Mercury toxicity due to the smelting of placer gold recovered by mercury amalgam. (1/2249)

A 19-year-old man developed tremor in both hands and fatigue after starting work at a placer gold mine where he was exposed to mercury-gold amalgam. Examination revealed an intention tremor, dysdiadochokinesis and mild rigidity. The 24-h urinary mercury concentration reached a peak of 715 nmol/l (143 ug/l) shortly before the clinical examination, after which he was removed from working in the gold room [Mercury No. Adverse Effect Level: 250 nmol/l (50 ug/l)]. On review 7 weeks later his tremor had almost resolved and the dysdiadochokinesis and rigidity had gone. The 24-h urinary mercury concentration had fallen to 160 nmol/l (32 ug/l). The principal exposure to mercury was considered to be the smelting of retorted gold with previously unrecognized residual mercury in it. The peak air concentration of mercury vapour during gold smelting was 0.533 mg/m3 (Mercury Vapour ACGIH TLV: 0.05 mg/m3 TWA). Several engineering and procedural controls were instituted. Carbon disulphide absorption during xanthate reagent mixing in a gold mine concentrator. (2/2249)

A xanthate reagent mixer at a gold mine concentrator was exposed to carbon disulphide by extensive skin contamination with xanthate powder and solution during the reagent mixing process. Absorption of carbon disulphide was confirmed by the detection of urinary 2-thiothiazolidine-4-carboxylic acid (TTCA). Drager colorimetric tube testing during subsequent mixing recorded a maximum concentration of at least 60 ppm carbon disulphide. An illness consisting of predominantly gastrointestinal symptoms began 20 h after the exposure. Although this may have been due to carbon disulphide toxicity this is by no means certain. The need for engineering controls, impervious protective clothing and full-face respirators with particulate and organic vapour cartridges is discussed. Particle bombardment mediated transformation and GFP expression in the moss Physcomitrella patens. (3/2249)

There are few plants facilitated for the study of development, morphogenesis and gene expression at the cellular level. The moss Physcomitrella patens can be a very useful plant with several advantages: simple life cycle containing a major haploid gametophyte stage, easy manipulation, small genome size (6 x 10(8) bp) and high similarities with higher plants. To establish the transformation system of mosses as a model for basic plant research, a series of experiments were performed. Mosses were cultured in cellophane overlaid BCD media, transformed by particle bombardment and selected by the choice of appropriate antibiotics. Initial transformants appeared 8 d or 14 d after selection, showing different sensitivities toward the antibiotics used. Heat treatment during the preparation of particles revealed that denaturing the DNA enabled a more efficient way to deliver a transgene into the chromosome. This was proven by the increase in the number of transformants by five times in the plants with denatured DNA. In the test for the repairing capacity of mosses, 154 and 195 transformants survived from 1 d and 3 d incubations, respectively, indicating that a longer period of incubation seemed to be recommendable for better survival. The selected transformants were further analyzed at the DNA and expression level. Transformed genes were confirmed by PCR where all the transformants showed the expected size of amplification. Histochemical beta-glucuronidase (GUS) and green fluorescent protein (GFP) expression also confirmed the integration of exogenous DNA. In a comparison of the two different forms of GFP, soluble-modified GFP (smGFP) expressed stronger signals than modified GFP (mGFP) due to its improved solubility. Confirmation of the transgene in the chloroplast transformation has improved the applicability of moss as a model system for the study of basic biological researches.  (+info)

Cutting edge: receptor-mediated endocytosis of heat shock proteins by professional antigen-presenting cells. (4/2249)

Immunization with heat shock proteins (HSPs) induces Ag-specific CTL responses. The specificity of the immune response is based on peptides associated with HSPs. To investigate how exogenous HSP/peptide complexes gain access to the MHC class I-restricted Ag presentation pathway, we incubated the monocytic cell line P388D1 and the dendritic cell line D2SC/1 with gold-labeled HSPs gp96 and HSC70. We show that HSPs bind specifically to the surface of these APCs and are internalized spontaneously by receptor-mediated endocytosis, demonstrating the existence of specific receptors for HSPs on these cells. In addition, we observe colocalization of internalized HSPs and surface MHC class I molecules in early and late endosomal structures. These findings provide possible explanations for the immunogenicity of HSP/peptide complexes and for the transfer of HSP-associated peptides onto MHC class I molecules.  (+info)

The secretory route of the leaderless protein interleukin 1beta involves exocytosis of endolysosome-related vesicles. (5/2249)

Interleukin 1beta (IL-1beta), a secretory protein lacking a signal peptide, does not follow the classical endoplasmic reticulum-to-Golgi pathway of secretion. Here we provide the evidence for a "leaderless" secretory route that uses regulated exocytosis of preterminal endocytic vesicles to transport cytosolic IL-1beta out of the cell. Indeed, although most of the IL-1beta precursor (proIL-1beta) localizes in the cytosol of activated human monocytes, a fraction is contained within vesicles that cofractionate with late endosomes and early lysosomes on Percoll density gradients and display ultrastructural features and markers typical of these organelles. The observation of organelles positive for both IL-1beta and the endolysosomal hydrolase cathepsin D or for both IL-1beta and the lysosomal marker Lamp-1 further suggests that they belong to the preterminal endocytic compartment. In addition, similarly to lysosomal hydrolases, secretion of IL-1beta is induced by acidotropic drugs. Treatment of monocytes with the sulfonylurea glibenclamide inhibits both IL-1beta secretion and vesicular accumulation, suggesting that this drug prevents the translocation of proIL-1beta from the cytosol into the vesicles. A high concentration of extracellular ATP and hypotonic medium increase secretion of IL-1beta but deplete the vesicular proIL-1beta content, indicating that exocytosis of proIL-1beta-containing vesicles is regulated by ATP and osmotic conditions.  (+info)

Leptospirosis and Ebola virus infection in five gold-panning villages in northeastern Gabon. (6/2249)

An exhaustive epidemiologic and serologic survey was carried out in five gold-panning villages situated in northeastern Gabon to estimate the degree of exposure of to leptospirosis and Ebola virus. The seroprevalence was 15.7% for leptospirosis and 10.2% for Ebola virus. Sixty years after the last seroepidemiologic survey of leptospirosis in Gabon, this study demonstrates the persistence of this infection among the endemic population and the need to consider it as a potential cause of hemorrhagic fever in Gabon. There was no significant statistical correlation between the serologic status of populations exposed to both infectious agents, indicating the lack of common risk factors for these diseases.  (+info)

Cellular responses to Plasmodium falciparum major surface antigens and their relationship to human activities associated with malaria transmission. (7/2249)

In Brazil, two types of activities have led to the worsening of malarial transmission in the Amazon region: prospecting/mining and agricultural settlements. In the present study, we analyze the cellular response of 52 of these individuals (14 gold-miners and 38 farmers) living within the same endemic area. Two Plasmodium falciparum major surface antigens (recombinant proteins) were used for cellular proliferative assays: circumsporozoite protein and merozoite surface protein-1. The frequency of these cellular responses were significantly higher among the miners (57-64%) than the farmers (10-20%) when either recombinant protein was used. Our data suggest that a higher exposure to malaria of the gold-miners contributed to their higher in vitro cellular response compared with the farmers. These findings point the way to further studies evaluating the influence of risk factors associated with the life styles of different social groups and the immune responses to these antigens.  (+info)

Tuning and controlling size, shape and surface characteristics of plasmonic gold nanoparticles can help define their photonic, catalytic, and spectroscopic properties, and therefore are crucial for the development of biosensors. In the present study, we fabricated a hemoglobin/gold nanoparticle (Hb/GNP) heterolayer immobilized on the Au micro-gap to confirm H2O2 detection with a signal-enhancement effect. The hemoglobin which contained the heme group catalyzed the reduction of H2O2. To facilitate the electron transfer between hemoglobin and Au micro-gap electrode, a gold nanoparticle was introduced. We present and analyze a method to improve the morphology and mechanical properties of gold thin films for use in optical sensors or other settings where good adhesion of gold to a substrate is of importance and where controlled topography/roughness is key. To improve the adhesion of thermally evaporated gold thin films, we introduce a gold deposition step on SU-8 photoresist prior to UV exposure but after the pre-bake step of SU-8 processing. Shrinkage and distribution of residual stresses, which occur during cross-linking of the SU-8 polymer layer in the post-exposure baking step, are responsible for the higher adhesion of the top gold film to the post-deposition cured SU-8 sublayer. The SU-8 underlayer can also be used to tune the resulting gold film morphology. Our promoter-free protocol is easily integrated with existing sensor microfabrication processes.

Nanoparticles have contributed significantly to the design of new drug delivery and targeting in cancer chemotherapy. Consequently, some of the desirable qualities of silver and gold nanoparticles have prompted their novel applications in cancer nanobiotechnology. Glioblastoma is the most common and highest grade primary brain tumors in adults. Despite limited recent advances in treatment of this neurotumor, the prognosis of patients with this tumor remains extremely poor. In this study, we have investigated the hypothesis that silver and gold nanoparticles exert differential cytotoxic effects on human astrocytoma (glioblastoma) U87 cells. Our results demonstrate silver and gold nanoparticles induced time- and concentration-related effects in lowering the survival of U87 cells. Consistent with our hypothesis, the effects induced by silver nanoparticles were much more pronounced compared to those induced by gold nanoparticles. Highly sensitive and selective DNA detection plays a central role in many fields of research, and various assay platforms have been developed. Compared to homogeneous DNA detection, surface-immobilized probes allow washing steps and signal amplification to give higher sensitivity. Previously research was focused on developing glass or gold-based surfaces for DNA immobilization; we herein report hydrogel-immobilized DNA. We have numerically investigated the influence of a nanoscale silicon tip in proximity to an illuminated gold nanoparticle. We describe how the position of the high-permittivity tip and the size of the nanoparticle impact the absorption, peak electric field and surface plasmon resonance wavelength under different illumination conditions. We detail the finite element method (FEM) approach we have used, whereby we specify a volume excitation field analytically and calculate the difference between this source field and the total field (i.e., scattered-field formulation). Protein encapsulated gold nanoclusters have received much attention due to the possibility of using them as a non-toxic fluorescent probe or marker for biomedical applications, however one major disadvantage currently is their low brightness and quantum yield in comparison to currently used fluorescent markers. A method of increasing the fluorescence emission of Human Serum Albumin (HSA) encapsulated gold nanoclusters (AuNCs) via a Polyallylamide hydrochloride (PAH) coating is described. PAH molecules with a molecular weight of ~17,500 Da were found to enhance the fluorescence emission of HSA-AuNCs by 3-fold when the protein/polymer concentration ratio is 2:1 in solution. Interestingly, the fluorescence lifetime of the AuNCs was found to decrease while the native tryptophan (TRP) fluorescence lifetime also decreased during the fluorescence emission intensity enhancement caused by the PAH binding. Determination of the ligand effect on catalysis requires the use of gold nanoclusters with protecting ligands as the only variable. Two isostructural Au38 nanoclusters, [Au38(L)20(Ph3P)4]2+ (L = alkynyl or thiolate), have been synthesized by a direct reduction method, and they have an unprecedented face-centered cubic (fcc)-type Au34 kernel surrounded by 4 AuL2 staple motifs, 4 Ph3P, and 12 bridging L ligands. In this report, a gold nanoparticle decorated single walled carbon nanotube (SWCNTs) nanocomposite was shown to possess synergistic intrinsic peroxidase like activity and enhanced affinity towards H2O2 oxidation. The gold nanoparticle decorated SWCNTs nanocomposite was characterized by high catalytic activity, enhanced stability from the gold nanoparticles and improved dispersion from the SWCNTs. Subsequently, this nanocomposite was proved to be a novel peroxidase mimetic with great potential to catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) in the presence of H2O2 to yield a blue colored product. As a proof of concept, the gold nanoparticle decorated SWCNTs composite was used as a robust nanoprobe for the detection of D-alanine with improved analytical characteristics. Taking into account the valuable intrinsic peroxidase activity of the nanohybrid, the present work may find widespread applications in the field of sensors and biosensors for diverse applications.

The relationship between environmental exposure and health outcomes is complex, multidirectional and dynamic. Therefore, we need an understanding of these linkages for effective health risk communication. Despite the severe health hazards, artisanal gold mining is widespread globally, with an estimated 30 million people engaged in it. In this study, the relationships between artisanal gold miners knowledge of environmental and health effects of Hg and compositional, contextual and occupational factors were assessed using generalized linear models (negative log-log regression). A cross-sectional survey in three urban gold mining hubs in Ghana (Prestea, Tarkwa and Damang), was carried out among 588 (482 male and 106 female) artisanal gold miners. The results showed that 89% of artisanal gold miners had very low to low levels of knowledge whereas 11% had moderate to very high levels of knowledge of deleterious health effects of Hg. An evaluation of the parameters that affect desorption/ionization show that careful treatments of AuNPs is needed, as subtle changes in the solution environment can result in subsequent changes in the mass spectra. A thorough evaluation of the parameters that affect desorption/ionization of peptides is presented here, and these parameters include: (i) AuNP-to-analyte ratio, (ii) AuNP size, (iii) solvent, (iv) AuNP surface composition, (v) pH and buffer effects, (vi) amino acid sequence, and (vii) additives such as fructose or glycerol. This study reports on current work involving the use of Surface Enhanced Raman Spectroscopy (SERS) for the intracellular detection of cell constituents in mouse fibroblast cells using gold nanoshells. Gold nanoshells were acquired from Nanospectra Biosciences that are based on a silica dielectric core and an outer gold shell layer. They have the unique property of a tunable surface plasmon resonance wavelength from the visible through the near infrared which allows control of the electromagnetic field strength on its surface. Hence gold nanoshells can serve as SERS substrates with plasmonic properties that are not aggregation dependent and thus can be expected to overcome the reproducibility problem that is generally associated with aggregation based colloidal metal nanoparticles. We demonstrate the amplified detection of BRCAI gene based on the gold nanoparticle labeled DNA sensor. The sensor was based on a sandwich detection strategy, which involved an immobilized capture probe DNA (DNA-c), Target DNA (DNA-t) and gold nanoparticle conjugated reporter probe DNA (DNA-r.AuNP). The sensor surface was characterized by scanning electron microscopy (SEM) and scanning tunneling microscopy (STM). Detection capability of the sensor was studied with I-V measurements using either scanning tunneling microscopy (STM) or Keithley 2400 Source Meter SMU Instrument. Shifting of the surface plasmon resonance wavelength induced by the variation of the thickness of insulating spacer between single layer graphene and Au nanoparticles is studied. The system demonstrates a blue-shift of 29 nm as the thickness of the spacer layer increases from 0 to 15 nm. This is due to the electromagnetic coupling between the localized surface plasmons excited in the nanoparticles and the graphene film. The strength of the coupling decays exponentially with a decay length of d/R = 0.36, where d is the spacer layer thickness and R is the diameter of the Au nanoparticles. The result agrees qualitatively well with the plasmon ruler equation. Interestingly, a further increment of the spacer layer thickness induces a red-shift of 17 nm in the resonance wavelength and the shift saturates when the thickness of the spacer layer increases above 20 nm. © 2012 Optical Society of America ...
In this work, a chiral hybrid superstructure has been achieved through a hierarchical, cooperative self-assembly/self-organization process in a multiple-component system comprising of gold nanorods, surfactants, and phospholipids film. This chiral hybrid superstructure presented a signate signal in plasmonic circular dichroism with negative collective plasmonic modes on one side of the plasmon resonance and positive modes on the other side. The origin of such an unusual optical activity is fundamentally different from that of chiral metal nanoparticles/nanoclusters in previous studies. We highlight here a possibility that helical alignment of gold nanorods in the hybrid superstructure would resemble mesogenic molecules in a cholesteric liquid crystalline mesophase, and the Coulomb dipole-dipole interactions between gold nanorods in such a chiral liquid crystalline-like mesophase would give rise to the plasmonic circular dichroism effect. Abstract Nanoparticle vehicles may improve the delivery of contrast agents and therapeutics to diseased tissues, but their rational design is currently impeded by a lack of robust technologies to characterize their in vivo behavior in real?time. This study demonstrates that fluorescent?labeled gold nanoparticles can be optimized for in vivo detection, perform pharmacokinetic analysis of nanoparticle designs, analyze tumor extravasation, and clearance kinetics in tumor?bearing animals. This optical imaging approach is non?invasive and high?throughput. Interestingly, these fluorescent gold nanoparticles can be used for multispectral imaging to compare several nanoparticle designs simultaneously within the same animal and eliminates the host?dependent variabilities across measured data. Together these results describe a novel platform for evaluating the performance of tumor?targeting nanoparticles, and provide new insights for the design of future nanotherapeutics.

Hollow gold nanoparticles (HGNPs) exposed to near-infrared (NIR) light yield photothermal effects that can trigger a variety of biological effects for potential biomedical applications. However, the mechanism of laser-triggered drug release has not been studied before. A tripeptide Ac-Glu-Glu-Cys-NH2 (Ac-EEC) was directly linked to the surface of HGNPs. The EEC-HGNPs conjugate was then complexed with cisplatin Pt(II) to give Ac-EEC(Pt)-HGNPs. Folic acid was introduced to the gold surface of Ac-EEC-HGNPs through a thioctic acid-terminated polyethylene glycol linker (F-PEG-TA) followed by complexation with Pt(II) to give F-Ac-EEC(Pt)-HGNPs. Laser treatment was instituted with a 15-ns pulsed laser at a repetition rate of 10 Hz. The released Pt(II) was quantified by inductively coupled plasma mass spectroscopy, and the nature of the released Pt-containing species was characterized by liquid chromatography-mass spectroscopy. The displacement adsorption process of 4-mercaptopyridine (4-MP) on trisodium citrate preadsorbed gold surface in aqueous solution was studied using atomic force microscopy (AFM). The measurement was done by monitoring the interaction force between a gold-coated AFM tip and a gold substrate, both presaturated by trisodium citrate. The addition of 4-MP was found to result in a remarkable change of tip−substrate adhesion force, which showed a gradual increase and finally reached a maximum. Using the chemical force titration technique, we estimated the surface pK1/2 of thus-formed 4-MP displacement film and compared with pure 4-MP self-assembled monolayer (SAM) formed on gold in ethanol solution. The current median survival of patients with glioblastoma multiforme (GBM), the most common type of glioma, remains at 14.6 months despite multimodal treatments (surgery, radiotherapy and chemotherapy). This research aims to study the feasibility of photothermal ablation of glioma using gold nanoshells that are heated upon laser irradiation at their resonance wavelength.
