Analysis of gabapentin in serum and plasma by solid-phase extraction and gas chromatography-mass spectrometry for therapeutic drug monitoring. (1/4528)

A simple method for the determination of gabapentin (Neurontin) is described. The method uses solid-phase extraction by disk column and derivatization followed by gas chromatographic-mass spectrometric analysis. The single-step derivatization with MTBSTFA produces a t-BDMS derivative of both the carboxylic and amine moieties of the molecule. Each step of the procedure was optimized to assure reliable performance of the method. The assay limit of detection was 0.1 microg/mL with a linear range from 1.0 to 35 microg/mL. Within-run (n = 3) and between-run (n = 40) coefficients of variation were less than 8.2 and 15.9%, respectively. The method has proven reliable in routine production for more than a year, producing clean chromatography with unique ion fragments, consistent ion mass ratios, and no interferences. Statistical analysis of the gabapentin concentrations measured in 1020 random specimens over a 2-month period showed a mean concentration of 6.07 microg/mL with a standard deviation of 5.28.  (+info)

Nuclear location of mammalian DNA polymerase activities. (2/4528)

Nuclei were isolated from monolayer cultures of mouse and human cells using a nonaqueous procedure of cell fractionation in which lyophilized cells were homogenized and centrifuged in 100% glycerol. In previous work we have shown that the nuclear pellet and cytoplasmic supernatant fraction contained 10% or less of the nucleic acids characteristic of the other cell fraction. Aqueous extracts made from fresh cultures and from nonaqueous material at each step of the fractionation procedure were assayed fro DNA polymerase activity. Activities were normalized to DNA contents of extracted material. Specific activity was preserved quantitatively through freezing and drying the cells, but was found to be unstable in glycerol suspensions with approximate half-lives and 1 h at 23 degrees and 4 h at 0-4 degrees. Activities were relatively stable at -25 degrees, however, so that by homogenizing only 15 min at 4 degrees and centrifuging at -25 degrees we preserved approximately 85% of the specific activity of fresh cultures in the nonaqueous nuclear fraction. Sedimentation analyses showed that the nuclear fraction contained both DNA polymerase-alpha and-beta in approximately the proportions expected if all polymerase activities were confined to the nucleus in living cells. DNA polymerase-alpha was found to be more unstable in glycerol suspensions than DNA polymerase-beta. Nuclear location of both activities was found in exponential cultures and in 3T3 mouse cultures synchronized in the G1 and S phases of the cell division cycle. We found no evidence for cytoplasmic factors affecting nuclear polymerase activities. We have concluded that the two major DNA polymerases are nuclear although one, DNA polymerase-alpha, frequently is present as a weakly bound nuclear protein.  (+info)

Native display of complete foreign protein domains on the surface of hepatitis B virus capsids. (3/4528)

The nucleocapsid of hepatitis B virus (HBV), or HBcAg, is a highly symmetric structure formed by multiple dimers of a single core protein that contains potent T helper epitopes in its 183-aa sequence. Both factors make HBcAg an unusually strong immunogen and an attractive candidate as a carrier for foreign epitopes. The immunodominant c/e1 epitope on the capsid has been suggested as a superior location to convey high immunogenicity to a heterologous sequence. Because of its central position, however, any c/e1 insert disrupts the core protein's primary sequence; hence, only peptides, or rather small protein fragments seemed to be compatible with particle formation. According to recent structural data, the epitope is located at the tips of prominent surface spikes formed by the very stable dimer interfaces. We therefore reasoned that much larger inserts might be tolerated, provided the individual parts of a corresponding fusion protein could fold independently. Using the green fluorescent protein (GFP) as a model insert, we show that the chimeric protein efficiently forms fluorescent particles; hence, all of its structurally important parts must be properly folded. We also demonstrate that the GFP domains are surface-exposed and that the chimeric particles elicit a potent humoral response against native GFP. Hence, proteins of at least up to 238 aa can be natively displayed on the surface of HBV core particles. Such chimeras may not only be useful as vaccines but may also open the way for high resolution structural analyses of nonassembling proteins by electron microscopy.  (+info)

Specificity of native-like interhelical hydrophobic contacts in the apomyoglobin intermediate. (4/4528)

On exposure to mildly acidic conditions, apomyoglobin forms a partially folded intermediate, I. The A, B, G, and H helices are significantly structured in this equilibrium intermediate, whereas the remainder of the protein is largely unfolded. We report here the effects of mutations at helix pairing sites on the stability of I in three classes of mutants that: (i) truncate hydrophobic side chains in native helix packing sites, (ii) truncate hydrophobic side chains not involved in interhelical contacts, and (iii) extend hydrophobic side chains at residues not involved in interhelical contacts. Class I mutants significantly decrease the stability and cooperativity of folding of the intermediate. Class II and III mutants show smaller effects on stability and have little effect on cooperativity. Qualitatively similar results to those found in I were obtained for all three classes of mutants in native myoglobin (N), demonstrating that hydrophobic burial is fairly specific to native helix packing sites in I as well as in N. These results suggest that hydrophobic burial along native-like interhelical contacts is important for the formation of the cooperatively folded intermediate.  (+info)

Stretching lattice models of protein folding. (5/4528)

A new class of experiments that probe folding of individual protein domains uses mechanical stretching to cause the transition. We show how stretching forces can be incorporated in lattice models of folding. For fast folding proteins, the analysis suggests a complex relation between the force dependence and the reaction coordinate for folding.  (+info)

Stabilization of L-ascorbic acid by superoxide dismutase and catalase. (6/4528)

The effects of superoxide dismutase (SOD) and catalase on the autoxidation rate of L-ascorbic acid (ASA) in the absence of metal ion catalysts were examined. The stabilization of ASA by SOD was confirmed, and the enzyme activity of SOD, which scavenges the superoxide anion formed during the autoxidation of ASA, contributed strongly to this stabilization. The stabilization of ASA by catalase was observed for the first time; however, the specific enzyme ability of catalase would not have been involved in the stabilization of ASA. Such proteins as bovine serum albumin (BSA) and ovalbumin also inhibited the autoxidation of ASA, therefore it seems that non-specific interaction between ASA and such proteins as catalase and BSA might stabilize ASA and that the non-enzymatic superoxide anion scavenging ability of proteins might be involved.  (+info)

Incorporation rates, stabilities, cytotoxicities and release of liposomal tetracycline and doxycycline in human serum. (7/4528)

Tetracycline and doxycycline were encapsulated in cationic, anionic and neutral liposomes. The amounts of antibiotic encapsulated, the stability of each preparation at 4 degrees C for 4 weeks, and the kinetics of the release of entrapped drug into human sera were assessed by high-performance liquid chromatography. The toxicities of the liposome preparations on human erythrocytes and HeLa 229 cells were evaluated in vitro. The results showed that doxycycline was entrapped more efficiently than tetracycline, and that doxycycline-entrapped liposomes were more stable at 4 degrees C and in human sera, and less cytotoxic than tetracycline-entrapped liposomes.  (+info)

Interaction of asparagine and EGF in the regulation of ornithine decarboxylase in IEC-6 cells. (8/4528)

Our laboratory has shown that asparagine (ASN) stimulates both ornithine decarboxylase (ODC) activity and gene expression in an intestinal epithelial cell line (IEC-6). The effect of ASN is specific, and other A- and N-system amino acids are almost as effective as ASN when added alone. In the present study, epidermal growth factor (EGF) was unable to increase ODC activity in cells maintained in a salt-glucose solution (Earle's balanced salt solution). However, the addition of ASN (10 mM) in the presence of EGF (30 ng/ml) increased the activity of ODC 0.5- to 4-fold over that stimulated by ASN alone. EGF also showed induction of ODC with glutamine and alpha-aminoisobutyric acid, but ODC induction was maximum with ASN and EGF. Thus the mechanism of the interaction between ASN and EGF is important for understanding the regulation of ODC under physiological conditions. Therefore, we examined the expression of the ODC gene and those for several protooncogenes under the same conditions. Increased expression of the genes for c-Jun and c-Fos but not for ODC occurred with EGF alone. The addition of ASN did not further increase the expression of the protooncogenes, but the combination of EGF and ASN further increased the expression of ODC over that of ASN alone. Western analysis showed no significant difference in the level of ODC protein in Earle's balanced salt solution, ASN, EGF, or EGF plus ASN. Addition of cycloheximide during ASN and ASN plus EGF treatment completely inhibited ODC activity without affecting the level of ODC protein. These results indicated that 1) the increased expression of protooncogenes in response to EGF is independent of increases in ODC activity and 2) potentiation between EGF and ASN on ODC activity may not be due to increased gene transcription but to posttranslational regulation and the requirement of ongoing protein synthesis involving a specific factor dependent on ASN.  (+info)

Title: Solution Stability - Plasma, Gastrointestinal, Bioassay. VOLUME: 9 ISSUE: 9. Author(s):Li Di and Edward H. Kerns. Affiliation:Wyeth Research, Princeton, NJ 08543-8000, USA.. Keywords:Plasma stability, solution stability, simulated gastro-intestinal fluids, stability-ph profile, high throughput, drug discovery, HPLC, LC-MS. Abstract: Solution stability of drug candidates in plasma, gastrointestinal fluids and bioassays is important in order to achieve low clearance, good oral bioavailability and have robust SAR. Screening of solution stability early in drug discovery can avoid pursuing hits with high risk of instability, prioritize chemical series, guide structural modification, and enhance the chance of project success. The conditions of solution stability methods are critical in generating relevant data and include: test compound concentration, enzyme source and preparation, limits of solubility, cosolvent, plasma protein binding effect, detection techniques (LC-UV vs. LC-MS), and what ...

AU - Galanti, Laurence. PY - 2017/8/8. Y1 - 2017/8/8. N2 - In hospitals, a major part of the drugs is administered via the intravenous route. When one wants to evaluate or know the stability of a drug in solution, it is necessary to know several physico-chemical parameters. Several reference works are available to help the hospital pharmacist with this research. However, reading these different sources can make you discover conflicting data. It is therefore necessary at this time to obtain the publications with contradictory results and to read them again. Seven criteria have been identified for judging the quality of a publication on physicochemical stability: full description of equipment, methods and analytical conditions of molecules studied; complete description of the procedures used to validate ...

Electron transfer plays a vital role in drug metabolism and underlying toxicity mechanisms. Currently, pharmaceutical research relies on pharmacokinetics (PK) and absorption, distribution, metabolism, elimination and toxicity (ADMET) measurements to understand and predict drug reactions in the body. Metabolic stability (and toxicity) prediction in the early phases of the drug discovery and development process is key in identifying a suitable lead compound for optimisation. Voltammetric methods have the potential to overcome the significant barrier of new drug failure rates, by giving insight into phase I metabolism events which can have a direct bearing on the stability and toxicity of the parent drug being dosed. Herein, we report for the first time a data-mining investigation into the voltammetric behaviour of reported drug molecules and their correlation with metabolic stability (indirectly measured via t½), as a potential predictor of drug stability/toxicity in vivo. We observed an inverse

Temperature is one of the primary factors affecting drug stability. The rate constant/ temperature relationship has traditionally been described by the Arrhenius equation,. where Ea is the activation energy and A is the frequency factor. This equation is a variant of the equation describing the effect of temperature on equilibrium processes that was developed by vant Hoff in 1887. Arrhenius applied his equation to various reaction processes.298 Comparison of the empirical Arrhenius equation to the Eyring equation, Eq. (2.7), shows some similarities and some differences. The frequency factor A in the Arrhenius equation corresponds to the product of the universal collision and entropy terms in Eq. (2.7) while the Ea term in Eq. (2.70) is related to the enthalpy term in Eq. (2.7).299 Because aplot of the logarithm of k against the reciprocal of absolute temperature generally yields a linear relationship (Arrhenius plots), the frequency factor and activation energy are regarded as independent of ...

There is an increasing demand for biomarkers in clinical treatment trials to demonstrate target engagement and to support disease modification claims. To be able to detect treatment related effects, a prerequisite is that the levels of the biomarker are stable over time or that the change over time is known. In the present study, the stability of alpha- and beta-cleaved soluble amyloid-beta protein precursor (sA beta PP alpha and sA beta PP beta), A beta(1-40) together with the phosphorylated form of neurofilament heavy/medium (pNfH/M) in cerebrospinal fluid (CSF) was analyzed in a cohort of 51 patients with Alzheimers disease. In addition, the stability of A beta(1-40), beta(1-42), and sA beta PP beta in plasma was explored. Plasma and CSF was sampled at baseline and after 6-months follow up, and all patients were on stable treatment with acetylcholinesterase inhibitors. During this 6-month longitudinal follow-up, we saw a small, but consistent and statistically significant increase in CSF ...

Since over 40 enzymes will be investigated for their mRNA abundance, processing, and degradation kinetics, the less tedious and more accurate Next Generation Sequencing of the entire mRNA repertoire of the cell is employed. To optimise the proportion of useful sequence, while including RNA fragments that are products of of degradation, rRNA is depleted using the eukaryotic Ribominus kit (Ambion). Two biological replicates are treated with Sinefungin and Actinomycin D to inhibit RNA processing and transcription, for different time periods. RNA is collected before treatment, 5 min after the addition of Sinefungin, 5 min after addition of ActD (+Sin), then 10 min after, 20, 30, 60 and 120 min. The samples are sequenced using the Illumina standard protocol, and the sequenced reads are aligned to the reference genome TREU927, and the number of reads aligned to each ORF are determined. ...

As prepared for delivery Governor Liebscher, distinguished guests, ladies and gentlemen, good morning to you all. It is a great pleasure for me to be here with you today, at the 33rd Economics Conference hosted by the Austrian National Bank-and, I must add, it is a delight to visit again this beautiful and historic city. I have been asked by the conference organizers to speak to the issue of financial sector stability and the IMFs work in this area-and I will seek to stay close to this topic in my remarks.. Financial sector stability has been described as the issue that central bankers worry about when they are not worrying about price stability. Experience regularly reminds us that there is a solid basis for this worry. The East Asian crisis during the late 1990s provided a striking example of how financial sector weaknesses can trigger economic turmoil and amplify the effects of adverse shocks on the economy, with severe economic and social consequences. Several industrial countries, ...

Fluorescent labelling of monoclonal antibodies (mAbs) is classically performed by chemical bioconjugation methods. The most frequent labelling technique to generate antibody-fluorophore conjugates (AFCs) involves the bioconjugation onto the mAb lysines of a dye bearing an N-hydroxysuccinimide ester or an isothiocyanate group. However, discrepancies between labelling experiments or kits can be observed, related to reproducibility issues, alteration of antigen binding, or mAb properties. The lack of information on labelling kits and the incomplete characterization of the obtained labelled mAbs largely contribute to these issues. In this work, we generated eight AFCs through either lysine or interchain cysteine cross-linking bioconjugation of green-emitting fluorophores (fluorescein or BODIPY) onto either trastuzumab or rituximab. This strategy allowed us to study the influence of fluorophore solubility, bioconjugation technology, and antibody nature on two known labelling procedures. The structures of

What Chirumbolo et al. are proposing is that a histamine molecule in water can cause the water molecules to arrange themselves around it in a kind of quasi-crystalline cluster -- a cluster having the same chemical and biological properties as the histamine itself. Further, the clusters are self-replicating: they can catalyse the formation of copies of themselves across multiple stages of dilution and succussion. This is a whole new polymorph of H2O, more stable at room temperature than liquid water ...
Storage: Lyophilized peptides although stable at room temperature for 3 months, should be stored desiccated below -18°C. Upon reconstitution of the peptide it should be stored at 4°C between 2-21 days and for future use below -18°C.. HGH Frag 176-191 is a fragment of the HGH peptide. Scientists found that if they truncated the peptide at the C terminal region they could isolate the fat loss attributes associated with HGH. Taking this fragment from HGH, including the peptide bonds from 176-191, they found they had developed a peptide that regulated fat loss 12.5 times better than regular HGH.. Introduction:. There is no question as to why bodybuilders of the modern era utilize human growth hormone. Its ability to regulate fat loss, pack on size and increase IGF-1 levels is just a few of the many attributes that many seek. HGH Frag 171-191 was developed so that bodybuilders and athletes not looking for the growth properties from HGH could still reap all the results of its amazing fat loss ...

The reaction is believed to proceed by the indicated conversion of a lithium carboxylate to the dilithium salt18 which is stable at room temperature in the absence of proton-donating solvents or reactants. However, the rapidity with which this dilithium salt is decomposed to a ketone in the presence of proton donors, accompanied by the rapidity of the subsequent reaction of the ketone with more methyllithium, can lead to a common side reaction in which an alkyldimethyl carbinol is formed. Both of the aforementioned reactions appear to be fast compared with 0.01 second usually required to disperse components with the conventional mixing techniques. This side reaction can be almost entirely avoided by taking precautions to minimize the possibility that high local concentrations of the geminal dialkoxide and the lithium reagent are present when a proton donor is added during either the reaction or the subsequent quenching. It is frequently possible to obtain only relatively small amounts of the ...

Although traditional 3D fluorescence spectroscopy is a powerful analytical technique, it is also notoriously slow, making it inappropriate for kinetic studies. Our paper presents a much faster way to collect 3D fluorescence measurements, using Hadamard barcodes to encode a white light source and the corresponding fluorescence signal. In this work, we used a submersible fiber-probe to measure both the degradation kinetics of spinach extract in acid, and the thermal degradation kinetics of rhodamine B.. Read More ...

About 10% of the study population showed excessive levels of perceived fall risk and were classified as anxious. Despite their low physiological fall risk, almost 40% of the anxious group experienced multiple or injurious falls during the one year follow-up. In keeping with previous studies, we anticipated that self imposed restriction on levels of physical activity would contribute to overall risk in this group.5 13 However, there was no indication of lower levels of activity, so other reasons need to be explored as to why anxious participants fell more than vigorous participants during follow-up.. It is possible that the anxious group had a residual physiological fall risk not encompassed by the physiological profile assessment. Thus, although the groups low physiological profile assessment score indicated that the integrity of individual sensorimotor components was adequate, their relatively poor performance on the coordinated stability test suggests that the integration of each of these ...

This paper studies the multidimensional stability of planar traveling waves for integrodifference equations. It is proved that for a Gaussian dispersal kernel, if the traveling wave is exponentially orbitally stable in one space dimension, then the corresponding planar wave is stable in $H^m(\mathbb{R}^N)$, $N\ge 4$, $m\ge [N/2]+1$, with the perturbation decaying at algebraic rate.

Previous studies have shown that equine SAA is more stable at room temperature than refrigerated. A small study at Cornell University confirmed this finding. Two equine samples with high values of 481 and 177 ug/ml showed a 2-3% decrease in concentration at 4ºC with storage for 24 hours versus a 3-5% decrease at room temperature. With storage for 72 hours, the drop at 4ºC was greater than at room temperature (7% versus 3-6% at 48 hours and 8-9% versus 2-4% at 72 hours). Very high values (,60 ug/dL) were still abnormal after 10 days of storage at either temperature (Hillstrom et al 2010). SAA concentrations are stable frozen (-20ºC), thus it is recommended that for storage ,24 hours, the samples should be frozen and shipped ensuring that they remain frozen. If freezing is not an option, then the samples should be maintained at room temperature. This also suggests that mildly increased concentrations may drop to within reference intervals (,20 ug/mL in horses) with storage. Stability is unknown ...

OPAT is used for treating a wide range of infections, including skin and soft tissue infections, osteoarticular infections, bacteraemia, endocarditis and complex intra-abdominal and urinary tract infections, even in presence of multiresistant microorganisms. Correct choice of antimicrobial agent and adequate patient selection are crucial for reaching therapeutic success and avoiding readmissions, treatment prolongation or treatment-related toxicity. The optimal antimicrobial for OPAT must be highly effective, have a long half-life and an adequate spectrum of action. Ceftriaxone and teicoplanin are currently the most prescribed antibiotics for OPAT, though daptomycin and ertapenem are also on the rise, due to their high efficiency, safety and wide spectrum of action. Antibiotics that are stable at room temperature can be administered through a continuous perfusion, though self-administration is preferable although it requires training of the patient or the caregiver. Thats a remarkable set of qualities for a simple string of carbon atoms, Yakobson said.. You could look at it as an ultimately thin graphene ribbon, reduced to just one atom, or an ultimately thin nanotube, he said. It could be useful for nanomechanical systems, in spintronic devices, as sensors, as strong and light materials for mechanical applications or for energy storage.. Regardless of the applications, he said, academically, its very exciting to know the strongest possible assembly of atoms.. Based on the calculations, he said carbyne might be the highest energy state for stable carbon. People usually look for what is ...
Prediction of shelf life is facilitated if the data include frequent intermediate stations (in line with the CPMP/ICH recommendations), are derived from several batches, consider a range of conditions, are of high precision, include analysis for breakdown products, and consider the physical properties of the formulation.. The Note for guidance on Evaluation of Stability Data (CPMP/ICH/420/02) provides detailed guidance on evaluation of the data from formal stability studies. The guideline presumes that the required amount of minimum data are available, as outlined under Section 9.4 Minimum data requirements.. For products intended for storage in a freezer no extrapolation beyond the period covered by the long term data is permitted (and testing of a single batch at an appropriate elevated temperature should also be conducted to address the effect of short term excursions outside the proposed label storage condition, such as during shipping or handling).. For other products, the assessment ...

Louis Roux from LifeAssay Diagnostics (Pty) Ltd. in South Africa, in collaboration with PATH, will develop an easy-to-use, low-cost, strip test for pregnant women in developing countries to detect the onset of proteinuria, which indicates a highly increased risk of preeclampsia/eclampsia. Preeclampsia/eclampsia is a major cause of maternal death, particularly in developing countries. Onset is rapid, and so early diagnosis is crucial, particularly in remote communities where health care is not immediately available. The test measures the ratio of protein to creatinine, which is more accurate than the current protein-only version, and they will produce a low-cost version that can be locally manufactured. They will evaluate the tests performance in comparison to three similar tools on a minimum of 1100 urine samples collected from the CAP trial in South Africa. The study also includes usability and stability evaluations to inform further prototype improvements and product insert development for ...

It is known that membrane technologies may have a major role to play in the development of environmental-friendly processes, either in gas or liquid separations. In liquid separations, one of the challenging issues is to get membranes with suitable separation properties able also to survive to the exposure of liquid solvents at various temperatures. Aromatic polyimides are known to exhibit a very good stability to a great number of solvents, but their permeability are low because of their rigid carbon skeleton and of their low available free volume; so their uses are mainly limited to gas separations or to filtration processes (MF, UF). To broaden the application area of polyimides in liquid separations, we have prepared and studied the properties of a block ether aromatic copolyimide series where the ether soft block acts both as a selective and a permeable block; we describe attempts to get asymmetric polyetherimide (PEI) membranes having molecular separation properties using various experimental

The durability of swings or climbing frames also depends on the materials used and their weather resistance. Play equipment with frames of highly durable woods such as oak or redwood is more expensive and less common at DIY markets. Discounters mostly offer standard equipment made of metal or so-called pressure-treated pine, which requires more maintenance.. Before purchasing their selection, parents should feel free to thoroughly inspect the equipment and consider carefully whether the equipment suits their purposes. Solid material and good stability should rate more highly than nice bright colours. Other important factors are:. · The equipment should be free from sharp edges or sharp ...
