Anti-proliferative effects of organic extracts from root bark of Juglans Regia L. (RBJR) on MDA-MB-231 human breast cancer cells: role of Bcl-2/Bax, caspases and Tp53. (2/2)

With increasing use of plant-based cancer chemotherapeutic agents, exploring the antiproliferative effects of phytochemicals has gained increasing momentum for anticancer drug design. The present study was undertaken to investigate the effect of root bark of Juglans regia (RBJR) organic extracts on cell proliferation, and to determine the molecular mechanism of RBJR-induced cell death by determining the expression of Bcl-2, Bax, caspases, Tp53, Mdm-2 and TNF-alpha in MDA-MB-231 human breast cancer cells. The results demonstrate that WNRB suppressed proliferation and induced apoptosis in a dose and time dependent manner by modulating expression of key genes. This involved characteristic changes in cytoplasmic and nuclear morphology, DNA fragmentation (TUNEL assay), levels of mRNA and expression of corresponding proteins. Real Time PCR and western blot analysis revealed that the expression of of Bax, caspases, tp53, and TNF-alpha was markedly increased in MBA-MB-231 cells treated with the RBJR extract. In contrast Bcl2 and mdm-2 expression was down regulated after exposure. In summary, our data suggest the presence of bioactive compound(s) in WNRB capable of killing breast carcinoma cells through induction of apoptosis, and therefore a candidate source of anticancer drugs.  (+info)

We think its an incredible, overlooked natural resource that is piling up on these farms, he says. We want to change the entire coffee industry to start looking at cascara as something just as valuable as coffee.. Including the cascara itself, the carbonated beverages are made using all organic ingredients, positioned as a healthy alternative to soda, and a more refreshing, less acidic alternative to kombucha. Gambhir, who had been an every-day coffee drinker, originally became interested in cascara a couple years ago when looking for ways to reduce his caffeine intake without eliminating it altogether. At 30mg per 12 ounce serving, his teas contain less than a third the caffeine found in an equivalent cup of coffee. Coffee as we know it developed in Yemen about 500 years ago, and as Gambhir began developing cascara tea recipes, he learned about qishr, a coffee fruit and ginger concoction Yemeni Sufis developed as a precursor to the hot beverage. That inspired him to add whole ginger root to ...

The Story behind Bowel Detox. Psyllium has been part of Indian Ayurvedic Medicine for nearly 5,000 years! It grows natively in India and parts of Asia and has been used to support gut health and to aid elimination. Ever since it was introduced to Spanish settlers in the 1600s, cascara sagrada bark has been used as a natural laxative throughout the world. Native American tribes shared this wonder with the Spaniards, and by the late 1800s, cascara was being exported overseas. The explosion of its use eventually led to overharvesting in the last century. But this resilient species of the buckthorn family has made a healthy resurgence. More than 2,000 years ago, the Phoenicians used activated charcoal to help purify their water. In fact, some sea voyagers stored their drinking water in charred barrels. Hippocrates, the father of medicine, recommended charcoal for vertigo and other conditions. Charcoal poultices and powders regained popularity in the 18th and 19th centuries ...

Over twenty-five years ago I learned something that really puzzled me. I learned that about 10% of the population react to nervines in a peculiar way. Herbs and essential oils that calm most people down are energizing to these people, while herbs and essential oils that are stimulating to most people, calm these people down. This bothered me, as the eclectic physicians claimed that one of the signs of a true remedy is a consistent action. For example, you dont see cascara sagrada causing constipation in some people. It always acts as a laxative.. About a decade later, I learned that the powerful stimulant, Chinese ephedra (which now is banned in the United States, unfortunately), would calm down children with ADHD. That kind of made sense, since Ritalin© is also a stimulant. But, the whole issue puzzled me. It was a few years later that I unraveled the secret. It had to do with nervous system balance. The remedies werent acting backward, its just that 10% of the population have an autonomic ...

Your notes are great examples for all the interested to hear of what we can do so easily. The predicament with the laxitives that are considered habit forming are that they replace or bypass healthy function so that with longterm use, the senna, for example, like others, will teach the body to relax its needed action. The cascara, contrary to that, stimulates the perastaltic action into a stronger function. We have used it for a period of years now and I see no negative, as long as dependance is not created and potassium loss is understood. . Great to hear of your success.
Rhamnus purshiana (cascara buckthorn, cascara, bearberry, and in the Chinook Jargon, chittem and chitticum; syn. Frangula purshiana, Rhamnus purshianus) is a species of buckthorn native to western North America from southern British Columbia south to central California, and eastward to northwestern Montana. The dried bark of cascara has been used for centuries as a laxative, first by Native Americans in the Pacific Northwest, and then by European/U.S. colonizers. The chemicals primarily responsible for the laxative action are the hydroxyanthracene glycosides (particularly cascarosides A, B, C and D), and emodin. These act as stimulant laxatives, with the hydroanthracene glycosides stimulating peristalsis, and emodin exciting smooth muscle cells in the large intestine. Cascara is a large shrub or small tree 4.5-10 m tall, with a trunk 20-50 cm in diameter. The outer bark is brownish to silver-grey with light splotching (often, in part, from lichens) and the inner surface of the bark is smooth and ...

The task of tracing visual language in book design is a challenge, but one that is useful to take up in order to help us understand how readers make meaning of what they read. Studying the lineage of some forms of layout also sheds light on how design fits into and contributes to culture in a wider sense. Childrens science books - the highly graphic and colourful ones published over the last 30 years or so in the UK - provide some excellent examples of complex visual language, partly because they tend to be more highly illustrated than books meant for adults. But how do diagrams, illustrations and different forms of text interact to produce content in these books? Four of them even possessed significantly stronger effects than DWMH, making the selected bioactive peptides useful for further research as new bioactive entities.
