Relative biological effectiveness of fission neutrons for producing micronuclei in the root-tip cells of onion seedlings after irradiation as dry seeds. (1/14)

The relative biological effectiveness (RBE) of mixed neutron and gamma-ray radiation emitted at a 252Cf source at the Research Institute for Radiation Biology and Medicine, Hiroshima University, compared with 60Co gamma-ray radiation was determined. The tissue-absorbed dose contribution of the accompanying gamma radiation was about 35.7% to the total tissue-absorbed dose from the 252Cf mixed radiation. The 252Cf mixed radiation and 60Co gamma rays produced approximate linear changes in the frequency of micronuclei induced in root-tip cells of Allium cepa L. onion seedlings after irradiation as dry dormant seeds with varying absorbed doses in onion seeds. Therefore, the RBE for radiation-induced micronuclei was calculated as the ratio of the slopes for the 252Cf mixed radiation and the 60Co gamma rays. The deduced RBE value of 252Cf mixed radiation to 60Co gamma rays to induce micronuclei in dry dormant onion seed cells was about 90.5 +/- 3.6 (+/- 1sigma); the RBE of neutrons from the 252Cf mixed radiation was about 150 +/- 6 (+/- 1sigma). Furthermore, the sensitivity ratio of the induction rate of micronuclei in dry dormant seeds to that in seedlings by neutrons from 252Cf mixed radiation was significantly different from that by 60Co gamma rays. From these results, we concluded that the repair efficiency of DNA damage induced by neutrons may be different from that by gamma rays.  (+info)

Carcinogenic effect of in utero 252Cf and 60Co irradiation in C57BL/6N x C3H/He F1 (B6C3F1) mice. (2/14)

C57BL/6N x C3H/He F1 mice were exposed in utero to 0, 1.0 and 2.7 Gy of 252Cf or 60Co at day 16.5th of gestation. Mice of both sexes were observed for 2 years. The females in the irradiated groups showed increases in the incidences of pituitary, mammary gland, liver and lung tumors. 252Cf was more effective in inducing tumors than was 60Co. Interestingly, the incidence of hematopoietic tumors decreased by irradiations with 252Cf but not with 60Co. The incidences of liver tumors in males increased by 252Cf-irradiation, whereas, the incidences of skin and soft tissue tumors increased by 60Co-irradiation. These results indicate that irradiation in utero during the late embryonic stage can induce tumors postnatally after a long latency. Moreover, females irradiated in utero had disfunction of the ovaries, evidence of impairment of the female's specific hormonal environment. This may be the cause of the low incidence of ovarian tumors and the high incidences of liver, lung and pituitary tumors in these female mice. Females with pituitary tumors had a high serum prolactin, which might be responsible for the concurrence of mammary gland tumors. These results indicate the importance of host factors in the development of radiation-induced tumors.  (+info)

252Cf relative biological effectiveness and inheritable effect of fission neutrons in mouse liver tumorigenesis. (3/14)

To determine the relative biological effectiveness of 252Cf fission neutrons versus 60Co gamma-rays for inducing liver tumorigenesis, C57BL/6NCrj x C3H/NCrj F1, hereafter called B6C3F1, mice were irradiated once either with 252Cf or 60Co and the tumorigenicity of the two types of radiation was studied. Individual groups of B6C3F1 mice (about 30 mice/group) were irradiated once with 252Cf at doses of 0, 3, 6, 12.5, 50, and 200 cGy for males and 0, 12.5, 50, and 200 cGy for females or with 60Co at doses of 0, 12.5, 50, and 200 cGy for both sexes. The groups were observed for 13 months after irradiation. The incidence of hepatic tumors in nonirradiated controls was 3.8% in males and 3.2% in females. 252Cf irradiation increased the incidence dose dependently in males and females, but less efficiently in females. The mean number and size of hepatic tumors were well correlated with tumor incidence. In contrast, 60Co was less efficient at inducing tumors. The relative biological effectiveness of 252Cf compared to 60Co was quite high in the low-dose range. Overall, the average relative biological effectiveness of 252Cf versus 60Co was 15.2 in males and 2.5 in females. In another experiment, 7-week-old male C3H mice, which are known to be carriers of hepatic tumorinogens, and that were the fathers of the B6C3F1 mice, were irradiated with 0, 50, and 200 cGy of 252Cf, and 2 weeks later they were mated with nonirradiated female C57BL mice. The control mice had nonirradiated fathers. The incidence of hepatic tumors in the offspring of irradiated fathers was 43% in males and 1.7% in females, whereas in the offspring of control mice with nonirradiated fathers the incidence was 3.2% in males and 3.3% in females. The multiplicity of the hepatic tumors was similar to the tumor incidence. Thus our results indicated a strong hepatocarcinogenic potential for 252Cf neutrons compared to 60Co gamma-rays, and that the cancer-prone genetic trait activated by 252Cf irradiation was inherited by first-generation offspring, especially in males.  (+info)

Analysis of unmodified endotoxin preparations by 252Cf plasma desorption mass spectrometry. Determination of molecular masses of the constituent native lipopolysaccharides. (4/14)

Nine unmodified endotoxin preparations constituted of Re-, Rd-, and Rc-type lipopolysaccharides (2 to 5 glycoses), representing four species of enterobacteria were analyzed by 252Cf plasma desorption mass spectrometry. The constituent lipopolysaccharides were characterized by the ion pair: (M-H)- and its corresponding lipid fragment ion. The lipid fragment ion is produced by cleavage of the glycosidic bond of the 3-deoxy-D-manno-oct-2-ulosonic acid unit that substitutes O-6' of the glucosamin beta 1'-6glucosamine ("lipid A backbone") disaccharide of the lipid A moiety. These lipid fragment ions were identical to the (M-H)- ions seen in the spectra of homologous isolated lipid A preparations that were obtained by hydrolysis (pH 4.5, 100 degrees C) promoted by sodium dodecyl sulfate. Since the molecular components present in the endotoxin preparations analyzed are known, the ion pair (M-H)(-)-lipid fragment ion defines the molecular compositions of each individual lipopolysaccharide. Heterogeneity of the R-type endotoxin preparations analyzed was due almost exclusively to differing lipid A moieties. In three Salmonella minnesota 595 Re endotoxin preparations 10 different lipopolysaccharides were identified, only two of which were common to all three preparations. Of the nine lipopolysaccharides identified in two S. minnesota R7 endotoxin preparations, only two were present in both.  (+info)

Structure of a streptococcal adhesin carbohydrate receptor. (6/14)

Interactions between complementary protein and carbohydrate structures on different genera of human oral bacteria have been implicated in the formation of dental plaque. The carbohydrate receptor on Streptococcus sanguis H1 (one of the primary colonizing species) that is specific for the adhesin on Capnocytophaga ochracea ATCC 33596 (a secondary colonizer) has been isolated from the streptococcal cell wall, purified, and structurally characterized. The hexasaccharide repeating unit of the polysaccharide was purified by reverse-phase, amino-bonded silica, and gel permeation high performance liquid chromatography. Earlier studies established that the repeating unit was a hexasaccharide composed of rhamnose, galactose, and glucose in the ration of 2:3:1, respectively. In the present study, determination of absolute configuration by gas chromatography of the trimethylsilyl (+)-2-butyl glycosides revealed that the rhamnose residues were of the L configuration while the hexoses were all D. 252Californium plasma desorption mass spectrometry of the native, the acetylated and the reduced and acetylated hexasaccharide determined that the molecular mass of the native hexasaccharide was 959, and that the 2 rhamnose residues were linked to each other at the nonreducing terminus of the linear molecule. Methylation analysis revealed the positions of the glycosidic linkages in the hexasaccharide and showed that a galactose residue was present at the reducing end. The structural characterization of the hexasaccharide was completed by one and two dimensional 1H and 13C NMR spectroscopy. Complete 1H and 13C assignments for each glycosyl residue were established by two-dimensional (1H,1H) correlation spectroscopy, homonuclear Hartmann-Hahn, and (13C,1H) correlation experiments. The configurations of the glycosidic linkages were inferred from the chemical shifts and coupling constants of the anomeric 1H and 13C resonances. The sequence of the glycosyl residues was determined by a heteronuclear multiple bond correlation experiment. These data show that the structure of the hexasaccharide repeating unit derived from the cell wall polysaccharide of S. sanguis H1 is: alpha-L-Rhap-(1----2)-alpha-L-Rhap-(1----3)-alpha-D-Galp- (1----3)-beta-D-Galp-(1----4)-beta-D-Glcp-(1----3)-alpha/beta-D-Gal.  (+info)

Teratogenic effect of californium-252 irradiation in rats. (7/14)

The teratogenicity of californium-252 (Cf-252) irradiation which generates approximately 70% 2.3 MeV fast neutron and 30% gamma rays was evaluated. A single whole body exposure of Cf-252 at various doses was given to pregnant rats on day 8 or 9 of pregnancy, followed by microscopic autopsy of the fetuses at the terminal stage of pregnancy to search for external and internal malformations. For comparison, pregnant rats were irradiated with various doses of cobalt-60 (Co-60) standard gamma rays at the same dose rate (1 rad/min.). The doses were 20-120 rad of Cf-252 and 80-220 rad of Co-60. Using frequency of radiation induced malformations observed on day 8 of pregnancy as an index, relative biological effectiveness (RBE) of 2.3-2.7 was obtained from the straight line obtained by modifying by the least squares method the frequency curves of malformed fetuses in total implants and in surviving fetuses. The types of malformations induced by Cf-252 and Co-60 irradiation were alike. Using fetal LD50 as an index, 2.4 was obtained as RBE when irradiated on day 8 of pregnancy and 3.1 as that when irradiated on day 9. The results showed that Cf-252 had stronger a teratogenic effect than Co-60 gamma rays.  (+info)

Identification of N-glycolylneuraminic acid-containing gangliosides of cat and sheep erythrocytes. 252Cf fission fragment ionization mass spectrometry in the analysis of glycosphingolipids. (8/14)

A number of gangliosides were isolated from cat and sheep erythrocytes for use in analyzing the specificity of a panel of human anti-heterophile monoclonal antibodies. The structures of these compounds were determined by a combination of different procedures, including sugar analysis, glycosidase treatment, periodate oxidation, TLC immunostaining, methylation analysis, and mass spectrometry. These methods identified the cat erythrocytes gangliosides (C1 and C2) as N-glycolylneuraminic acid (NeuGc)-containing hematosides; C1 was shown to be NeuGc alpha 2----8NeuGc alpha 2----3Gal beta I----4Glc-Cer [NeuGc)2GD3) and C2 to be NeuAc alpha 2----8NeuGc alpha 2----3Gal beta 1----4Glc-Cer [NeuAc-NeuGc-)GD3). The two sheep gangliosides (S1 and S2) were found to be novel glycolipids based on the paragloboside sequence; S1 was identified as NeuGc alpha 2----8NeuGc alpha 2----3Gal beta 1----4GlcNAc beta 1----3Gal beta 1----4Glc-Cer [NeuGc)2-disialylparagloboside) and S2 as NeuAc alpha 2----8NeuGc alpha 2----3Gal beta 1----4GlcNAc beta 1----3Gal beta 1----4Glc-Cer [NeuAc-NeuGc-)-disialylparagloboside). Structural analysis of these compounds was aided by the use of 252Cf fission fragment ionization time-of-flight mass spectrometry. This method provided easily interpretable spectra on methylated derivatives which were particularly useful in determining the sialic acid composition of the gangliosides and the sequence of their disialosyl side chains.  (+info)

All californium isotopes are radioactive; the long-lived isotopes are produced from berkelium-249 or from californium-249. They are californium-249 (351-year half-life), californium-250 (13-year half-life), californium-251 (898-year half-life), and californium-252 (2.645-year half-life). The isotope californium-249 has been used in tracer levels and microgram amounts to investigate the chemistry of californium (which exhibits an oxidation state of +3 in aqueous solution) and for preparing microgram quantities of compounds such as the oxychloride CfOCl, the oxides Cf2O3 and CfO2, and the trichloride CfCl3. The dihalides CfCl2, CfBr2, and CfI2 have been made. There is also some evidence from electrochemical studies in solutions for a +2 state. Metallic californium has been prepared and is structurally similar to metallic americium, curium, berkelium, and most lanthanoid metals. ...
Sepsis and septic shock result from an exacerbated systemic inflammatory reaction to infection. Their incidence is rising, and they have recently become the main cause of death in intensive care units. Septic shock is defined as sepsis accompanied by life-threatening refractory hypotension, for which excessive nitric oxide (NO), produced by inducible NO synthase iNOS, is thought responsible. LPS, a vital outer membrane component of Gram-negative bacteria, mimics most of the septic effects and is widely used as a model for septic shock. TLR4 is the signal-transducing receptor for LPS, evidenced by the resistance of TLR4-deficient C3H/HeJ and C57BL/10ScNJ mice. As expected, we found that TLR4 deficiency precludes LPS-induced cytokine production, independent of the purity of the LPS preparation. However, various conventional LPS preparations induced NO in TLR4-deficient mice to the same level as in control animals, while ultrapure LPS did not, indicating the presence of NO-producing contaminant(s). ...
One microgram of 252-Cf emits 2.314 × 10^6 fast neutrons/s with a 2.645-year half-life. Typical costs for loan and return of a ,7μg source total ~$11,000. (This fee is waived under the University Loan Program for university research and teaching applications.) Similar costs for a source in the range of 7μg to 3 mg (neutron intensities ranging up to 7 × 10^9/s ) total ~$20,000, while sources in the 3- to 5-mg range total ~$28,000. Loan/return costs of pre-existing sources from inventory containing ,5 mg total ~$32,000. Sources containing ,8 mg typically require custom fabrication, but a source containing the maximum permitted 252-Cf content of 50 mg (neutron intensity ~10^11/s) can be obtained for ~$51,000. None of these costs include transportation charges. Sources with neutron intensities ,10^8/s can be obtained at lower cost from commercial vendors, but potential costs for end-of-use source disposal should be evaluated. Loan costs for high-intensity sources compare very favorably with ...

In the past, scientists used a stationary neutron source to calibrate the reactor and to make sure it was accurately measuring emitted neutrons, but that doesnt recreate how neutrons actually bounce around. Tossing a lump of californium on the moving train improved the accuracy 10-fold, according to the scientists.. Researchers at Princeton used toy trains for calibration decades ago in an older reactor, but anytime scientists build radioactive trains, people tend to pay attention.. An no, the train didnt glow bright green or gain super powers-californium is only slightly radioactive, after all, and the toy train is now chugging around the Christmas tree in the labs lobby.. These particular particles pass through most measuring systems undetected, but the FNS uses a gate and capture technique that slows down neutrons and captures them in special glass fibers loaded with lithium. That process produces a unique flash of light, which custom electronics in the FNS recognize and analyze to determine radiation level. This technology is less susceptible to false triggers from other forms of radiation and can significantly improve reliable identification of neutrons in the mixed radiation field found in deep space. This improved measurement will help protect crews on future exploration missions ...
These conjugates can have the carbohydrate moiety linked directly to the oligonucleotide or can be linked to a lipid or a peptide/protein. Due to the diversity of carbohydrate receptors present on the cells surface, oligosaccharides are useful for targeting conjugates to specific cells. Conjugation of antisense ODNs to a neoglycoprotein in which the protein has several mannose-6-phosphate residues, is readily internalized by cells having a receptor for this ligand on their surfaces. ...
Two assumptions are commonly made in the estimation of genetic risk: (1) that the seven specific loci in the mouse constitute a suitable basis for extrapolation to genetic disease in humans, and (2) that mutations are induced by radiation damage (energy-loss events leading to double-stranded damage) occurring within the gene and are induced linearly with dose, at least at low doses. Recent evidence on the mutability of repeat sequences is reviewed that suggests that neither of these assumptions is as well founded as we like to think. Repeat sequences are common in the human genome, and alterations in them may have health consequences. Many of them are unstable, both spontaneously and after irradiation. The fact that changes in DNA repeat sequences can clearly arise as a result of radiation damage outside the sequence concerned and the likely involvement of some sort of signal transduction process mean that the nature of the radiation dose response cannot be assumed. While the time has not come ...
The stability of the HIV-1 core in the cytoplasm is crucial for productive HIV-1 infection. Mutations that stabilize or destabilize the core showed defects in HIV-1 reverse transcription and infection. We developed a novel and simple assay to measure stability of in vitro-assembled HIV-1 CA-NC complexes. This assay allowed us to demonstrate that cytosolic extracts strongly stabilize the HIV-1 core (Fricke et al., J Virol 87:10587-10597, 2013). By using our novel assay, one can measure the ability of different drugs to modulate the stability of in vitro-assembled HIV-1 CA-NC complexes, such as PF74, CAP-1, IXN-053, cyclosporine A, Bi2, and the peptide CAI. We also found that purified CPSF6 (1-321) protein stabilizes in vitro-assembled HIV-1 CA-NC complexes (Fricke et al., J Virol 87:10587-10597, 2013). Here we describe in detail the use of this capsid stability assay. We believe that our assay can be a powerful tool to assess HIV-1 capsid stability in vitro. ...
In the course of this review, my usual carry Glock 19 went off line and needed some small parts replaced. While awaiting replacement, I swapped to my Glock 17. Lo and behold I found myself comfortably carrying the larger gun without much difficulty, and have done so for the last few weeks.. I have successfully concealed the gun in this position from my office staff, patients and hoplophobic wife. Hugs can occur-so long as I am offset a little to the right. The only time I was ever made was when one of the kids, whose head is about waist high, ran to sneak attack me with a hug and rang his bell on the chunk of metal on the waistband. God knows what he made of that . . .. AIWB carry has another, far more significant downside: the possibility of shooting oneself during a transition.. While shooting oneself always sucks, there are places where bullet wounds are eminently survivable (e.g., the buttocks). The femoral artery is not one of them. By the same token, you really need to pay attention when ...

The even-even N=90 isotones with Z=60-66 are known to undergo a first order phase transition. Such a phase transition in atomic nuclei is characterized by a sudden change of the shape of the nucleus due to changes in the location of the potential minimum. In these proceedings we report a measurement of the B4/2 ratio of 148Ce, which will probe the location of the low-Z boundary of the N=90 phase transitional region. The measured B4/2 value is compared to the prediction from the X(5) symmetry within the interacting boson model at the critical point between the geometrical limits of vibrators and rigid/axial rotors. Carbohydrate - receptor interactions are an integral part of biological events. They play an important role in many cellular processes, such as cell-cell adhesion, cell differentiation and in-cell signaling. Carbohydrates can interact with a receptor by using several types of intermolecular interactions. One of the most important is the interaction of a carbohydrates apolar part with aromatic amino acid residues, known as dispersion interaction or CH/π interaction. In the study presented here, we attempted for the first time to quantify how the CH/π interaction contributes to a more general carbohydrate - protein interaction. We used a combined experimental approach, creating single and double point mutants with high level computational methods, and applied both to Ralstonia solanacearum (RSL) lectin complexes with α-l-Me-fucoside. Experimentally measured binding affinities were compared with computed carbohydrate-aromatic amino acid residue interaction energies. Experimental binding affinities for

Our interest in the title compound, C36H27N3O9, arises from its use as an important precursor in the synthesis of artificial receptors based on trimethoxybenzene-derived core (for a review on acyclic carbohydrate receptors containing a trimethyl- or triethylbenzene scaffold, see: Mazik, 2009). The title compound crystallizes in the space group P-1 with one molecule in the asymmetric part of the unit cell. The interplanar angles between the phthalimido residues are 6.67 (4) (B/C), 48.86 (3) (B/D) and 53.23 (3)° (C/D). According to the three-dimensional arrangement of substituents around the benzene ring the present conformational isomer can be named 1-up, 3,5-down tris(phthalimidomethyl), 2,4-up, 6-down- trimethoxybenzene. The molecular conformation is stabilized by seven C-H···O hydrogen bonds (Desiraju, 2002; Desiraju & Steiner, 1999; Mazik et al., 2001; Steiner, 2002) with phthalimido O atoms O(4), O(7), O(8), O(9) and the ether O atoms O(2), O(3) acting as acceptors [C-H···Ophthal 2.38 ...

This invention relates to a method of forming a selectively perforate membrane suitable for high resolution screen printing by homogeneously irradiating a sheet of synthetic resin, e.g., a polycarbonate resin, with heavy fission fragments whereupon the sheet is selectively irradiated in an oxygen atmosphere with electromagnetic radiation less than 4,000A (or a high intensity electron beam) to enhance the etch rate of those damage tracks lying within the electromagnetically irradiated area of the sheet. Apertures less than 200A in diameter then are etched through the sheet in areas of dual irradiation whereupon etching is abruptly terminated and the sheet is annealed at a temperature above 80 DEG C to obliterate fission track damage. The selectively apertured sheet then is etched again to enlarge the apertures to a diameter between 1,000A and 100,000A.
By Reta McDermott. Christopher Brav from the Mathematical Institute at the University of Oxford will present Ping-Pong and Thin Monodromy at 2:30 p.m. today in 122 Cardwell Hall. Abstract: Monodromy groups of low order hypergeometric differential equations have been well understood since the 19th century, with the examples arising in algebraic geometry typically being arithmetic. While some general results about monodromy groups of higher order equations have been known for over 20 years, many basic questions remain open.. Brav will discuss joint work with Hugh Thomas that gives the first examples of nonarithmetic or thin monodromy groups of higher rank arising in algebraic geometry. More precisely, Brav and Thomas use the ping-pong lemma of Fricke and Klein to show that the monodromy groups of certain one-parameter families of Calabi-Yau manifolds split as free products and then apply a cohomological criterion to deduce that the groups are not arithmetic.. ...

TY - GEN. T1 - APPLICATIONS OF LASER DESORPTION MASS SPECTROMETRY.. AU - Cotter, Robert J.. AU - Tabet, Jean Claude. AU - van Breemen, Richard. PY - 1984. Y1 - 1984. N2 - The laser desorption technique can be used for the mass spectral analysis of nonvolatile/thermally labile compounds such as peptides, oligosaccharides, glycolipids, etc. In our laboratory a laser desorption time-of-flight mass spectrometer has been used for structural confirmation of several glucuronic acid conjugates, synthesized by an immobilized-enzyme mediated technique. Laser desorption spectra of several small peptides produce both C-terminal and N-terminal fragments. The molecular ions are formed by cationization (alkali ion attachment) and the alakali ion is generally found on the C-terminal fragments, suggesting a ionization/fragmentation scheme similar to that observed by chemical ionization.. Biophysical models are the analytical tools by which the effects of radiation may be interpreted, allowing assessment and prediction of the effects of low dose radiation. Many developments have taken place in modelling since the 1980s. This volume is a review of the current state of radiation effect modelling, as discussed by scientists at an international workshop held in Padua, Italy in September 1991. Research on existing models and data sets used to test different models are described in detail. The applications of models to radiation biology and radiological protection are reviewed, as are recent developments in the modelling of the carcinogenic process. The interaction of mixed radiation, trace theory and the effects of radiation on DNA are also considered. The book should be of interest to radiation biophysicsts and biologists, medical physicists and others involved in radiological therapy and protection.

Steve Jobs would have loved this band. Capsulas songs are perfect for any Apple product rollout: a quick burst, exhilarating and different - but not too different. Capsula sounds like a lot of other things youve heard by now, but in a way thats hard to pin down. Not unlike Mr. Jobss revolution, Capsula is manufactured from a lot of parts made by other bands, in this case such high-energy acts as the Strokes or the Hives or the Vines, or the Who or Sonic Youth. What we try to do is pick different sounds from different periods of time that we like, Coni Duchess says by phone from somewhere on the road between Nashville and Atlanta. We dont belong to any particular scene.. But the guitar in Capsula is superior, I think, considering that most American indie-rockers rarely solo. Voices Underground, by the way, finishes with a nice quote from Cheap Tricks Shes Tight. I can see why Rolling Stone critic David Fricke jumped on the Capsula bandwagon so early: because they sound a lot like ...

Controllers may have some splainin to do. It will be interesting to see if this circle-to-land was therefore foisted on this crew, although I didnt hear them discussing the alternative, and their ATC communications certainly seemed normal, even a little punchy. As Juan Brown points out, the right wing low just before impact when the aircraft had been in a left turn suggests a stall of the right wing while they were trying to maneuver at approach speed. These guys got bit hard, and, again, it sounds a bit like the Lear at Teterboro, where rather than go-around when the approach became unstable, after botching the circle-to-land such that the aircraft couldnt be aligned with the runway, the Captain instead tried to muscle it in without adequate airspeed for the manoevre, stalled, and crashed about 1 km from the runway ...
