We report on the development of azide-coronatine as a useful platform for azide alkyne cycloaddition ("click chemistry")-mediated synthesis of molecular probes. (+)-Azido-coronatine was synthesized in 10 steps with 11% yield using improved synthesis of coronafacic acid, in which the highly exo-selective Diels-Alder reaction (endo:exo > 1:25) is the key step. Azido coronatine was as effective as the original coronatine in a stomatal opening assay, and was easily modified to a fluorescein isothiocyanate (FITC)-labeled probe with high yield.  (+info)

Alternative approaches with lower activation barriers and copper-free reactions were proposed. In this paper, a facile and efficient approach combining distillation-precipitation polymerization (DPP) and click chemistry was developed to synthesize boronic acid ligand-modified magnetic nanoparticles for the enrichment of glycoproteins. The copper (I)-catalyzed alkyne azide 1,3-dipolar cycloaddition (CuAAC) or click reaction, is a highly versatile reaction that can be performed under a variety of reaction conditions including various solvents, a wide pH and temperature range, and using different copper sources, with or without additional ligands or reducing agents. This reaction is highly selective and can be performed in the presence of other functional moieties. The flexibility and selectivity has resulted in growing interest in the application of CuAAC in various fields. In this review, we briefly describe the importance of the structural folding of peptides and proteins and how the 1,4-disubstituted triazole product of the CuAAC reaction is a suitable isoster for an amide bond. However the major focus of the review is the application of this reaction to produce peptide conjugates for tagging and targeting purpose, linkers for multifunctional biomacromolecules, and reporter ions for peptide and protein analysis.

For all these applications mNPs surfaces must be tailored to improve biocompatibility properties or reduce aggregation but also to insert molecules on the particles with precise control of their architecture and surface density. So new methodologies for controlled and efficient functionalization of such materials are needed. In this contribution we will describe the use of various chemical approaches (microwave, click chemistry, photochemistry) for mNPs functionalization and present results of improve grafting rates (up to 40 fold increase). These are dPEG® compounds from Quanta Biodesign that contain discrete length PEGs. This can improve reproducibility in experiments when compared to using PEGs of variable length. PEGS have high aqueous solubility and a substances aqueous solubility can be improved by attaching a PEG. Some of these compounds contain a single reactive end for attachment to proteins such as through amines or sulfhydryls. This list includes branched PEGs which are more compact than long linear PEGs. There are also compounds with two reactive ends for crosslinking. Some of the compounds have azide groups which can be attached to alkynes using copper click chemistry or to strained alkynes through copper-free click chemistry. Our research projects rely on efficient chemistry. Organometallic and coordination complexes are synthesized in straightforward and modular fashions in order to study them in the biological context. Unfortunately, the clinical efficacy of these drugs is currently hindered by the development of undesirable side effects and resistance during treatment. The molecular mechanisms underlying these effects are still unclear. For decades, research has focused on DNA as the main cellular target of Pt(II) compounds. Glycopolymers consisting of styrene (St) and pentafluorostyrene (PFS) were synthesized by a combination of nitroxide-mediated polymerization and "click" chemistry. A series of well-defined homopolymers as well as block and random copolymers of St and PFS were obtained with different ratios by using Bloc Builder as an alkoxyamine initiator. Some copolymers showed self-assembly behavior into regular nanospheres with diameters ranging from 70 to 720 nm by applying the nanoprecipitation technique. In addition, a thiol−glycoside (2,3,4,6-tetra-O-acetyl-1-thio-β-d-glucopyranose) was reacted under ambient conditions with PFS moieties on the polymeric backbone utilizing a thiol−para fluoro "click" reaction. The therapeutic efficacy of antibodies can be successfully improved through targeted delivery of potent cytotoxic drugs in the form of antibody-drug conjugates. However, conventional conjugation strategies lead to heterogeneous conjugates with undefined stoichiometry and sites, even with considerable batch-to-batch variability. In this study, we have developed a chemo-enzymatic strategy by equipping the C-terminus of anti-CD20 ofatumumab with a click handle using Sortase A, followed by ligation of the payload based on a strain-promoted azide-alkyne cycloaddition to produce homogeneous conjugates. The resulting antibody-drug conjugates fully retained their antigen binding capability and proved to be internalized and trafficked to the lysosome, which released the payload with a favorable efficacy in vitro and in vivo. Coumarins were recently shown to constitute a novel class of mechanism-based carbonic anhydrase (CA, EC 4.2.1.1) inhibitors. We demonstrate that sulfocoumarins (1,2-benzoxathiine 2,2-dioxides) possess a similar mechanism of action, acting as effective CA inhibitors. The sulfocoumarins were hydrolyzed by the esterase CA activity to 2-hydroxyphenyl-vinylsulfonic acids, which thereafter bind to the enzyme in a region rarely occupied by other classes of inhibitors. The X-ray structure of one of these compounds in adduct with a modified CA II enzyme possessing two amino acid residues from the CA IX active site, allowed us to decipher the inhibition mechanism. The sulfonic acid was observed anchored to the zinc-coordinated water molecule, making favorable interactions with Thr200 and Pro201. Surface engineering provides a powerful tool to create model systems to investigate the complex interactions between cells and the extracellular matrix (ECM). Cell adhesive peptide ligands (e.g. Arginine-Glycine-Aspatate (RGD) sequence) and carbohydrates are abundant in the ECM, and both are involved in mediating cell adhesion. However, how these interact is not well understood in vivo. Model systems to investigate their collective roles are rare. We have developed a robust surface patterning method allowing spatial control of carbohydrate (i.e. mannose) and RGD on glass while overcoming non-specific biomolecular interactions.
