The 4-ethoxy group on the 2,4-pentadien-1-one was found to be effective to facilitate the cycloaddition with dienophiles. The predominance of oxidative stress in many tumour cell environments provides a means to selectively target these cells via protein oxidation. The zinc fingers of transcription factors utilise cysteine thiols for structural zinc coordination. Redox control of DNA binding regulates transcription and therefore the overall rates of proliferation, apoptosis and necrosis in the carcinoma. We report here the adverse effects of glutathione peroxidase (GPx) mimics towards zinc finger motifs and PC12 cell survival. Nanomolar catalyst concentrations facilitated H2O2-induced oxidation of an Sp1 transcription factor fragment. In PC12 cells GPx catalysis triggered a significant increase in cell death, correlating with severity of oxidative stress. As a consequence, we conclude that GPx mimics are potential chemotherapeutic agents.  (+info)

Synthesis and properties of benzo[a]phenoxazinium chalcogen analogues as novel broad-spectrum antimicrobial photosensitizers. (3/16)

The goal of this investigation was to develop improved photosensitizers for use as antimicrobial drugs in photodynamic therapy of localized infections. Replacement of the oxygen atom in 5-(ethylamino)-9-diethylaminobenzo[a]phenoxazinium chloride (1) with sulfur and selenium afforded thiazinium and selenazinium analogues 2 and 3, respectively. All three dyes are water soluble, lipophilic, and red light absorbers. The relative photodynamic activities of the chalcogen series were evaluated against a panel of prototypical pathogenic microorganisms: the Gram-positive Enterococcus faecalis, the Gram-negative Escherichia coli, and the fungus Candida albicans. Selenium dye 3 was highly effective as a broad-spectrum antimicrobial photosensitizer with fluences of 4-32 J/cm2 killing 2-5 more logs of all cell types than sulfur dye 2, which was slightly more effective than oxygen analogue 1. These data, taken with the findings of uptake and retention studies, suggest that the superior activity of selenium derivative 3 can be attributed to its much higher triplet quantum yield.  (+info)

The Group II-IV nitride semiconductors are emerging as promising alternatives to III-nitrides in ultraviolet LED applications. These materials have wurtzite-derived orthorhombic crystal structures and can be obtained by substituting pairs of Group III atoms in a III-nitride for a single Group II atom and a single Group IV atom. Here we will focus on MgSiN$_{2}$, which is the equivalent II-IV nitride to wurtzite AlN. A detailed comparison of the properties obtained by first principles calculations and experiment of these two systems will be performed. Interestingly, a recent study by Ohno et al. (2014) demonstrated that deletion of SNPH is counterproductive to axonal survival in acute demyelination. In the current study, we sought to identify proteins regulating mitochondrial docking in axons. By applying a proteomic approach and cell biological assays combined with time-lapse imaging in live neurons, we reveal that LC8 enhances the SNPH-mediated mitochondrial docking efficiency through modulating the dynamic interaction between the docking receptor and axonal cytoskeleton. Four lines of evidence support this view. First, SNPH selectively interacted with LC8 via its seven-residue LC8-binding motif (ERAIQTD). The SNPH-LC8 complex was detected in vivo by coimmunoprecipitation of brain homogenates from snph wild-type but not snph (−/−) mice; the interaction occurred independently of the dynein motor complex. Second, SNPH recruited LC8 to axonal mitochondria via its LC8-binding motif. Deleting this motif reduced the efficiency of SNPH in docking axonal mitochondria.
