Cytoskeletal components of an invasion machine--the apical complex of Toxoplasma gondii. (1/147)

The apical complex of Toxoplasma gondii is widely believed to serve essential functions in both invasion of its host cells (including human cells), and in replication of the parasite. The understanding of apical complex function, the basis for its novel structure, and the mechanism for its motility are greatly impeded by lack of knowledge of its molecular composition. We have partially purified the conoid/apical complex, identified approximately 200 proteins that represent 70% of its cytoskeletal protein components, characterized seven novel proteins, and determined the sequence of recruitment of five of these proteins into the cytoskeleton during cell division. Our results provide new markers for the different subcompartments within the apical complex, and revealed previously unknown cellular compartments, which facilitate our understanding of how the invasion machinery is built. Surprisingly, the extreme apical and extreme basal structures of this highly polarized cell originate in the same location and at the same time very early during parasite replication.  (+info)

Regulation of ABCA1 expression in human keratinocytes and murine epidermis. (2/147)

Keratinocytes require abundant cholesterol for cutaneous permeability barrier function; hence, the regulation of cholesterol homeostasis is of great importance. ABCA1 is a membrane transporter responsible for cholesterol efflux and plays a pivotal role in regulating cellular cholesterol levels. We demonstrate that ABCA1 is expressed in cultured human keratinocytes (CHKs) and murine epidermis. Liver X receptor (LXR) activation markedly stimulates ABCA1 mRNA and protein levels in CHKs and mouse epidermis. In addition to LXR, activators of peroxisome proliferator-activated receptor (PPAR)alpha, PPARbeta/delta, and retinoid X receptor (RXR), but neither PPARgamma nor retinoic acid receptor, also increase ABCA1 expression in CHKs. Increases in cholesterol supply induced by LDL or mevalonate stimulate ABCA1 expression, whereas inhibiting cholesterol synthesis with statins or cholesterol sulfate decreases ABCA1 expression in CHKs. After acute permeability barrier disruption by either tape-stripping or acetone treatment, ABCA1 expression declines, and this attenuates cellular cholesterol efflux, making more cholesterol available for regeneration of the barrier. In addition, during fetal epidermal development, ABCA1 expression decreases at days 18-22 of gestation (term = 22 days), leaving more cholesterol available during the critical period of barrier formation. Together, our results show that ABCA1 is expressed in keratinocytes, where it is negatively regulated by a decrease in cellular cholesterol levels or altered permeability barrier requirements and positively regulated by activators of LXR, PPARs, and RXR or increases in cellular cholesterol levels.  (+info)

p53 is preferentially recruited to the promoters of growth arrest genes p21 and GADD45 during replicative senescence of normal human fibroblasts. (3/147)

Replicative senescence is the terminal growth arrest that most normal human cells enter into after a fixed number of divisions in vitro, limiting the proliferative potential of a cell and preventing genomic instability caused by critically short telomeres. Thus, senescence presents a tumor-suppressive mechanism and a barrier to tumor formation. However, senescent cells are inherently resistant to apoptosis and, as they accumulate in aging tissues, may contribute to organ dysfunction and promote tumor progression as part of the stromal environment. Replicative life span in normal human cells can be extended by inactivation of the tumor suppressor gene p53 or its direct target, the cyclin-dependent kinase inhibitor p21, suggesting a direct role for this pathway in senescence. However, p53 recruitment to promoters of target genes during replicative senescence has not been shown in live cells. In this study, we used chromatin immunoprecipitation to determine that p53 preferentially occupied the promoters of growth arrest genes p21 and GADD45 in senescent normal human diploid fibroblasts but not the promoters of other target genes that recruited p53 following doxorubicin-induced DNA damage, such as apoptosis regulators TNFRSF10b, TNFRSF6, and PUMA. This differential recruitment of p53 in senescent versus doxorubicin-treated fibroblasts was accompanied by differences in post-translational modification of p53. These data provide mechanisms for both the growth arrest mediated by p53 and the resistant nature of senescent cells to apoptosis despite p53 activity.  (+info)

Role of ABCG1 and ABCA1 in regulation of neuronal cholesterol efflux to apolipoprotein E discs and suppression of amyloid-beta peptide generation. (4/147)

Maintenance of an adequate supply of cholesterol is important for neuronal function, whereas excess cholesterol promotes amyloid precursor protein (APP) cleavage generating toxic amyloid-beta (Abeta) peptides. To gain insights into the pathways that regulate neuronal cholesterol level, we investigated the potential for reconstituted apolipoprotein E (apoE) discs, resembling nascent lipoprotein complexes in the central nervous system, to stimulate neuronal [3H]cholesterol efflux. ApoE discs potently accelerated cholesterol efflux from primary human neurons and cell lines. The process was saturable (17.5 microg of apoE/ml) and was not influenced by APOE genotype. High performance liquid chromatography analysis of cholesterol and cholesterol metabolites effluxed from neurons indicated that <25% of the released cholesterol was modified to polar products (e.g. 24-hydroxycholesterol) that diffuse from neuronal membranes. Thus, most cholesterol (approximately 75%) appeared to be effluxed from neurons in a native state via a transporter pathway. ATP-binding cassette transporters ABCA1, ABCA2, and ABCG1 were detected in neurons and neuroblastoma cell lines and expression of these cDNAs revealed that ABCA1 and ABCG1 stimulated cholesterol efflux to apoE discs. In addition, ABCA1 and ABCG1 expression in Chinese hamster ovary cells that stably express human APP significantly reduced Abeta generation, whereas ABCA2 did not modulate either cholesterol efflux or Abeta generation. These data indicate that ABCA1 and ABCG1 play a significant role in the regulation of neuronal cholesterol efflux to apoE discs and in suppression of APP processing to generate Abeta peptides.  (+info)

Scrambling of phospholipids activates red cell membrane cholesterol. (5/147)

Cholesterol is predicted to associate more strongly with the outer than the inner leaflet of plasma membrane bilayers based on the relative in vitro affinities of their phospholipids. Complex formation with the high-affinity species (especially saturated sphingomyelins) is said to reduce the chemical activity (escape potential or fugacity) of the sterol. We therefore tested the hypothesis that scrambling the sidedness of plasma membrane phospholipids of intact cells will increase the chemical activity of outer surface cholesterol. Upon activating the plasma membrane scramblase in intact human red cells by introducing ionomycin to raise cytoplasmic Ca++, phosphatidylserine became exposed and, concomitantly, the chemical activity of exofacial cholesterol was increased. (This was gauged by its susceptibility to cholesterol oxidase and its rate of transfer to cyclodextrin.) Similar behavior was observed in human fibroblasts. Two other treatments known to activate cell surface cholesterol (namely, exposure to glutaraldehyde and to low-ionic-strength buffer) also brought phosphatidylserine to the cell surface but by a Ca++-independent mechanism. Given that phospholipid scrambling is important in blood coagulation and apoptosis, the concomitant activation of cell surface cholesterol could contribute to these and other pathophysiological signaling processes.  (+info)

Central role of p53 in the suntan response and pathologic hyperpigmentation. (6/147)

UV-induced pigmentation (suntanning) requires induction of alpha-melanocyte-stimulating hormone (alpha-MSH) secretion by keratinocytes. alpha-MSH and other bioactive peptides are cleavage products of pro-opiomelanocortin (POMC). Here we provide biochemical and genetic evidence demonstrating that UV induction of POMC/MSH in skin is directly controlled by p53. Whereas p53 potently stimulates the POMC promoter in response to UV, the absence of p53, as in knockout mice, is associated with absence of the UV-tanning response. The same pathway produces beta-endorphin, another POMC derivative, which potentially contributes to sun-seeking behaviors. Furthermore, several instances of UV-independent pathologic pigmentation are shown to involve p53 "mimicking" the tanning response. p53 thus functions as a sensor/effector for UV pigmentation, which is a nearly constant environmental exposure. Moreover, this pathway is activated in numerous conditions of pathologic pigmentation and thus mimics the tanning response.  (+info)

Up-regulation of MET expression by alpha-melanocyte-stimulating hormone and MITF allows hepatocyte growth factor to protect melanocytes and melanoma cells from apoptosis. (7/147)

The MET proto-oncogene encodes for the hepatocyte growth factor (HGF) receptor, a plasma membrane tyrosine kinase that is involved in melanocyte growth and melanoma development. In mouse melanoma cells, Met expression is increased by alphaMSH via the activation of the cAMP pathway. However, the mechanism by which cAMP regulates MET and the biological consequences of this increase were not known. In the present report, we show that alphaMSH regulates MET expression in both human melanocytes and mouse melanoma cells through a transcriptional mechanism that requires MITF. Furthermore, the adenovirus driven expression of MITF is sufficient to increase MET in melanoma cells. Functional analysis of the MET promoter allows us to identify an E-box motif conserved in both human and mouse promoter that mediates the effect of MITF. Interestingly, up-regulation of MET expression by cAMP leads to an exacerbated HGF signaling and allows HGF to protect melanocytes and melanoma cells from apoptosis. Thus, physiological stimuli or pathological events that would induce MITF expression may lead to increased MET expression thereby favoring melanoma survival. These observations strengthen the roles of MITF and MET in melanoma development.  (+info)

Divergent polyamine metabolism in the Apicomplexa. (8/147)

The lead enzymes of polyamine biosynthesis, i.e. ornithine decarboxylase (ODC) and arginine decarboxylase (ADC), were not detected in Toxoplasma gondii [the limit of detection for ODC and ADC was 5 pmol min(-1) (mg protein)(-1)], indicating that T. gondii lacks a forward-directed polyamine biosynthetic pathway, and is therefore a polyamine auxotroph. The biochemical results were supported by results obtained from data-mining the T. gondii genome. However, it was possible to demonstrate the presence of a highly active backconversion pathway that formed spermidine from spermine, and putrescine from spermidine, via the combined action of spermidine/spermine N(1)-acetyltransferase (SSAT) or spermidine N(1)-acetyltransferase (SAT) and polyamine oxidase (PAO). With spermine as the substrate, T. gondii SSAT had a specific activity of 1.84 nmol min(-1) (mg protein)(-1), and an apparent K(m) for spermine of 180 mM; with spermidine as the substrate, the SAT had a specific activity of 3.95 nmol min(-1) (mg protein)(-1), and a K(m) for spermidine of 240 mM. T. gondii PAO had a specific activity of 10.6 nmol min(-1) (mg protein)(-1), and a K(m) for acetylspermine of 36 mM. Furthermore, the results demonstrated that T. gondii SSAT was 50 % inhibited by 30 mM di(ethyl)norspermine. The parasite actively transported arginine and ornithine, which were converted via the arginine dihydrolase pathway to citrulline and carbamoyl phosphate, resulting in the formation of ATP via carbamate kinase. The lack of polyamine biosynthesis by T. gondii is contrasted with polyamine metabolism by other apicomplexans.  (+info)

Herpes simplex virus type 2 (HSV2), the most common cause of genital herpes, increases a womans risk of HIV acquisition from 3-6 fold, perhaps because HSV2-infected women have increased numbers of HIV target cells (CD4 T cells and dendritic cells) in the cervical mucosa. However, recent clinical trials showed no impact of HSV2 suppression on HIV acquisition rates. The reasons for this negative result are unclear. The investigators propose to examine the effect of valacyclovir (a widely used herpes medication) treatment on cervical immunology and HIV target cells in the cervix. The study will take the form of a randomized, double-blind, placebo-controlled crossover trial. Primary endpoints will be (1) the number of CD4 T cells on a cervical cytobrush and (2) the number of immature dendritic cells per cervical cytobrush ...
Bacterial vaginosis (BV), a disruption of the normal vaginal flora, has recently been associated with a greater than 3-fold increased risk of female-to-male HIV-1 transmission in a manner not accounted for by plasma HIV-1 RNA load(1). We hypothesize that compared to cervicovaginal fluid (CVF) from healthy individuals, BV CVF induces an elevated pro-inflammatory and pro-chemotactic response in cells of the foreskin which may facilitate transmission. Because foreskin epithelial cells are one of the first cells to encounter HIV during female-to-male transmission, we propose to compare the effects of healthy vs. BV CVF on primary epithelial cells cultured from human foreskin tissues. These studies will take advantage of the extensive experience the PIs UCSF mentor has had in culturing primary epithelial cells from genital tract tissue, and examining the response of these cells to genital secretions(2). We propose to expose epithelial cells cultured from foreskin tissue to healthy and BV CVF, and to ...
Vactruth [2]. Warning: Some people may find the information in this article disturbing and the images graphic.. Every year, some infants are circumcised. During this surgical procedure, part of the childs protective penile tissue is removed. This tissue removed from his penis may be sold to companies and institutions seeking the rich human fibroblast cells and other cells it contains. Most people are unaware that for decades, vaccine companies have been using these foreskin cells to research, grow and develop vaccines.. Certain microorganisms used by vaccine companies need living human cells to replicate. The cells within foreskin are being used for this purpose. Foreskin cells can be used to turn a wild-type microorganism found in nature into a genetically modified microorganism for use in vaccines.. Normal human foreskin keratinocytes cotransfected with the neomycin resistance gene and recombinant human papillomavirus (HPV) DNAs (types 16, 18, 31, and 33) that have a high or moderate association with cervical malignancy acquired immortality and contained integrated and transcriptionally active viral genomes. Only transcripts from the intact E6 and E7 genes were detected in at least one cell line, suggesting that one or both of these genes are responsible for immortalization. Recombinant HPV DNAs with low or no oncogenic potential for cervical cancer (HPV1a, -5, -6b, and -11) induced small G418-resistant colonies that senesced as did the nontransfected cells. These colonies contained only episomal virus DNA; therefore, integration of HPV sequences is important for immortalization of keratinocytes. This study suggests that the virus-encoded immortalization function contributes to the pathogenesis of cervical carcinoma. ...
The foreskin, like the eyelid, also serves an important protective and hygienic function. The foreskin protects the delicate glans of the penis and puts the urethra at a distance form its environment, protecting it from foreign contaminants of all kinds while simultaneously shielding the penis from injury. It is a double fold of skin which offers two layers of protection. At birth, the foreskin is usually attached to the glans (head) of the penis by a synechia, akin to how a fingernail is attached to a finger.[1] The preputial cavity is sealed by the synechia so it cannot be infected. In infancy, the foreskins tubular neck (prepucial orifice) is often long and narrow while the sphincter muscle in the tip of the foreskin keeps its opening closed. This acts as an extension of the urethra.[2][3] Together, these properties prevent the entry of contaminants.[4] The foreskin protects the infant glans penis from ammonia in diapers (nappies) and prevents meatal ulcer and meatal stenosis.[5] [6] [7] ...
Pardon the tangent, but as long as yall are on the subject of circumcision, does anyone have a foreskin that doesnt retract? My six year old sons foreskin doesnt retract, and hes beyond the age of two or three or four when the foreskin is supposed to naturally pull back. The condition is called phimosis. Some doctors freak out about it and recommend circumcision. Our pediatrician says we should leave it as is, and just see if it eventually retracts, or if not, let him participate in the decision about whether to get cut after he reaches puberty. The doctor says it will not restrict the growth of his penis. But Im wondering: do any of you grown ups have a foreskin that doesnt retract? Hows it working? And how does it feel? Any hygienic concerns? Im cut so I cant imagine having a foreskin, let alone one that doesnt retract ...
Methods Inter-rater comparison of the responses on foreskin status and circumcision in a self-report questionnaire with the findings of a physical examination by an experienced well-trained urologist was performed for patients who visited a healthcare facility in Kanagawa, Japan. Foreskin status was defined using a five-point graphical scale based on the degree to which the foreskin covers the foreskin and the glans penis in a non-erectile condition: type 1, a fully exposed glans penis; types 2-4, the glans penis partly covered by the foreskin and type 5, phimosis. Linear weighted κ and per cent agreement were used to evaluate the reliability of responses. ...

One of the primary orgasmic nerve receptors in the foreskin. In the penis, they are concentrated in the ridged band, inner foreskin and the frenulum.. A normal foreskin contains thousands of these coiled fine-touch receptors. They are able to detect subtle changes in motion, as well as fine gradations in texture. Circumcision typically removes most of these receptors.. Reference: Wikipedia entry for Meissners Corpuscles. ...

Results: Vaginal epithelial thickness was similar in women using injectable HCs compared to non-injectable HC users. The frequency of CD4+ T cells in the vaginal squamous epithelium of injectable HC users was significantly higher than non-injectable HC users (p=0.028). CD68+ macrophage cell density did not differ between women using injectable HCs and those not using injectable HCs, although macrophages were closer to the vaginal luminal surface in injectable HC users than those not using HCs (p=0.021). Furthermore, the frequency of mucosal CD68+ macrophages during the acute infection were positively associated with the concentration of the RANTES (beta coefficient (β)=0.779, p=0.024), MCP-1 (β=0.453, p=0.041), IP-10 (β=0.568, p=0.042), IL-7 (β=1.332, p=0.018), IL-9 (β=0.336, p=0.015), and IL-17 (β=1.058, p=0.007) in CVL, after adjusting for multiple comparisons ...
Cloaking immune cells with antibodies that block T cell trafficking to the gut can substantially reduce the risk of viral transmission in a non-human primate model of HIV infection, scientists report.. The findings suggest that drugs that are already in clinical trials for inflammatory bowel diseases might be effective in the treatment or prevention of HIV infection.. The results were published Monday in Nature Medicine.. We were surprised by the effects that we observed, says senior author Aftab Ansari, PhD, professor of pathology and laboratory medicine at Emory University School of Medicine and Yerkes National Primate Research Center.. Our goal was to demonstrate that blocking CD4+ T cell trafficking to gut tissues could decrease the level of virus in the gut and help mount an effective immune response, but we found that administration of the antibody could actually prevent transmission. The results of this study suggest that this form of therapy could counteract pathologies that lead to ...

For those uncomfortable with metric measurements, 43.3 square centimetres is 6.71 square inches, and 36.8 square centimetres is 5.7 square inches. To my knowledge, this is the first study of foreskin surface area in the literature - I have never been able to identify any evidence for the 15 square inches commonly claimed by the anti-circumcision lobby, which appears to be merely a guess, and apparently a rather over-inflated guess. So now that we have hard data, we can expect to see that figure disappear. I wish I wasnt joking - Ive never yet seen the anti-circumcision lobby abandon any of their myths in the face of contrary evidence - but I might be pleasantly surprised ...

There is no need for stitches in the postnatal time and there is practically no blood shed. Simple surgery!. But this simple act takes away an area of skin whose underside, which faces the head of the penis, is a mucous membrane, as is the head of the penis. After circumcision the skin of the glans loses its mucous quality, becoming more like outer skin. This desensitizes some of the nerve endings of the head. Also, with the loss of the foreskin the penis loses a natural gliding mechanism that is helpful for insertion during intercourse.. One of the arguments for circumcision is that the uncircumcised head naturally collects smegma (old mucous secretion, etc.) If allowed to build up may be an ideal breeding ground for germs. But if a boy can be taught to brush his teeth, clean his ears and wipe his anus he can also be taught to retract his foreskin and wash the head of his penis! As a baby the foreskin is tight over the head and has a membrane attached which closes the area (sort of like a ...
Foreskin restoration by manual tugging with method 3, which is manual restoring by tugging the foreskin of the penis of a circumcised or intact man.
The penis -- in its unaltered state with an intact foreskin -- is pretty clearly designed for sexual activity where it is inserted, and where it can move around during sexual activities without a need for any special assistance. Not only do you not have to do anything at all to your foreskin at all during any kind of intercourse, its designed in such a way to work optimally with that activity. While we need to be careful how we talk about genitals, sex and design, since we cant consult with the designer and so can only theorize, we can safely say that even if someone chooses not to reproduce, the penis clearly has a lot to do with reproduction, and the way it is, foreskin and all, is conducive to that purpose through vaginal intercourse without aid. Given the massive population of the planet, and knowing the majority of men worldwide are not circumcised, you can rest assured the uncircumcised penis works just fine in that regard. While the practice of circumcision dates back further than ...

The resale value of neonate foreskins is astronomically dizzying in that from one boys foreskin can be grown bio-engineered skin in a lab to the size of a football field. Thats 4 acres of new skin or around 200,000 units of manufactured skin, which is enough skin to cover about 250 people and sells at $3,000 a square foot. Considering that there are 1.25 million neonate foreskins circumcised each year in the U.S alone this translates to one of the most lucrative trades, if not THE most lucrative trade in human body parts ever in the history of humanity ...

Question - White spots on foreskin, no itching, no pain, tight foreskin. Ointment?. Ask a Doctor about diagnosis, treatment and medication for Vitiligo, Ask a Dermatologist
Pull the foreskin all the way back so its tight on the shaft. Then lubricate it and just rub up and down, stretching and pulling it at the same time as rubbing. Just concentrate on the foreskin - the glans is too sensitive unless its well lubricated. The foreskin sensation is very sharp - almost painful - and produces a big orgasm. If you dont stop, the sensation is strong enough to maintain the erection.. ...

In drawing the foreskin forward in this manner there is danger that its outer portion will be cut farther back than will its inner portion; hence, after the first clip of the scissors the traction upon the clamps should be relaxed and the reflected portion of the foreskin should be cut farther if necessary. Two clamps are then placed upon the orifice of the foreskin at its lower edge and an incision is made between them. This incision is far shorter than the dorsal one. The two clamps on the left side are then drawn outward and left half of the foreskin is removed, care being taken that the incision through the inner layer of the foreskin shall be nearly parallel to the corona of the glans, and that the incision through the external layer shall be directly opposite to it when only slight traction is made upon the clamps. The best result is obtained when the portion of the inner layer which is left is a third or a half of an inch in width. The right half of the foreskin is next cut away. Any ...
Why the big deal over a petty thing like foreskin? Are you all angry because you think if you werent circumsized youd feel a little more pleasure? Face it, men dont feel as much as women. Pleasure shouldnt be a big issue. You want stronger emotions too? Why dont you be content. Foreskin isnt like a clit. You dont get a drastic change in feeling if you lose a little skin. The skin doesnt have feeling anyways. Its the head that has feeling. The clot is sensitive. Your outer skin is nothing like a clit. Stop listening to women. They dont have the slightest clue on those things nor do they even care. Havent you learned of their selfishness yet ...

The big studios seem to be running out of superheros for the summer movie season. They must be scraping the bottom of the barrel if the best they can do now is Thor and the Green Lantern.. I have a solution: A brand new superhero, minted just last week, and a big, big Hollywood star who seems hyped to play him.. Weve recently been introduced to the comic book Foreskin Man, the quite blatantly, Medieval-style anti-Semitic creation of one Matthew Hess, a key figure behind the ballot initiative to ban circumcision in San Francisco. The comics hero fights off the dark, Shylock-looking character known as Mohel Monster who brandishes his knife and looks to cut up Jewish babies penises. Foreskin Man is a waspy, blond haired hunk in a yellow cape whose real identity is the mild, mannered bachelor Miles Hastwick.. ...

And now when I start to clean myself after jerking off, Most of the time this Frenulum really bothers me. I try to slowly pull down my foreskin. That area on my skin has turned a bit white. Like as if it has gotten stretched too much. What should I do? Does anyone else here have the same problem? In males who have been circumcised, the circumcision scar refers to the scar after a circumcision has healed. In some cases, the scar can be darker-colored, and, in all cases, it will encircle the shaft of the penis. The scar, which completely encircles the shaft of the penis, is located at the boundary of the shaft skin and the inner foreskin remnant, which is the portion of the foreskin that was not removed during circumcision. This foreskin remnant is mucosa that lies between the glans and the circumcision scar, which results in dissimilar tissue healing together. In adult circumcision part of the frenulum may remain intact. The foreskin remnant is dried mucosa and can often have a different color and texture than the rest of the penile skin. It can be pinkish or light-colored, and it typically becomes covered with keratin to protect it from a dry environment. Some circumcision scars result in a marked color difference on the shaft. The characteristics of the circumcision scar depend on the ...
The World Health Organization say that there are lots of uses for the foreskin, such as keeping the glans penis moist, protecting it or to make sexual intercourse feel nicer since it has so many nerves in it.[3] The removal of the foreskin, the circumcision, reduces diseases.. However, Moses and Bailey (1998), say that it has not been demonstrated that [the foreskin] is associated with increased male sexual pleasure.[4]. ...
The most common masturbation technique among males is to hold the penis with a loose fist and then to move the hand up and down the shaft. This type of stimulation is typically all that is required to achieve orgasm and ejaculation. The speed of the hand motion will vary, although it is common for the speed to increase as ejaculation nears and for it to decrease during the ejaculation itself.[18]. Male masturbation techniques may differ between males who have been circumcised and those who have not. Some techniques which may work for one individual can be difficult or uncomfortable for another. For males who have not been circumcised, stimulation of the penis typically comes from the pumping of the foreskin, whereby the foreskin is held and slid up and down over the glans, which, depending on foreskin length, is completely or partially covered and then uncovered in a rapid motion. The outer foreskin glides smoothly over the inner foreskin. The glans itself may widen and lengthen as the ...
Tight foreskin refers to phimosis, a normal condition in babies but consider abnormal in older children and adults where foreskin is very tight to be pulled back over the glans penis.
Those boys or men who havent been circumcised are exposed to the danger of getting phimosis. This is a condition which happens when the foreskin of a male penis cannot be moved back or pulled fully thus causing problems. When boys grow older their foreskin should become retractable. If not its best to get phimosis treatment while the boys are still young as if you delay it for later age it can cause more problems for the person. If when a boy is two or three years old you notice that this skin of his penis is not loosening up you should seek help from an expert doctor for phimosis treatment. Sometimes mothers fail to recognize this condition as it doesnt cause much problem for young boys. But as they grow older, if they dont get phimosis treatment they face difficulty in retracting their skin. Without phimosis treatment they will begin to face difficulty in urinating, the penis might also get sore or red and especially when having sexual intercourse it can cause problems if phimosis treatment ...

Thanks very much for this incredibly helpful site. I have a question about stretching exercises. Like many others on the site, I find it difficult to make a lot of time for these. So I was wondering how effective it would be to retract the foreskin as far as possible (maybe even over the glans, though thats still painful for me if an erection then ensues) and leave it that way for as long as possible - the advantage being that you can get on with other stuff while the foreskin is already in stretch mode. Will this do the trick, or do I have to make time for regular two-fingered stretching exercises ...
Foreskin balanitis is often due to prepuce, smegma, and not paying attention to local hygiene. This will lead to the accumulation of dirt within the foreskin and bacterial infection, and hence symptoms onset. Early symptoms include glans and foreskin surface edema, congestion, reddish urethral area, the emergence of wounds and erosion. the symptoms can develop into superficial ulcers, purulent discharge outflow, and the patient will feel itchy or have a burning sensation followed by pain. After ulcer is developed, pus will be secreted and it will stink. Severe cases can lead to fatigue, fever, inguinal lymph node swollen and pain.. ...

July 13th, 2012 at 11:01 am the foreskin is not a birth defect and being born a healthy male in this country does not require surgical correction. all normal human beings-male and female-are born with a foreskin. information about the foreskin and its NUMEROUS functions, are always missing from the discussion about circumcision. depending on the amount of skin cut off, circumcision robs a male of as much as 80 percent or more of his penile skin and cuts off necessary veins, arteries and capillaries. the foreskin contains more than 20,000 nerve endings, which is why genital cutting is excruciatingly painful. it is often done without anesthesia (inhuman and CRUEL) because the foreskins are sold and must remain in pristine condition. you can find this information on the internet. anyone considering genital cutting for their newborn owes it to themselves and their son, to watch the actual procedure. download an actual circumcision, educate yourself about this unncessary surgery. the screams of a ...
Phimosis is a condition where the foreskin is too tight. Mr Gill provides treatment for this condition in Harrogate, North Yorkshire.
Gomco Clamp and Plastibell Device. The techniques for performing circumcision with the Gomco clamp and the Plastibell device have many of the same features. Following preparation of the infant and inspection of the penis and scrotum as described above, the foreskin is grasped with two mosquito clamps at the 10 oclock and 2 oclock positions (Figure 2a). The nondominant hand holds these clamps and uses them to elevate the foreskin. A third mosquito clamp is used as a probe to gently lyse any adhesions under the foreskin from the 8 oclock to the 4 oclock positions and is then clamped approximately two-thirds of the distance from the foreskin opening to the corona. This action creates the crushed area for the dorsal slit. The third clamp remains in place for one minute. The dorsal slit is then cut through the middle of the crushed area, using tissue scissors (Figure 2b). The foreskin is peeled back, and any additional adhesions are lysed using a blunt probe. Attention must be paid to preserving ...
Both the upper (endocervix and uterus) and lower (ectocervix and vagina) female genital tract mucosa are considered to be target sites for sexual transmission of HIV. There are a few reports on the T cell and antigen-presenting cell distribution in human endometrial tissue however, there is little known about the expression of the HIV co-receptor CCR5 and HIV-binding C-type lectin receptors on endometrial cell subsets. We therefore assessed endometrial tissue sections from HIV seronegative women undergoing hysterectomy of a benign and non-inflammatory cause for phenotypic characterization of potential HIV target cells and receptors by immunohistochemistry. Langerin was expressed on intraepithelial CD1a+CD4+ and CD11c+CD4+ Langerhans cells. Furthermore, CCR5+CD4+CD3+ T cells, DC-SIGN+MR+CD11c+ myeloid dendritic cells and MR+CD68+ macrophages were found within or adjacent to the epithelium of the uterine lumen. In addition, occasional CD123+BDCA-2+ plasmacytoid dendritic cells were detected deep ...

Does your anti-circumcision stance get you laid more?. It does, but not in the way you might be thinking. Part of being an intactivist involved restoring my own foreskin as much as possible. The nerve endings didnt grow back, of course, but I was able to completely reverse the keratinization damage by stretching my remaining shaft skin over the glans. When the head of the penis is kept moist and sensitive like nature intended, the result is a significant increase in sensation. The gliding mechanism of the foreskin is restored to some extent, as well. That combination allows me to perform much longer (and more gently) during sex than I could in the past.. This work shows that three-dimensional images from MPLSM of endogenous tissue fluorescence can effectively distinguish between normal, precancerous, and cancerous epithelial tissues. The high-resolution capability of MPLSM enabled the identification of three morphologic features (nuclear density ratio, keratin, and epithelial thickness), which showed statistically significant differences between normal and both precancerous and cancerous tissues. Moreover, statistically significant differences were observed in the keratin thickness of dysplasia and both types of SCCs and in the epithelial thickness of dysplasia and CIS. The lack of statistically significant differences in the epithelial thickness of dysplasia and SCCs may be because the imaging depth was limited by signal attenuation in the SCCs, thus underestimating the epithelial thickness in these samples. The statistically significant morphologic changes that accompany precancer and cancer development in the MPSLM images ( Table 1) are ...

20. At the age of 17 and in my final year of school I still could not retract my foreskin. Talking to a circumcised friend about it, he said that something should be done and he persuaded me to try and pull the skin back. Of course, as I did so, with him watching, I got a hard-on, which made it even more difficult, but eventually, and in great pain, it came back as I shot a load of cum all over the place! Strangely enough, at boarding school we had no medical examinations, but Im sure if we had, Id have been circumcised without delay. Ive always had a fascination with circumcision, as most of my school friends were cut and it always seemed that the successful boys were all circumcised. I know of 2 boys who returned after the holidays without their foreskins, but as they were in different years, never discussed it with them.. Even when I married at age 27 my helmet was still quite tender, but by now the foreskin was loose and easy to manipulate.. I had my first bout of posthitis/balanitis when ...
Hi. I am a 29 year old male and I just started having a problem with my non-circumsized penis. At first It started itching, then I noticed that the inside of the foreskin looked dry and flaky. After, the foreskin started to get swollen. Now its hard for me to expose my penis from my foreskin due to it being swollen and I keep on feeling something coming out rather than urine. I have checked it out and its clear and sticky. Recently I started to drink alot of water and since then I have been using the restroom more than usual. Also, this really started when my girlfriend and I last had intercourse. We were on the road and I did not get a chance to wash after intercourse until the next day and a half. She has been my only partner since a year weve been together and I am sure I have been hers and we do not have any STDs ...
Oh my goodness, your brother was not taught proper care of his intact penis at ALL! I am a mom to two intact boys. Neither has ever had an infection or any sort of problem with their foreskin or penis. Im guessing that your brother was taught to OVERclean in and around his foreskin. But just as we teach our little girls to avoid getting soap inside their labia, little boys need to be taught to avoid getting soap inside the foreskin. Water is all it ever needs, and it takes less care than keeping fingers clean, believe me. I left my boys alone for the most part - they played with it in the bath, stretching it and allowing water to circulate as needed. Thats IT for care of the intact penis. We knew one little boy who had an infection in his intact penis because his mother forcibly retracted it to lather it up with soap. Bad idea. Think about it, the equipment of girls is much more hidden and has many more folds and pockets, and yet we dont worry about cleaning and scrubbing out every square ...
