Combination interferon-alpha2a and 13-cis-retinoic acid enhances radiosensitization of human malignant glioma cells in vitro. (1/311)

We investigated the individual and combined effects of cis-retinoic acid (CRA) and/or IFN-alpha (IFN) and/or radiation therapy (RT) against a human glioma cell line (American Type Culture Collection; U373MG) to evaluate the possible radiosensitization properties of these agents in vitro. Glioma cells were incubated for 24 h in 96-well plates (2 x 10(2) cells/well) in standard culture medium. Sets of U373 (n = 12) were exposed to CRA (3 x 10(6) microM), IFN (25 units/ml), CRA plus IFN, or standard culture medium. After an additional 24 h of incubation, the U373 cells were subjected to increasing radiation doses (up to 16 Gy). Glioma cells were harvested 92 h after irradiation, and cell survival curves were determined from [3H]thymidine incorporation data (over the last 24 h). The experiment was repeated for both the untreated control group and the combined CRA/IFN group. To verify the [3H]thymidine assays, a clonogenic assay was also performed. Single cell suspensions of U373 cells were plated out in six-well plates (n = 3). After chemical and RT treatment, colonies of 50 cells or more were counted, and cell survival curves were generated as fractions of nonirradiated controls. The amount of RT (in Gy) that would cause a 50% survival fraction (lethal dose 50 or LD50) was calculated from the survival curves by regression analysis. The following LD50s were obtained: [table: see text] The results showed that for both the [3H]thymidine incorporation assay and the clonogenic assay, the combination of IFN/CRA rendered U373 cells more susceptible to ionizing radiation than the untreated control or either single agent alone.  (+info)

Young women taking isotretinoin still conceive. Role of physicians in preventing disaster. (2/311)

QUESTION: One of my adolescent patients was prescribed isotretinoin for severe acne by a dermatologist. I was shocked to discover she does not use any means of contraception. The dermatologist insists he told her about the need for contraception. How can we do better? ANSWER: Clearly this dermatologist, like many of his colleagues, does not comply with the Pregnancy Prevention Program. Until physicians become more aware of this program, babies will continue to be born with embryopathy due to isotretinoin.  (+info)

13-cis-Retinoic acid alters neural crest cells expressing Krox-20 and Pax-2 in macaque embryos. (3/311)

This study investigates hindbrain and associated neural crest (NCC), otocyst, and pharyngeal arch development in monkey embryos following teratogenic exposure to 13-cis-retinoic acid (cRA). cRA was orally administered (5 mg/kg) to pregnant long-tailed macaques (Macaca fascicularis) between gestational days (GD) 12 and 27. Embryos were surgically collected at desired stages during treatment, analyzed for external morphological changes, and processed for immunohistochemistry. Two transiently expressed nuclear proteins, Krox-20 and Pax-2, were used as markers for the target cellular and anatomical structures. Rhombomere (r) expression patterns of Pax-2 (r4/r6) and Krox-20 (r3/r5) were maintained after cRA treatment, but r4 and r5 were substantially reduced in size. In untreated embryos, Krox-20 immunoreactive NCC derived from r5 migrated caudally around the developing otocyst to contribute to the third pharyngeal arch mesenchyme. In cRA-treated embryos, a subpopulation of NCC rostral to the otocyst also showed Krox-20 immunoreactivity, but there was a substantial reduction in Krox-20 post-otic NCC. Pax-2 immunoreactive NCC migrating from r4 to the second pharyngeal arch were substantially reduced in numbers in treated embryos. Alteration in the otic anlage included delayed invagination, abnormal relationship with the adjacent hindbrain epithelium, and altered expression boundaries for Pax-2. cRA-associated changes in the pharyngeal arch region due to cRA included truncation of the distal portion of the first arch and reduction in the size of the second arch. These alterations in hindbrain, neural crest, otic anlage, and pharyngeal arch morphogenesis could contribute to some of the craniofacial malformations in the macaque fetus associated with exposure to cRA.  (+info)

Induction of matrix metalloprotease-1 gene expression by retinoic acid in the human pancreatic tumour cell line Dan-G. (4/311)

We have investigated the effects of retinoic acid (RA) on matrix metalloprotease-1 (MMP-1) gene expression in the human pancreatic tumour cell line Dan-G. 13-cis RA results in a time- and dose-dependent increase of MMP-1 protein concentration. These stimulatory effects were paralleled by a time- and dose-dependent increase of MMP-1 mRNA steady-state concentrations. Nuclear run-on analysis revealed that the increase of MMP-1 mRNA was partially due to an increase of MMP-1 gene transcription. In addition, 13-cis RA treatment results in an increase of MMP-1 mRNA stability. These data demonstrate that RA stimulates MMP-1 gene expression in human pancreatic carcinoma cells by transcriptional and post-transcriptional mechanisms.  (+info)

Regulation of AML2/CBFA3 in hematopoietic cells through the retinoic acid receptor alpha-dependent signaling pathway. (5/311)

AML2 is a member of the acute myelogenous leukemia, AML family of transcription factors. The biologic functions of AML1 and AML3 have been well characterized; however, the functional role of AML2 remains unknown. In this study, we found that AML2 protein expressed predominantly in cells of hematopoietic origin is a nuclear serine phosphoprotein associated with the nuclear matrix, and its expression is not cell cycle-related. In HL-60 cells AML2 expression can be induced by all three natural retinoids, all-trans-retinoic acid (RA), 13-cis-RA, and 9-cis-RA in a dose-dependent manner. A synthetic retinoic acid derivative, 4HPR, which neither activates RA receptor (RAR) alpha nor retinoic X receptor alpha was unable to induce the expression of AML2. A RAR-selective activator, TTNPB, induced AML2 expression similar to RA. Our study further showed that AGN193109, a potent RARalpha antagonist, suppressed AML2 expression induced by RA and that a retinoic X receptor pan agonist AGN194204 had no effect on its expression. Taken together, these studies conclusively demonstrated that the expression of AML2 in HL-60 cells is regulated through the RARalpha-specific signaling pathway. Our study further showed that after all-trans-retinoic acid priming, AML2 expression could be augmented by vitamin D(3). Based on these studies we hypothesize that AML2 expression is normally regulated by retinoid/vitamin D nuclear receptors mainly through the RARalpha-dependent signaling pathway and that it may play a role in hematopoietic cell differentiation.  (+info)

Up-regulation of retinoic acid receptor beta expression in renal cancers in vivo correlates with response to 13-cis-retinoic acid and interferon-alpha-2a. (6/311)

Retinoic acid receptor-beta (RAR-beta) mRNA is not expressed by retinoid-resistant renal cancer cell lines but is present in retinoid-sensitive SK-RC-06 renal cancer cells and increases following incubation with retinoic acid (RA), suggesting that the antitumor action of RA is mediated through RAR-beta (A. D. Hoffman et al., Clin. Cancer Res., 2: 1077-1082, 2996). To determine whether RAR-beta expression correlates in vivo with major clinical response to patients with renal cell carcinoma (RCC) who were treated with retinoid-based therapy, we used in situ hybridization to analyze RAR-beta expression in tumor specimens obtained from patients who were treated on a clinical trial with 13-cis-RA and IFN-alpha. Thirty-three tissue specimens were analyzed (23 pretreatment and 10 on-treatment). mRNA expression was based on staining intensity, with scores within tumor cells ranging from 0 to 2, where a score of 0 indicated absence of staining, a score of 1 indicated weak staining, and a score of 2 indicated strong staining. RAR-beta expression was present in 22 of 23 (96%) pretreatment and 9 of 10 (90%) on-treatment specimens. Pretreatment levels of expression did not associate with the site of biopsy and did not predict for major clinical response to RA plus IFN-alpha therapy (two-sided Fisher's exact test, P = 0.826). However, an increase in the intensity of RAR-beta mRNA expression was detected in four of five (80%) patients who achieved a major response but in none of the five patients with progressive disease in whom sequential biopsies were available (two-sided Fisher's exact test, P = 0.048). These data show that RAR-beta transcripts increase in tumor cells of RCC patients who clinically respond to retinoid-based therapy. Retinoids that potently induce RAR-beta expression should be evaluated in the treatment of advanced RCC.  (+info)

Retinoic acids inhibit inducible nitric oxide synthase expression in mesangial cells. (7/311)

BACKGROUND: Nitric oxide (NO) release as a result of cytokine-mediated activation of inducible nitric oxide synthase (iNOS) in mesangial cells can be sustained and lead to oxidative injury in various forms of glomerular inflammation. Inhibition of iNOS expression and/or activity could therefore be an effective anti-inflammatory strategy. The present study was undertaken to explore whether retinoids, which are known to have anti-inflammatory and immuno-modulatory actions, can attenuate cytokine-stimulated iNOS expression and enzyme activity in murine mesangial cells. METHODS: Expression of iNOS was evaluated by NO production (nitrite analysis), protein (Western blot analysis) and mRNA (RT-PCR analysis) levels in mesangial cells stimulated by a combination of lipopolysaccharide (LPS) and interferon-gamma (IFN-gamma) in the presence and absence of all-trans-retinoic acid (ATRA) or its active metabolite, 13-cis-retinoic acid (13-cis-RA). Changes in iNOS enzyme activity were assessed by calculating conversion of L-[14C]arginine to L-[14C]citrulline. The levels of transcription factors nuclear factor-kappaB (NF-kappaB) and activated protein-1 (AP-1) in nuclear extracts prepared from mesangial cells stimulated by a combination of LPS and IFN-gamma in the presence and absence of ATRA was assessed by immunoblot analysis. The effect of both retinoids on transforming growth factor-beta1 (TGF-beta1) levels was also assessed by a quantitative enzyme immunoassay method. RESULTS: The combination of LPS/IFN-gamma stimulated NO production, induced iNOS expression (mRNA and protein) and increased iNOS enzyme activity. ATRA and 13-cis-RA dose-dependently attenuated NO production. This effect was most pronounced at ATRA concentration of 10 microM. At this concentration, ATRA attenuated iNOS expression (mRNA and protein levels) and enzyme activity. ATRA also reduced nuclear levels of both subunits (p50 and p65) of NF-kappaB. TGF-beta1 levels in mesangial cells stimulated with LPS/IFN-gamma in presence of ATRA or 13-cis-RA were also reduced indicating that TGF-beta1 did not mediate the suppressive effect of retinoids on iNOS. CONCLUSIONS: Our studies demonstrate that the retinoids ATRA and 13-cis-RA attenuate iNOS expression and activity in cytokine-stimulated murine mesangial cells. These retinoids may emerge as naturally occurring compounds for treatment of inflammatory glomerular diseases.  (+info)

Nuclear retinoid acid receptor beta in bronchial epithelium of smokers before and during chemoprevention. (8/311)

BACKGROUND: Retinoids can reverse neoplastic lesions and prevent second primary tumors in the aerodigestive tract. These effects are thought to be mediated by nuclear retinoic acid receptors (RARs) and retinoid X receptors (RXRs), each receptor group including three subtypes (alpha, beta, and gamma). Previously, we found that RARbeta expression was suppressed in lung cancer. In this study, we investigated whether expression of RARbeta is modulated by chemopreventive intervention. METHODS: Using in situ hybridization, we analyzed RARbeta messenger RNA (mRNA) expression in bronchial biopsy specimens from heavy smokers, at baseline and after 6 months of treatment with 13-cis-retinoic acid (13-cis-RA) or placebo. Since we had previously detected RARbeta expression in 90% of bronchial specimens from nonsmokers, we considered loss of RARbeta mRNA expression in at least one of six biopsy specimens at baseline in this study to be aberrant. RESULTS: RARbeta mRNA expression was aberrant in 30 (85.7%) of 35 subjects in the 13-cis-RA group and in 24 (72.7%) of 33 subjects in the placebo group. After 6 months of 13-cis-RA treatment, the number of subjects who were RARbeta positive in all six biopsy specimens increased from five of 35 to 13 of 35 (2.6-fold), so that the percentage of individuals with aberrant RARbeta expression decreased to 62.9% (22 of 35), which represents a statistically significant difference from baseline expression (two-sided P =.01). In the placebo group, no statistically significant difference in RARbeta expression was observed between baseline and 6 months. RARbeta expression was not related to current smoking status or reversal of squamous metaplasia. CONCLUSIONS: These results indicate that RARbeta is an independent marker of response to 13-cis-RA and may serve as an intermediate biomarker in chemoprevention trials of upper aerodigestive tract cancers.  (+info)

Isotretinoin is a derivative of vitamin A, used in the treatment of severe recalcitrant nodular acne that has not responded to other therapies. It is a potent inhibitor of sebaceous gland function and keratinization. Isotretinoin is also known to have anti-inflammatory properties. It is taken orally in the form of capsules and its use requires careful monitoring due to potential teratogenic effects and other side effects, such as dryness of the skin and mucous membranes, mood changes, and liver enzyme abnormalities.

Acne vulgaris is a common skin condition characterized by the formation of various types of blemishes on the skin, such as blackheads, whiteheads, papules, pustules, and cysts or nodules. These lesions typically appear on areas of the body that have a high concentration of sebaceous glands, including the face, neck, chest, back, and shoulders.

Acne vulgaris occurs when hair follicles become clogged with dead skin cells and excess oil (sebum) produced by the sebaceous glands. This blockage provides an ideal environment for bacteria, particularly Propionibacterium acnes, to multiply, leading to inflammation and infection. The severity of acne vulgaris can range from mild with only a few scattered comedones (blackheads or whiteheads) to severe cystic acne, which can cause significant scarring and emotional distress.

The exact causes of acne vulgaris are not fully understood, but several factors contribute to its development, including:

1. Hormonal changes during puberty, menstruation, pregnancy, or due to conditions like polycystic ovary syndrome (PCOS)
2. Genetic predisposition
3. Use of certain medications, such as corticosteroids and lithium
4. Excessive production of sebum due to overactive sebaceous glands
5. Accumulation of dead skin cells that clog pores
6. Bacterial infection (particularly Propionibacterium acnes)
7. Inflammation caused by the body's immune response to bacterial infection and clogged pores

Treatment for acne vulgaris depends on its severity and can include over-the-counter or prescription topical treatments, oral medications, chemical peels, light therapies, or even hormonal therapies in some cases. It is essential to seek professional medical advice from a dermatologist or healthcare provider to determine the most appropriate treatment plan for individual needs.

Dermatologic agents are medications, chemicals, or other substances that are applied to the skin (dermis) for therapeutic or cosmetic purposes. They can be used to treat various skin conditions such as acne, eczema, psoriasis, fungal infections, and wounds. Dermatologic agents include topical corticosteroids, antibiotics, antifungals, retinoids, benzoyl peroxide, salicylic acid, and many others. They can come in various forms such as creams, ointments, gels, lotions, solutions, and patches. It is important to follow the instructions for use carefully to ensure safety and effectiveness.

Keratolytic agents are substances that cause the softening and sloughing off of excess keratin, the protein that makes up the outermost layer of the skin (stratum corneum). These agents help to break down and remove dead skin cells, increase moisture retention, and promote the growth of new skin cells. They are commonly used in the treatment of various dermatological conditions such as psoriasis, eczema, warts, calluses, and ichthyosis. Examples of keratolytic agents include salicylic acid, urea, lactic acid, and retinoic acid.

Sebum is an oily, waxy substance that is produced by the sebaceous glands in the skin of mammals. It is composed mainly of triglycerides, wax esters, squalene, and free fatty acids, as well as smaller amounts of metabolites and other substances. Sebum plays an important role in the maintenance of the skin's barrier function and in the regulation of its moisture levels. It also has antimicrobial properties that help to protect the skin from infection. Excessive sebum production can contribute to the development of acne and other skin conditions.

Facial dermatoses refer to various skin conditions that affect the face. These can include a wide range of disorders, such as:

1. Acne vulgaris: A common skin condition characterized by the formation of comedones (blackheads and whiteheads) and inflammatory papules, pustules, and nodules. It primarily affects the face, neck, chest, and back.
2. Rosacea: A chronic skin condition that causes redness, flushing, and visible blood vessels on the face, along with bumps or pimples and sometimes eye irritation.
3. Seborrheic dermatitis: A common inflammatory skin disorder that causes a red, itchy, and flaky rash, often on the scalp, face, and eyebrows. It can also affect other oily areas of the body, like the sides of the nose and behind the ears.
4. Atopic dermatitis (eczema): A chronic inflammatory skin condition that causes red, itchy, and scaly patches on the skin. While it can occur anywhere on the body, it frequently affects the face, especially in infants and young children.
5. Psoriasis: An autoimmune disorder that results in thick, scaly, silvery, or red patches on the skin. It can affect any part of the body, including the face.
6. Contact dermatitis: A skin reaction caused by direct contact with an allergen or irritant, resulting in redness, itching, and inflammation. The face can be affected when allergens or irritants come into contact with the skin through cosmetics, skincare products, or other substances.
7. Lupus erythematosus: An autoimmune disorder that can cause a butterfly-shaped rash on the cheeks and nose, along with other symptoms like joint pain, fatigue, and photosensitivity.
8. Perioral dermatitis: A inflammatory skin condition that causes redness, small bumps, and dryness around the mouth, often mistaken for acne. It can also affect the skin around the nose and eyes.
9. Vitiligo: An autoimmune disorder that results in the loss of pigmentation in patches of skin, which can occur on the face and other parts of the body.
10. Tinea faciei: A fungal infection that affects the facial skin, causing red, scaly, or itchy patches. It is also known as ringworm of the face.

These are just a few examples of skin conditions that can affect the face. If you experience any unusual symptoms or changes in your skin, it's essential to consult a dermatologist for proper diagnosis and treatment.

Acitretin is a synthetic form of retinoic acid, which is a type of vitamin A. It is used to treat severe psoriasis and other skin conditions. Acitretin works by slowing down the rapid growth of skin cells that cause the symptoms of psoriasis. It comes in the form of a capsule and is taken orally.

Common side effects of acitretin include dryness of the skin, lips, and mouth, itching, peeling, redness, or stickiness of the palms and soles, hair loss, and changes in nail growth. Less common but more serious side effects can include liver damage, increased levels of lipids in the blood, and birth defects if taken during pregnancy.

It is important to note that acitretin can cause birth defects, so women who are pregnant or planning to become pregnant should not take this medication. Additionally, because acitretin can remain in the body for a long time, it is recommended that women of childbearing age use effective contraception while taking this medication and for at least three years after stopping it.

Rosacea is a chronic skin condition primarily characterized by persistent redness, inflammation, and visible blood vessels on the face, particularly the nose, cheeks, forehead, and chin. It can also cause small, red, pus-filled bumps. Rosacea typically affects adults between 30 and 50 years old, with fair skin types being more susceptible. The exact cause of rosacea is unknown, but it's believed to be a combination of genetic and environmental factors, including abnormal facial blood vessels, immune system issues, and certain triggers (such as sun exposure, emotional stress, hot or cold weather, heavy exercise, alcohol consumption, spicy foods, and certain skin care products). There is no cure for rosacea, but various treatments can help control its symptoms and improve the appearance of the skin. These may include topical medications, oral antibiotics, laser therapy, and lifestyle modifications to avoid triggers.

"Drug-induced abnormalities" refer to physical or physiological changes that occur as a result of taking medication or drugs. These abnormalities can affect various organs and systems in the body and can range from minor symptoms, such as nausea or dizziness, to more serious conditions, such as liver damage or heart rhythm disturbances.

Drug-induced abnormalities can occur for several reasons, including:

1. Direct toxicity: Some drugs can directly damage cells and tissues in the body, leading to abnormalities.
2. Altered metabolism: Drugs can interfere with normal metabolic processes in the body, leading to the accumulation of harmful substances or the depletion of essential nutrients.
3. Hormonal imbalances: Some drugs can affect hormone levels in the body, leading to abnormalities.
4. Allergic reactions: Some people may have allergic reactions to certain drugs, which can cause a range of symptoms, including rashes, swelling, and difficulty breathing.
5. Interactions with other drugs: Taking multiple medications or drugs at the same time can increase the risk of drug-induced abnormalities.

It is important for healthcare providers to monitor patients closely for signs of drug-induced abnormalities and to adjust medication dosages or switch to alternative treatments as necessary. Patients should also inform their healthcare providers of any symptoms they experience while taking medication, as these may be related to drug-induced abnormalities.

Sebaceous glands are microscopic, exocrine glands that are found in the dermis of mammalian skin. They are attached to hair follicles and produce an oily substance called sebum, which is composed of triglycerides, wax esters, squalene, and metabolites of fat-producing cells (fatty acids, cholesterol). Sebum is released through a duct onto the surface of the skin, where it forms a protective barrier that helps to prevent water loss, keeps the skin and hair moisturized, and has antibacterial properties.

Sebaceous glands are distributed throughout the body, but they are most numerous on the face, scalp, and upper trunk. They can also be found in other areas of the body such as the eyelids (where they are known as meibomian glands), the external ear canal, and the genital area.

Abnormalities in sebaceous gland function can lead to various skin conditions, including acne, seborrheic dermatitis, and certain types of skin cancer.

Folliculitis is a medical condition characterized by inflammation of one or more hair follicles, typically appearing as small red bumps or pimples that surround the affected follicle. It can occur anywhere on the body where hair grows, but it's most common in areas exposed to friction, heat, and tight clothing such as the neck, back, legs, arms, and buttocks.

Folliculitis can be caused by various factors, including bacterial or fungal infections, irritation from shaving or waxing, ingrown hairs, and exposure to chemicals or sweat. The severity of folliculitis ranges from mild cases that resolve on their own within a few days to severe cases that may require medical treatment.

Treatment for folliculitis depends on the underlying cause. For bacterial infections, antibiotics may be prescribed, while antifungal medications are used for fungal infections. In some cases, topical treatments such as creams or gels may be sufficient to treat mild folliculitis, while more severe cases may require oral medication or other medical interventions.

Topical administration refers to a route of administering a medication or treatment directly to a specific area of the body, such as the skin, mucous membranes, or eyes. This method allows the drug to be applied directly to the site where it is needed, which can increase its effectiveness and reduce potential side effects compared to systemic administration (taking the medication by mouth or injecting it into a vein or muscle).

Topical medications come in various forms, including creams, ointments, gels, lotions, solutions, sprays, and patches. They may be used to treat localized conditions such as skin infections, rashes, inflammation, or pain, or to deliver medication to the eyes or mucous membranes for local or systemic effects.

When applying topical medications, it is important to follow the instructions carefully to ensure proper absorption and avoid irritation or other adverse reactions. This may include cleaning the area before application, covering the treated area with a dressing, or avoiding exposure to sunlight or water after application, depending on the specific medication and its intended use.

Hyperostosis is a medical term that refers to an excessive growth or abnormal thickening of bone tissue. It can occur as a result of various conditions, such as inflammation, injury, or genetic disorders. The extra bone growth can cause pain, stiffness, and limited mobility in the affected area. In some cases, hyperostosis can also lead to deformities and other complications.

There are several types of hyperostosis, including:

1. Diffuse idiopathic skeletal hyperostosis (DISH): This is a condition that affects the spine, causing calcification and stiffening of the ligaments and bone spurs to form along the edges of the vertebrae. It is often asymptomatic but can cause pain and stiffness in some cases.
2. Flat bone hyperostosis: This type of hyperostosis affects the flat bones of the body, such as the skull, ribs, and pelvis. It can be caused by various conditions, including Paget's disease, fibrous dysplasia, and certain types of cancer.
3. Focal hyperostosis: This refers to localized areas of bone overgrowth that can occur in response to injury, infection, or inflammation. Examples include heterotopic ossification (the formation of bone in soft tissues) and Freiberg's infarction (a condition that affects the joint surface of the metatarsal bones in the foot).
4. Hyperostosis frontalis interna: This is a benign condition that causes thickening of the inner table of the frontal bone in the skull. It is more common in women and often asymptomatic but can cause headaches and other symptoms in some cases.

Treatment for hyperostosis depends on the underlying cause and severity of the condition. In some cases, no treatment may be necessary. However, if the condition causes pain or limits mobility, various treatments may be recommended, such as medication, physical therapy, or surgery.

Leukoplakia is a medical term used to describe a white or gray patch that develops on the mucous membranes lining the inside of the mouth. These patches are typically caused by excessive cell growth and cannot be easily scraped off. Leukoplakia is often associated with long-term tobacco use, including smoking and chewing tobacco, as well as alcohol consumption. While most cases of leukoplakia are benign, a small percentage can develop into oral cancer, so it's essential to have any suspicious patches evaluated by a healthcare professional.

Retinoids are a class of chemical compounds that are derivatives of vitamin A. They are widely used in dermatology for the treatment of various skin conditions, including acne, psoriasis, and photoaging. Retinoids can help to reduce inflammation, improve skin texture and tone, and stimulate collagen production.

Retinoids work by binding to specific receptors in the skin cells, which triggers a series of biochemical reactions that regulate gene expression and promote cell differentiation and turnover. This can help to unclog pores, reduce the appearance of fine lines and wrinkles, and improve the overall health and appearance of the skin.

There are several different types of retinoids used in skincare products, including retinoic acid, retinaldehyde, and retinol. Retinoic acid is the most potent form of retinoid and is available by prescription only. Retinaldehyde and retinol are weaker forms of retinoid that can be found in over-the-counter skincare products.

While retinoids can be highly effective for treating various skin conditions, they can also cause side effects such as dryness, irritation, and sensitivity to the sun. It is important to use retinoids as directed by a healthcare professional and to follow proper sun protection measures when using these products.

Oral administration is a route of giving medications or other substances by mouth. This can be in the form of tablets, capsules, liquids, pastes, or other forms that can be swallowed. Once ingested, the substance is absorbed through the gastrointestinal tract and enters the bloodstream to reach its intended target site in the body. Oral administration is a common and convenient route of medication delivery, but it may not be appropriate for all substances or in certain situations, such as when rapid onset of action is required or when the patient has difficulty swallowing.

Propionibacterium acnes is a gram-positive, rod-shaped bacterium that naturally colonizes the skin, predominantly in areas with a high density of sebaceous glands such as the face, back, and chest. It is part of the normal skin flora but can contribute to the development of acne vulgaris when it proliferates excessively and clogs the pilosebaceous units (hair follicles).

The bacterium metabolizes sebum, producing propionic acid and other short-chain fatty acids as byproducts. In acne, these byproducts can cause an inflammatory response in the skin, leading to the formation of papules, pustules, and nodules. Propionibacterium acnes has also been implicated in various other skin conditions and occasionally in opportunistic infections in other parts of the body, particularly in immunocompromised individuals or following surgical procedures.

Benzoyl peroxide is a medication used in the treatment of acne. It is available in various forms, including creams, gels, and washes. Benzoyl peroxide works by reducing the amount of bacteria on the skin and helping to unclog pores. It is typically applied to the affected area once or twice a day.

Benzoyl peroxide can cause side effects such as dryness, redness, and irritation of the skin. It is important to follow the directions for use carefully and start with a lower concentration if you are new to using this medication. If you experience severe or persistent side effects, it is recommended that you speak with a healthcare provider.

It is also important to note that benzoyl peroxide can bleach clothing and hair, so it is best to apply it carefully and allow it to fully absorb into the skin before dressing or coming into contact with fabrics.

Leukoplakia, oral is a predominantly white patch or plaque that cannot be characterized clinically or pathologically as any other disease. It is an oral potentially malignant disorder (OPMD) and represents a significant risk for the development of squamous cell carcinoma. The lesions are typically caused by chronic irritation, such as smoking or smokeless tobacco use, and are most commonly found on the tongue, floor of the mouth, and buccal mucosa. The diagnosis is confirmed through a biopsy, and management includes removal of causative factors and close monitoring for any signs of malignant transformation.
