lck) was significantly elevated in the ZnDF+LP, ZnDF and DR groups compared to the C group. In contrast, the expression of PLCgamma1 and PKC was unaffected. There was a significant negative correlation between T lymphocyte p56(lck) expression and serum Zn (r= -0.65, P = 0.0007) or femur Zn (r = -0.73, P = 0.0001) concentrations. We propose that elevated T lymphocyte p56(lck) may contribute to altered thymoctye maturation, apoptosis and lymphopenia in Zn deficiency and protein-energy malnutrition syndromes.  (+info)

Intracellular trafficking pathways in the assembly of connexins into gap junctions. (8/3761)

Trafficking pathways underlying the assembly of connexins into gap junctions were examined using living COS-7 cells expressing a range of connexin-aequorin (Cx-Aeq) chimeras. By measuring the chemiluminescence of the aequorin fusion partner, the translocation of oligomerized connexins from intracellular stores to the plasma membrane was shown to occur at different rates that depended on the connexin isoform. Treatment of COS-7 cells expressing Cx32-Aeq and Cx43-Aeq with brefeldin A inhibited the movement of these chimera to the plasma membrane by 84 +/- 4 and 88 +/- 4%, respectively. Nocodazole treatment of the cells expressing Cx32-Aeq and Cx43-Aeq produced 29 +/- 16 and 4 +/- 7% inhibition, respectively. In contrast, the transport of Cx26 to the plasma membrane, studied using a construct (Cx26/43T-Aeq) in which the short cytoplasmic carboxyl-terminal tail of Cx26 was replaced with the extended carboxyl terminus of Cx43, was inhibited 89 +/- 5% by nocodazole and was minimally affected by exposure of cells to brefeldin A (17 +/-11%). The transfer of Lucifer yellow across gap junctions between cells expressing wild-type Cx32, Cx43, and the corresponding Cx32-Aeq and Cx43-Aeq chimeras was reduced by nocodazole treatment and abolished by brefeldin A treatment. However, the extent of dye coupling between cells expressing wild-type Cx26 or the Cx26/43T-Aeq chimeras was not significantly affected by brefeldin A treatment, but after nocodazole treatment, transfer of dye to neighboring cells was greatly reduced. These contrasting effects of brefeldin A and nocodazole on the trafficking properties and intercellular dye transfer are interpreted to suggest that two pathways contribute to the routing of connexins to the gap junction.  (+info)

Vitamin B12 is very important for the human body. Early diagnosis of vitamin B12 deficiency is essential because it is associated with many problems such as fatigue, lethargy, depression, poor memory, breathlessness, headaches, pale skin, mania, psychosis, etc. Most of the vitamin B12 molecules in serum bind to specific proteins as a complex; so, although researchers have developed some sensitive methods to quantify vitamin B12, few can be used for human serum detection of vitamin B12 because of disturbed releasing process. In this study, we are aiming to develop a rapid and accurate chemiluminescence immunoassay (CLIA) for human serum testing ...

In this research, the increase in sensitivity and decrease in diagnose limit of chemiluminescence system was made possible by producing gold/silver nanoalloy as the catalyst and introducing it in combination with Amplex red. Amplex red has unique luminescence properties (its oxidized form has fluorescence - chemiluminescence ability), and it is used as the probe. The observation of fluorescence caused by Amplex red in the absence of vitamin C a few minutes after the end of luminescence reaction confirms the presence of hydrogen peroxide as the oxidant in chemiluminescence system. In fact, hydrogen peroxide and its other active forms make possible the oxidation of Amplex red and its conversion into fluorescence form. Vitamin C, having antioxidant properties, is able to decompose hydrogen peroxide and disturb chemiluminescence reaction. Therefore, the designed luminescence system was used in the measurement of vitamin C in medical products ...

Peri- and post-operative (up to day 7 after surgery) neutrophil chemiluminescence and the plasma concentrations of interleukin 6 (IL-6), IL-8, IL-10 and tumour necrosis factor alpha (TNF-alpha) were evaluated in the blood of patients undergoing liver transplantation. IL-6, IL-8 and IL-10 levels increased during early reperfusion and then returned to normal mostly within the first post-operative day. TNF-alpha was increased during the whole period observed. Spontaneous as well as activated neutrophil chemiluminescence was depressed in early reperfusion and remained low during the whole period followed. Samples collected during early reperfusion provided positive correlation for IL-6 vs IL-8 as well as for IL-6 and IL-8 vs chemiluminescence. The data were also evaluated with respect to the outcome of transplantation. Since IL-8, IL-10 and TNF-alpha levels increased significantly during the first post-operative week, mainly in a group of patients who developed serious complications within the first ...
297283835 - EP 0898620 B1 2002-11-06 - ASSAY OF PEROXIDASE ACTIVITY - [origin: WO9739142A1] Chemiluminescent detection of molecules of synthetic or natural origin such as proteins and nucleic acids (DNA and RNA), as well as other biologic molecules, is increasingly replacing radioactive detection as the method of choice where sensitivity is critical. In such assays, luminescence is customarily achieved by the oxidation of a luminol or isoluminol substrate in the presence of an oxidizing agent such as hydrogen peroxide or hydrogen peroxide source, such as perborate, and a peroxidase catalyst such as horseradish peroxidase. To obtain useful levels of luminescence (e.g., detectable levels) by customary techniques, a luminescent enhancer is also present during oxidation. It has been found in the practice of the present invention that azine enhancers have contained an impurity which reduces the properties of the chemiluminescent assay working solutions. The present invention describes a working solution, a

Minute WB Stripping Solution is designed to rapidly strip antibodies bound to antigens on Western blotting membranes using chemiluminescent substrate. One of the major advantages of chemiluminescent detection is high sensitivity and the ability to strip reagents from a blot and re-probe the same blot. Re-probing Western blots are very useful for optimizing the blotting conditions such as testing different antibodydilution and membrane blocking conditions. The key to this process is to useconditions that cause the release of antibody from the antigen without causing a significant amount of antigen to be released from the membrane. During the stripping procedure, small amount of antigen is inevitably lost from the membrane. It is important to minimize this loss by stripping the antibody under gentle conditions. Minute WB stripping solution is able to achieve maximum removal of antibodies from a membrane and preserving the integrity of the antigen in 10-15 min without using harsh conditions suchas ...

The effect of binding the Trp-free motor domain mutant of Dictyostelium discoideum, rabbit skeletal muscle myosin S1, and tropomyosin on the dynamics and conformation of actin filaments was characterized by an analysis of steady- state tryptophan phosphorescence spectra and phosphorescence decay kinetics over a temperature range of 140-293 K. The binding of the Trp-free motor domain mutant of D. discoideum to actin caused red shifts in the phosphorescence spectrum of two internal Trp residues of actin and affected the intrinsic lifetime of each emitter, decreasing by roughly twofold the short phosphorescence lifetime components (t 1 and t 2) and increasing by ;20% the longest component (t3). The alteration of actin phosphorescence by the motor protein suggests that i), structural changes occur deep down in the core of actin and that ii), subtle changes in conformation appear also on the surface but in regions distant from the motor domain binding site. When actin formed complexes with skeletal ...
Well-defined mechanistic principles have emerged to rationalize micellar enhancement of chemiluminescence. The review of Lin and Yamada[1] focuses on how micelles may be used to improve chemiluminescence signals by changes that affect the reaction rate. These occur in the microenvironment (i.e. polarity, viscosity and/or acidity, etc.), in the chemical and photophysical pathway and in the solubilization, concentration and organization of the solute/reactant. We shall now use these principles as a framework for discussing this work and it will become clear that they are highly inter-related rather than mutually exclusive.[2]. There follow examples of micellar enhancement which have been explained by changes in the microenvironment. In the interaction of sulfite groups in drugs with dissolved oxygen in presence of acidic rhodamine 6G, the surfactant Tween 60 can enhance chemiluminescence by 200%, attributable to a microenvironment that leads to an increase in the fluorescence quantum yield of ...

Sequencing by synthesis involves taking a single strand of the DNA to be sequenced and then synthesizing its complementary strand enzymatically. The Pyrosequencing method is based on detecting the activity of DNA polymerase (a DNA synthesizing enzyme) with another chemiluminescent enzyme. Essentially, the method allows sequencing of a single strand of DNA by synthesizing the complementary strand along it, one base pair at a time, and detecting which base was actually added at each step. The template DNA is immobilized, and solutions of A, C, G, and T nucleotides are added and removed after the reaction, sequentially. Light is produced only when the nucleotide solution complements the first unpaired base of the template. The sequence of solutions which produce chemiluminescent signals allows the determination of the sequence of the template ...

The wavelengths of chemiluminescence emission that are analytically useful depend on the characteristics of the detector. Visible emission (though it is seldom visible to the naked eye) has a wavelength range of about 400-750 nm, corresponding to enthalpy changes of exothermic reactions of between 180 and 300 kJ mol−1, provided that there is a pathway to an excited state that relaxes with the loss of a photon (see Figure 1.1). Emission intensity is proportional to the concentration of the emitting species, which is either an intermediate or a product in an electronically excited state. This concentration depends on the rate of the reaction producing it. Analytical detection of chemiluminescence usually involves no wavelength selection, i.e., it is emission photometry rather than emission spectrophotometry. Selectivity is achieved by on-line treatments rather than by processing of the signal, which has little fine-structure.[1]. Because of this, the importance of chemiluminescence spectroscopy ...
Bioluminescent Reporter plasmid to express bacterial luciferase (Lux) in Mycobacteria Background. Bioluminescence, the production of light by luciferase-catalysed reactions, is a versatile reporter technology with multiple applications both in vitro and in vivo. In vivo bioluminescence imaging (BLI) represents one of its most outstanding uses by allowing the non-invasive localisation of luciferase-expressing cells within an animal. Applied to the study of infectious diseases, BLI permits the detection of microorganisms from within living animals thus allowing the spatiotemporal study of infection in real-time in the same host. Moreover, using luciferase as a reporter of gene expression, it is possible to establish when and where a gene function is needed, shedding light on bacterial pathogenesis. Finally, BLI constitutes an easy and rapid method to test novel antimicrobial compounds in vivo.. Three main luciferin-luciferase systems have been utilised for BLI. The first system is represented by ...
Sensitive and feasible electrochemiluminescence (ECL) bioassay relies on the achievement of the ECL signal tags with high and stable signal intensity. In this work, strong electrochemiluminescence (ECL) emission was achieved from metal organic framework (MOFs) accelerator enriched Quantum Dots (CdTe), which was applied as an efficient ECL signal tag for trace biomarker detection. It is particularly worth noting that a novel mechanism to drastically enhance the ECL intensity of CdTe is established because the isoreticular metal organic framework-3 (IRMOF-3) with 2-amino terephthalic acid (2-NH2-BDC) as organic ligand is not only devoted for loading large amount of CdTe via encapsulating effect and internal/external decoration, but also as a novel co-reactant accelerator for promoting the conversion of co-reactant S2O82- into the sulfate radical anion (SO4•-), further boosting the electrochemiluminescence (ECL) emission of CdTe ...
CTX : Testing is performed using the Roche Cobas 6000 e601 analyzer and the Roche Beta-CrossLaps assay, a 2-site immunometric (sandwich) assay using electrochemiluminescence detection. Patient specimen, biotinylated monoclonal beta-CrossLaps-specific antibody, and monoclonal beta-CrossLaps-specific antibody labeled with ruthenium react to form a complex. Streptavidin-coated microparticles act as the solid phase to which the complex binds. Voltage is applied to the electrode, inducing a chemiluminescent emission from the ruthenium, which is then measured against a calibration curve to determine the amount of beta-CrossLaps in the patient specimen. This assay is specific for crosslinked isomerized type I collagen fragments, independent of the nature of the crosslink (eg, pyrrole, pyridinolines). The assay specificity is guaranteed through the use of 2 monoclonal antibodies, each recognizing linear beta-8AA octapeptides (EKAHD-beta-GGR). The assay, therefore, quantifies all type I collagen degradation
OBJECTIVE: Activated phagocytes, generating a variety of powerful inflammatory mediators, such as oxygen and nitrogen species, may participate in oxidative stress-mediated inflammation and organ toxicity. At present, great attention is devoted to the important class of phenolic compounds - coumarins - due to their antiinflammatory/antioxidant activities. We compared two synthetic phenylcoumarins: 7-hydroxy-3- (4-hydroxyphenyl) coumarin (HHC; 0.01-100 mu mol/l) and its hydrogenated analogue: 7-hydroxy-3-(4-hydroxyphenyl)-3,4-dihydrocoumarin (HHDC; 0.01-100 mu mol/l) as their ability to inhibit reactive oxygen species (ROS) generation in human neutrophils and nitric oxide (NO) production by RAW 264.7 macrophages in vitro, with respect to some of their physicochemical characteristics. METHODS: ROS production was measured with luminol-enhanced chemiluminescence (CL) in the microplate luminometer Immunotech LM-01T, nitrite formation was determined by the Griess reaction - spectrophotometrically. ...

The temporal effects of luciferase reaction luminescence have only been discussed in the context of light intensity (flash vs. glow). However, alterations in the color of the light emitted over the course of the luciferase reaction have not been reported. Here, we show a temporal change in the light color emitted during the reaction catalyzed by unmodified firefly luciferase when concentrations of one of the substrates, adenosine triphosphate (ATP), are gradually increased. The temporal color change from green to red occurs within the first few minutes of the luciferase reaction when an ATP-containing solution is either added or synthesized in situ with the aid of an autocatalytic reaction occurring simultaneously ...

The possibility that the mode of action of sulphasalazine and its active metabolite 5-amino-salicylic acid (5ASA) involves modification of toxic oxygen metabolite production by neutrophils has been investigated by measuring the effect of these drugs on luminol-dependent chemiluminescence, superoxide release and oxygen consumption by stimulated neutrophils in vitro. 5ASA, and to a lesser extent sulphasalazine, had profound inhibitory effects on the luminol dependent chemiluminescent response of neutrophils stimulated with formyl-methionyl-leucyl-phenylalanine (1 microM) + cytochalasin B (5 micrograms/ml). A concentration of 50 microM 5ASA or sulphasalazine produced 93.8 (2.3)% and 65.7 (3.7)% inhibition of control responses respectively. The concentration of 5ASA and sulphasalazine producing 50% inhibition of chemiluminescence were 3.6 (1.8) microM and 16.5 (6) microM respectively. Both drugs had little effect on the chemiluminescent response of neutrophils stimulated with phorbol myristate ...

A carbon fiber ionizer was used to extract ATP from bacterial cells for generating ATP bioluminescence. Our methodology was tested using Staphylococcus epidermidis and Escherichia coli, which were aerosolized with an atomizer, and then indoor bioaerosols were also used for testing the methodology. Bioaerosol concentrations were estimated without culturing which requires several days for colony formation. Correlation equations were obtained for results acquired using our methodology (Relative Luminescent Unit (RLU)/m3) and a culture-based (Colony Forming Unit (CFU)/m3) method; ...
Isolated equine alveolar macrophages were shown to generate a luminol-dependent light response when challenged with a phagocytic stimulus. The chemiluminescent response was not detected with luminol prepared at 1.0 x 10(-5) or 1.0 x 10(-4) molar concentrations, but was readily quantitated when used at a 1.0 x 10(-3) molar concentration. Challenge of the alveolar macrophages with latex particles or with equine IgG-coated sheep red blood cells elicited the luminol-dependent light response, whereas unchallenged equine alveolar macrophages or those challenged with unopsonized erythrocytes failed to emit light above background levels. Latex-bead-challenged macrophages released 8.06 times the total amount of light as those equine alveolar macrophages challenged with equine IgG-opsonized erythrocytes. This study represents the first investigation on chemiluminescence and equine alveolar macrophages. ...
The authors reported on the cervical histologic results of 209 patients demonstrating equivocal results for hrHPV from SurePath (204 patients) or ThinPrep (5 patients) vials, and compared patients in this cohort with atypical squamous cells of undetermined significance (ASC-US) cytology on the index cervical Papanicolaou (Pap) test (Group 1; n = 148 patients) with a patient cohort demonstrating unequivocal positive hrHPV test results (Group 2; n = 148 patients). The chemiluminescence intensity of hrHPV tests from patients in Group 2 were correlated with the presence and severity of dysplasia on subsequent histologic results, and patients were thereby stratified for their subsequent risk of cervical intraepithelial neoplasia (CIN) types II/III. ...

While lateral flow test formats can be utilized with whole blood and low sample volumes, their diagnostic characteristics are inferior to immunoassays based on chemiluminescence immunoassay (CLIA) or enzyme-linked immunosorbent assay (ELISA) technology. CLIAs and ELISAs can be automated to a high degree but commonly require larger serum or plasma volumes for sample processing. We addressed the suitability of EDTA-anticoagulated whole blood as an alternative sample material for antibody testing against SARS-CoV-2 by electro-CLIA (ECLIA; Roche, Rotkreuz, Switzerland) and ELISA (IgG and IgA; Euroimmun, Germany). Simultaneously drawn venous serum and EDTA-anticoagulated whole blood samples from 223 individuals were included. Correction of the whole blood results for hematocrit led to a good agreement with the serum results for weakly to moderately positive antibody signals. In receiver-operating characteristic curve analysis, all three assays displayed comparable diagnostic accuracy (area under the ...
A new flow-injection chemiluminescence method for the determination of iron is described. It is based upon the chemiluminescence reaction of iron (Ⅱ)-lucigenin system with Jones reductor. The chemiluminescence signal-to-noise ration can be improved apparently by the addition of cetyltrimethyl- ammonium bromide and emulsor OP. The present method has a wide linear of 1×10-11~1×10-5 g/mL for Fe(Ⅱ). The detection limit is 2 × 10-12 g/mL. The relative standard deviation is 2.0% (1× 10-8 g/mL Fe, n = 11). The method has been applied to the determination of iron in hair samples with satisfactory results. The mechanism of this reaction is also investigated in detail.

By using single tryptophan protein phosphorescence, we follow site-specific internal protein dynamics over a broad temperature range and demonstrate three independent dynamic processes. Process I is seen at temperatures below the bulk solvent Tg, has low activation energy, and is likely due to fast vibrations that may be enabled by water mobility on the protein surface. Process II is observed above 170 K, with activation energy typical of β relaxations in a glass; it has the same temperature ...
Figure D11.1 - On-column coaxial flow interface between capillary electrophoresis and chemiluminescence detection (adapted from reference D9.1).. As with HPLC, there is an inherent problem of compatibility between the conditions needed for separation and those needed for chemiluminescence. Additionally, there is a potential problem with the stability of chemiluminescence reagents. Both of these are addressed by using the post-column mode rather than pre-column. Post-column interfaces are devices for mixing the eluent with the chemiluminescence reagents and for this purpose designs may make use of merging flow, coaxial flow or reservoir mixing. Interfaces may also be classified as off-, on- or end-column, depending on the site of detection and on whether this is isolated from the high voltage supply used for capillary electrophoresis.. The simplest interface, off-column and merging flow, did not find widespread application. Buffer flowed from a reservoir through the separation capillary and ...
The unique cathodic electrochemiluminescence(ECL) emission of Ru(bpy)32+(bpy=2,2′-bipyridine) was observed via Nafion film at Au electrode[Au/Nafion/Ru(bpy)32+] at about 0.20 V(vs. Ag/AgCl) and applied to the determination of several amino acids without prior derivatization with high sensitivity. The cathodic electrochemiluminescence(ECL) exhibits the detection limits and linear ranges of several amino acids comparable to or better than those of capillary electrophoresis with conventional ECL detection method(at 1.10-1.20 V vs. Ag/AgCl) based on precolumn derivatization. The results suggest that the cathodic ECL is promising for the detection of amino acids in bioanalysis.. ...
Аннотация: Two methods for phenol determination based on the suppression of chemiluminescence (CL) produced upon HRP-catalysed oxidation of luminol were developed. One method used an enhanced CL reaction with 3-(100-phenothiazinyl)-propane-1-sulfonate and 4-morpholinopyridine as primary and secondary enhancers, respectively, and another one is free of the enhancers. A comparison of the phenol assay with enhancers showed much higher sensitivity and lower LDL value than the assay without enhancers. In addition, the amount of HRP used in the assay with enhancers is 40 times less than in the assay without them. The study of the specificity of the developed assays demonstrated that in the assay without enhancers several phenol compounds, for instance, unsubstituted phenol, suppressed the CL intensity, whereas other phenols, such as 4-chlorophenol and 2,4-dichlorophenol, increased the light output. In contrast to that assay, in the assay with enhancers all tested phenols suppressed the CL ...
Continuous increase in the number and the variety of anthropogenic sources of electromagnetic radiation causes a high interest in studying the effects ultrahigh frequency on living organisms. In the present research inflence of UHF EMR (15 W, 2.45 GHz) for 5 and 15 min on morphological and genetic peculiarities of Photobacterium phosphoreum colonies was studied. It has been revealed that UHF EMR affected colony growth parameters, induced transcriptional activity of luciferase encoding gene expression and that the effect was depended on exposure duration. The subsequent cultivation of bacteria during a two week period after treatment showed maintaining of the increased luxb mRNA level in irradiated colonies. Opposite bacterial stress responses were detected to UHF EMR and elevated temperature treatments that assumed UHF EMR comprised of not only thermal but specifi component of non-thermal nature.. The validity of chemiluminescence-based methods relies upon the uniqueness of the relationship between the concentration of the analyte and the intensity of chemiluminescence produced. We have examined the chemiluminescence of luminol (5-amino-2,3-dihydro-1,4-phthalazinedione) in the presence of O2, without added H2O2, to measure nanomolar concentrations of total Fe(II) (both inorganically and organically complexed) in aqueous samples. To test the validity of the method, we have developed a kinetic model that describes the two-step oxidation of luminol by superoxide and hydroxyl (or hydroxyl-like) radicals produced from the oxidation of Fe(II) by O2 by synthesis of existing published data. This model was coupled with a model for Fe(II) oxidation by O2 in the presence and absence of the naturally occurring Suwannee River fulvic acid (SRFA). The production of chemiluminescence depended upon the concentrations of free radicals in both the sample and luminol reagent and the pH at which the reactions were
Presence of anti-HCV antibodies indicates prior exposure to HCV infection. However, the assay may be negative early in acute HCV infection, in immunosuppressed individuals. Anti-HCV antibodies can be detected by rapid immunoassays, enzyme-linked immunosorbent assays (ELISA), or chemiluminescent immunoassays. Quality of some of the test kits for anti-HCV may be a reason for concern.. Rapid immunoassay tests use a unique combination of modified HCV antigens conserved across all genotypes. However, some studies have demonstrated that rapid tests cannot be solely relied upon in screening patients from high- risk population in India. ELISA tests indicate presence of antibodies against HCV core, NS3 and NS5 regions. A study in North India has demonstrated the value of blood banks screening using two ELISA kits concurrently to improve detection of contaminated blood.. Enhanced chemiluminescence immunoassay (ECi) a quantitative assay based on chemiluminescent reaction, which is as good as ELISA.. ...

The limit of detection calculated from repeated measurements of the zero calibrator was found to be ,0.6 kilounits/L in all analyses. This is equivalent to the readable limit of the analyzer. One laboratory performed measurements in a serum sample with a CA 125 value of 0.8 kilounits/L. The limit of detection obtained for serum was 1.1 kilounits/L, which is highly sufficient for diagnostic purposes.. According to the manufacturer, the assay is linear up to 5000 kilounits/L. This was checked in four laboratories in a total of 24 dilution series covering three decades of CA 125 concentrations, using either human sera or diluent for dilution. On the basis of an acceptance range of 90-110% recovery, the linearity of the assay when serum was used as diluent was confirmed throughout the range studied (highest serum concentration ∼4500 kilounits/L) with the exception of the highest dilution, which exceeded the acceptance criteria in 5 of 12 experiments. The acceptance criteria were not met with the ...

This work demonstrates an aptasensor for ultrasensitive electrochemiluminescence (ECL) detection of thrombin based on an off-on-off approach. The system is composed of an Eu3+-doped CdS nanocrystals (CdS:Eu NCs) film on glassy carbon electrode (GCE) as ECL emitter. Then gold nanoparticles (AuNPs) labeled hairpin-DNA probe (ssDNA1) containing thrombin-binding aptamer (TBA) sequence was linked on the NCs film, which led to ECL quenching (off) as a result of Förster-resonance energy transfer (FRET) between the CdS:Eu NC film and the proximal AuNPs. Upon the occurrence of hybridization with its complementary DNA (ssDNA2), an ECL enhancement (on) occurred owing to the interactions of the excited CdS:Eu NCs with ECL-induced surface plasmon resonance (SPR) in AuNPs at large separation. Thrombin could induce ssDNA1 forming a G-quadruplex and cause the AuNPs to be close to CdS:Eu NCs film again, which resulted in an enhanced ECL quenching (off). This off-on-off system showed a maximum 7.4-fold ...

The bacterial luciferase gene cassette (lux) is unique among bioluminescent bioreporter systems due to its ability to synthesize and/or scavenge all of the substrate compounds required for its production of light. As a result, the lux system has the unique ability to autonomously produce a luminescent signal, either continuously or in response to the presence of a specific trigger, across a wide array of organismal hosts. While originally employed extensively as a bacterial bioreporter system for the detection of specific chemical signals in environmental samples, the use of lux as a bioreporter technology has continuously expanded over the last 30 years to include expression in eukaryotic cells such as Saccharomyces cerevisiae and even human cell lines as well. Under these conditions, the lux system has been developed for use as a biomedical detection tool for toxicity screening and visualization of tumors in small animal models. As the technologies for lux signal detection continue to improve, it is

The Difference Between Fluorescence & Phosphorescence: What is Fluorescence? Some materials absorb invisible, high-energy black (ultraviolet) light, use part of the energy, then emit nanoseconds later bright visible light, but only while the ultraviolet light shines on it. What is Phosphorescence? Phosphorescence is similar to fluorescence. Both involve absorption of energy. Glow-in-the-dark light (phosphorescence) is not as bright as fluorescence but lasts much longer. Phosphorescence is controlled by forbidden quantum physics transitions. Quantum Physics Transitions Quantum physics transitions are changes in molecular states made possible by energy absorption. Energy changes are quantized if they can only be absorbed or emitted in predetermined values or quanta. After absorption, lower energy photons (light particles) can be re-emitted. Practical Uses Fluorescence is utilized in medicine, while phosphorescence is used for glow-in-the-dark products. Hobbyist Uses Mineralogists treasure ...

Increased oxidative stress plays a significant role in the etiology of bone diseases. Heightened levels of H2O2 disrupt bone homeostasis, leading to greater bone resorption than bone formation. Organochalcogen compounds could act as free radical trapping agents or glutathione peroxidase mimetics, reducing oxidative stress in inflammatory diseases. In this report, we synthesized and screened a library of organoselenium and organotellurium compounds for hydrogen peroxide scavenging activity, using macrophagic cell lines RAW264.7 and THP-1, as well as human mono- and poly-nuclear cells. These cells were stimulated to release H2O2, using phorbol 12-myristate 13-acetate, with and without organochalogens. Released H2O2 was then measured using a chemiluminescent assay over a period of 2 h. The screening identified an organoselenium compound which scavenged H2O2 more effectively than the vitamin E analog, Trolox. We also found that this organoselenium compound protected MC3T3 cells against H2O2 -induced ...
We also decided that we needed to increase the concentration of the HRP in the cuvet. The Fluorimeter was not working and so we just used visual analysis to see if any fluorescence was occurring. The first concentration in the cuvet we tested was 2mM- which did not produce any luminescence. The next concentration we attempted was 10mM which also did not result in any luminescence. We then decided to double this final concentration to 20mM HRP while maintaining 3mM concentration of Luminol. This sample resulted in fluorescence of the luminol in the form of a blue light! We plan on running different variations of the HRP concentration beginning with 20mM tomorrow when conducting the chemiluminescence assay. All other reagents will remain constant ...
