Anti-proliferative effects of organic extracts from root bark of Juglans Regia L. (RBJR) on MDA-MB-231 human breast cancer cells: role of Bcl-2/Bax, caspases and Tp53. (2/2)

With increasing use of plant-based cancer chemotherapeutic agents, exploring the antiproliferative effects of phytochemicals has gained increasing momentum for anticancer drug design. The present study was undertaken to investigate the effect of root bark of Juglans regia (RBJR) organic extracts on cell proliferation, and to determine the molecular mechanism of RBJR-induced cell death by determining the expression of Bcl-2, Bax, caspases, Tp53, Mdm-2 and TNF-alpha in MDA-MB-231 human breast cancer cells. The results demonstrate that WNRB suppressed proliferation and induced apoptosis in a dose and time dependent manner by modulating expression of key genes. This involved characteristic changes in cytoplasmic and nuclear morphology, DNA fragmentation (TUNEL assay), levels of mRNA and expression of corresponding proteins. Real Time PCR and western blot analysis revealed that the expression of of Bax, caspases, tp53, and TNF-alpha was markedly increased in MBA-MB-231 cells treated with the RBJR extract. In contrast Bcl2 and mdm-2 expression was down regulated after exposure. In summary, our data suggest the presence of bioactive compound(s) in WNRB capable of killing breast carcinoma cells through induction of apoptosis, and therefore a candidate source of anticancer drugs.  (+info)

Over twenty-five years ago I learned something that really puzzled me. I learned that about 10% of the population react to nervines in a peculiar way. Herbs and essential oils that calm most people down are energizing to these people, while herbs and essential oils that are stimulating to most people, calm these people down. This bothered me, as the eclectic physicians claimed that one of the signs of a true remedy is a consistent action. For example, you dont see cascara sagrada causing constipation in some people. It always acts as a laxative.. About a decade later, I learned that the powerful stimulant, Chinese ephedra (which now is banned in the United States, unfortunately), would calm down children with ADHD. That kind of made sense, since Ritalin© is also a stimulant. But, the whole issue puzzled me. It was a few years later that I unraveled the secret. It had to do with nervous system balance. The remedies werent acting backward, its just that 10% of the population have an autonomic ...

Rhamnus purshiana (cascara buckthorn, cascara, bearberry, and in the Chinook Jargon, chittem and chitticum; syn. Frangula purshiana, Rhamnus purshianus) is a species of buckthorn native to western North America from southern British Columbia south to central California, and eastward to northwestern Montana. The dried bark of cascara has been used for centuries as a laxative, first by Native Americans in the Pacific Northwest, and then by European/U.S. colonizers. The chemicals primarily responsible for the laxative action are the hydroxyanthracene glycosides (particularly cascarosides A, B, C and D), and emodin. These act as stimulant laxatives, with the hydroanthracene glycosides stimulating peristalsis, and emodin exciting smooth muscle cells in the large intestine. Cascara is a large shrub or small tree 4.5-10 m tall, with a trunk 20-50 cm in diameter. The outer bark is brownish to silver-grey with light splotching (often, in part, from lichens) and the inner surface of the bark is smooth and ...

The task of tracing visual language in book design is a challenge, but one that is useful to take up in order to help us understand how readers make meaning of what they read. Studying the lineage of some forms of layout also sheds light on how design fits into and contributes to culture in a wider sense. Childrens science books - the highly graphic and colourful ones published over the last 30 years or so in the UK - provide some excellent examples of complex visual language, partly because they tend to be more highly illustrated than books meant for adults. But how do diagrams, illustrations and different forms of text interact to produce content in these books? Meaning does not reside in the book alone, but is dependent on context: the particular conditions of reading, and the wider social and cultural environment. This is an exploration of book design as a medium of communication.. Many changes have occurred in British childrens publishing over the past 30 years. These have had a visible ...

HERBS -- Therapeutic use. Abstract:This study aims to present the in vitro inhibitory effect of Psidium guajava and Juglans regia leaf extracts on the main developer of acne lesions, Propionibacterium acnes (P. acnes), and other organisms that are isolated from acne lesions. Thirty-eight subjects (males and females) who had various types of acne were enrolled in the study. The contents of the acne lesions were cultured and the frequency of P. acnes (alone and with Staphylococci spp.) was 47%, whereas the frequencies for Staphylococcus aureus and Staphylococcus epidermidis were 13% and 24%, respectively. The antimicrobial activities of Psidium guajava and Juglans regia leaf extracts, determined by disk diffusion method (zone of inhibition), were compared to tea tree oil (TTO), doxycycline and clindamycin antibiotics. The zones of inhibition due to the Psidium guajava and Juglans regia leaf extracts ranged from 15.8-17.6 mm against P. acnes, 11.3-15.7 mm against S. aureus and 12.9-15.5 mm against ...
If the pollen from the male flower settles on the female flower at the point that they are receptive, fertilisation is likely to occur and the female flower will go on to develop into nuts. The time of pollen shedding from the male flower does not always overlap well with the time of female flower receptivity to pollen. This condition is referred to as dichogamy. To overcome this problem growers can select another walnut cultivar (a pollinator) the male flowers of which open at the same time as the female flowers from the main cultivar. The pollinator should be situated upwind from the main crop. If you have other walnuts upwind from your site you should not have problems with this ...

In this study, morphological characteristics as well as ten of Simple Sequence Repeat (SSRs) loci were used to analyze the genetic diversity and relationships among 12 natural populations of Persian walnut (Juglans regia L.) in northern and western regions of Iran. The results showed that there was a high level of genetic diversity among the walnuts, both in terms of their SSRs loci as well as morphological traits. The nut weight ranged from 11.5 to 17.2 g, kernel weight from 3.2 to 6.3 g, and kernel percentage from 28 to 46.7%. In SSRs analysis, the number of alleles per locus ranged from 6 to 11, with a total of 83 alleles and average of 8.3 alleles and 4.9 effective alleles per locus. The expected heterozygosity (He) varied between 0.70 and 0.87, with an average of 0.79 per locus. The proportion of genetic differences among the walnut populations accounted for 19% of the total variation. The overall gene flow among populations equaled 1.10. The 12 walnut populations were separated into four main
