Novel screening method for potential skin-whitening compounds by a luciferase reporter assay. (2/19)

Measurement of the melanin content by using B16 melanoma cells is generally applied to find novel skin-whitening agents. However, this measurement method using B16 melanoma cells has such disadvantages, as the time taken, its sensitivity, and troublesomeness. We therefore attempted in the present study to establish a reporter assay system by measuring the tyrosinase promoter activity to use for convenient, high-throughput screening of new melanogenesis inhibitors. We first confirmed the validity of this reporter assay system by using such known skin-whitening agents, as arbutin, sulforaphane, and theaflavin 3,3'-digallate. We then compared the effect of 56 compounds on the tyrosinase promoter activity to test this reporter assay system. Carnosol, and rottlerin strongly inhibited the tyrosinase promoter activity. Moreover, carnosol and rottlerin decreased melanin synthesis and tyrosinase expression in a dose-dependent manner when using B16 melanoma cells. These results indicate this new luciferase reported assay system to be an effective and convenient method for screening potential skin-whitening compounds.  (+info)

Optimization of xylanase production from Aspergillus niger for biobleaching of eucalyptus pulp. (4/19)

A crude endo-xylanase produced by Aspergillus niger BCC14405 was investigated for its potential in pre-bleaching of chemical pulp from eucalyptus. The optimal fermentation conditions on the basis of optimization using response surface methodology included cultivation in a complex medium comprising wheat bran, rice bran, and soybean meal supplemented with yeast extract, glucose, peptone, and lactose with a starting pH of 6.0 for 7 d. This resulted in production of 89.5 IU/mL of xylanase with minor cellulase activity. Proteomic analysis using LC/MS/MS revealed that the crude enzyme was a composite of hemicellulolytic enzymes, including endo-beta-1,4-xylanase and other hemicellulolytic enzymes attacking arabinoxylan and mannan. Pretreatment of the pulp at a xylanase dosage of 10 IU/g increased the brightness ceiling after the C-Eop-H bleaching step up to 3.0% using a chlorine charge with a C-factor of 0.16-0.20. Xylanase treatment also led to reduction in chlorine charge of at least 20%, with an acceptable brightness level. The enzyme pretreatment resulted in a slight increase in pulp viscosity, suggesting an increase in relative cellulose content. The crude enzyme was potent in the enzyme-aided bleaching of chemical pulp in an environmentally friendly pulping process.  (+info)

Inhibitory effect of [6]-gingerol on melanogenesis in B16F10 melanoma cells and a possible mechanism of action. (5/19)

[6]-Gingerol is an active component of ginger that shows antipyretic and anti-inflammation activities. To find a novel skin-whitening agent, the melanogeneis inhibitory effects and action mechanisms of [6]-gingerol were investigated. In the present study, the effects of [6]-gingerol on mushroom tyrosinase, tyrosinase activity, and melanin content were determined spectrophotometrically, and the expression of tyrosinase and related proteins in B16F10 murine melanoma cells was evaluated by Western blotting. Furthermore, a possible signaling pathway involved in [6]-gingerolmediated depigmentation was investigated by means of specific inhibitors. The results revealed that [6]-gingerol (25-100 microM) effectively suppresses murine tyrosinase activity and decreases the amount of melanin in a dose-dependent manner. Additionally, it also effectively decreased the intracellular reactive oxygen species (ROS) level in a dose-dependent pattern in the same dose range. Our results indicate that [6]-gingerol inhibits melanogenesis of B16F10 melanoma and can function as a good skinwhitening agent.  (+info)

The prevalence of occupational asthma among denim bleachery workers in Kayseri. (7/19)

The bleachery procedure is the most frequent method used to decolorize denims since sandblasting has been shown to cause silicosis. The aim of this study was to determined the prevalence of occupational asthma among denim bleachery workers in Kayseri. The study was conducted in 4 factories, in which jean bleachery was performed, in Kayseri between December 2008 and February 2009. Overall, forty-four subjects, 22 from the bleachery section and 22 from the other sections, were included. A questionnaire about respiratory symptoms was administered. Pulmonary function tests (PFTs) and serial peak expiratory flow (PEF) measurements were performed. All subjects were evaluated by posteroanterior chest x-rays. The prevalence of occupational asthma (OA) in the bleachery and other section workers was 23.8% and 9.1%, respectively (p> 0.05). Within workers, exercise dyspnea (23.3%) and wheezing (20.9%) were the most frequent symptoms. The relationship between the duration of employment and PFTs in bleachery workers (n= 21) was negatively correlated and statistically significant with FEV(1), FEF(2)(5)-(7)(5) (moderate; r= -0.477, -0.449, respectively; p< 0.05) and FEV(1)/FVC, FEV(1)% (well; r= -0.588, -0.509, respectively; p< 0.05). The results of the present study suggest that exposure to denim-bleaching agents plays an important role in the occurrence of respiratory symptoms, reduction in pulmonary functions, and induction of occupational asthma.  (+info)

Among these are the stereocilia, each with a core of several hundred actin filaments that arise from their apical surfaces and that bear the mechanotransduction apparatus at their tips. Actin turnover in stereocilia has previously been studied by transfecting neonatal rat hair cells in culture with a β-actin-GFP fusion, and evidence was found that actin is replaced, from the top down, in 2-3 days. Overexpression of the actin-binding protein espin causes elongation of ...

Skin-whitening and skin-condition-improving effects of topical oxidized glutathione: a double-blind and placebo-controlled clinical trial in healthy women Fumiko Watanabe,1 Erika Hashizume,1 Gertrude P Chan,2 Ayako Kamimura11Healthcare Products Development Center, KYOWA HAKKO BIO CO., LTD., Tsukuba, Ibaraki, Japan; 2Clinical Trial Management and Testing Associates, Inc., Filinvest Corporate City, Alabang, Muntinlupa City, PhilippinesPurpose: Glutathione is a tripeptide consisting of cysteine, glycine, and glutamate and functions as a major antioxidant. It is synthesized endogenously in humans. Glutathione protects thiol protein groups from oxidation and is involved in cellular detoxification for maintenance of the cell environment. Reduced glutathione (GSH) has a skin-whitening effect in humans through its tyrosinase inhibitory activity, but in the case of oxidized glutathione (GSSG) this effect is unclear. We examined the skin-whitening and skin-condition effects of topical GSSG in healthy women
Denim jeans or coats never came up short on style in the realm of design. I wager everybody has their own pair of denim jeans. They are ordinary, adaptable and utilitarian to wear. Our denims and our garments need unique consideration and care. Washing your preferred pair of denims with care will keep going for quite a long while. To keep your jeans and coats in flawless condition, here are some helpful hints on the most proficient method to wash and think about your denims.. Washing. The most prescribed approach to wash your denims is through hand-wash. In the event that you have a lot of time to save in your clothing, attempt to hand-wash your denims. This will avoid your jeans to loosen up, release or blur. At the point when you hand wash your jeans, be enthusiastic about the sew lining. Earth and residue remains in the trim line which make them noticeable. Utilize a decent quality cleanser and delicately brush the material. For occupied individuals, you can likewise wash your preferred pair ...
Im sad because this is the last outfit I shot with Portia during our last meeting, boo! How great is she? In this outfit I went for double blue denim paired with black denim, so really triple denim!!! Crazy!! I am actually very into it, though. This longline denim shirt (which I think was originally intended as a fitted dress, god love the 90s) from Cow Vintage is a nice alternative to the sort of denim overshirt I usually go for, and its quite a different piece which I like. I also love this frayed denim t-shirt from the Topshop x Marques Almeida collection - I seriously still want every denim piece from that collab, and most of it is on sale now! My coat is also worthy of a mention, you might remember it from my feature with Primark in Company Magazine way back in October but I finally got my hands on one last month. I think its going to be the ideal transitional coat as it (hopefully) starts to warm up a little at some point in the not too distant future, but as far as trench coats or macs ...
The ability of the gastrointestinal epithelium to act as a barrier is critically important in terms of preventing infection. There are many components to epithelial defence, including the ability of the epithelium to secrete electrolytes, water, and mucus, and the ability (by virtue of tight junctions) to physically restrict the movement of microbes and microbial products into the lamina propria. Impairment of these defensive functions could therefore predispose an organism to bouts of infection and inflammation. Intestinal inflammation has been shown to produce prolonged alterations in smooth muscle function, which could contribute to symptom generation in disorders such as irritable bowel syndrome.24 In a series of studies over the past few years, we have sought to determine if a bout of inflammation of the intestine would similarly result in prolonged epithelial dysfunction.7,8 Indeed, colitis induced by a hapten (TNBS) and enteritis induced by Nippostrongylus brasiliensis were both found to ...
Bangladesh has become a hotspot for denim products sourcing thanks to the countrys expertise in denim manufacturing and competitive advantages due to low production cost. Meanwhile, China has lost its grip in the denim sector to some extent due to a rise in the production cost. This change in Chinese denim sector is diverting more orders to Bangladeshi denim exporters. Right now, we are getting a large number of orders from the international buyers. This is because China is moving away from low-end products, Sayeed Ahmad Chowdhury, general manager (operation) at Square Denim, told the daily sun. The number of orders increased by 20-25 percent in a year, he added. Currently, our production capacity is 1.5 million yards a month, which will increase to three million from next month since the demand is continuously rising, Sayeed Ahmad said. He said to make a good use of Chinas backtrack on low-end clothing, Bangladeshi entrepreneurs should focus to tap this opportunity by taking more orders ...
Generation of reactive oxygen intermediates (ROI) following antigen receptor ligation is critical to promote cellular responses. However, the effect of antioxidant treatment on humoral immunity during a viral infection was unknown. Mice were infected with lymphocytic choriomeningitis virus (LCMV) and treated with Mn(III)tetrakis(4-benzoic acid)porphyrin chloride (MnTBAP), a superoxide dismutase mimetic, from days 0 to 8 postinfection. On day 8, at the peak of the splenic response in vehicle-treated mice, virus-specific IgM and IgG antibody-secreting cells (ASC) were decreased 22- and 457-fold in MnTBAP-treated animals. By day 38, LCMV-specific IgG ASC were decreased 5-fold in the bone marrow of drug-treated mice, and virus-specific antibodies were of lower affinity. Interestingly, antioxidant treatment had no effect on the number of LCMV-specific IgG memory B cells. In addition to decreases in ASC, MnTBAP treatment decreased the number of functional virus-specific CD4+ T cells. The decreased numbers of

ADVANCED DENIMs Diresul Indicolor concept opens up a rich new vista of production and design possibilities. Apart from eliminating hydrosulfite in the dyeing process, it makes manufacturers more versatile by offering them the choice of solid or ring denim and a wider range of blues and navies, blacks and grays. It enables them to create new effects by using sustainable techniques at either the dyeing or wash-down stage, and guarantees overall higher-quality results.. In the past, most of the features and effects that make jeans so special and so versatile have been created during wash-down using chlorine and permanganates, use of which is controversial. Now ADVANCED DENIM allows you to use peroxide-based wash-downs and sidestep any environmental issues.. All this means is that manufacturers and consumers who care about sustainability now have a real choice without compromising on any fabulous visual effects: ADVANCED DENIM. ...
Certain features of the dynamics of homogeneous populations on NNs are generic, resulting from two ubiquitous topological properties: heterogeneity and assortativity. In general, the dynamical process that begins when the population enters the NN and finishes when mutation-selection equilibrium is achieved is characterized by as many different time scales as different degrees of robustness are found in the NN. These time scales successively show up along the transient and affect the process of mutation fixation. As time elapses, regions of increasingly higher degree are visited, causing entrapment of the population. The actual time spent on a NN should increase as the difference between the fitness of the current phenotype and that of neighbouring alternatives grows, because fitter phenotypes trap populations for longer times.. Some of the features here described directly affect evolutionary dynamics on realistic NNs and, therefore, quantitative properties of the actual mutation substitution ...
The ideal timing for you to nourish your skin is right after the moisture retention step aimed at imparting the skin texture with a tenderly soft and dewy moist touch. This is because the nourishing, whitening, lifting, anti-ageing, and other effective constituents provided in a facial cream can now be more efficiently absorbed to pamper the skin in a more holistic manner while beefing up its barrier functions. That explains why it is so important for you to choose a facial cream of top-grade quality. ...

The science behind the Dry Brine: The addition of salt alone will allow proteins to bind more readily to water during the cooking process, so in the simplified sense that brines improve texture, flavor and moisture retention, a salt heavy dry rub can be thought of as a dry brine.. When making a dry brine, salt is usually mixed with other dry seasonings such as herbs and spices and rubbed onto the surface of the protein. A good starting point for the amount of salt to use is around 1% based on the proteins weight.. The salt rub is left on for a given period of time, (anywhere from 4-48 hours), and then cooked as is, usually without being rinsed. Although this method doesnt introduce excess water to be absorbed, the salting does allow the protein to bind moisture more tightly, yielding a moister finished product, assuming of course that the protein is cooked properly.. To understand whats really happening, you have to look at the structure of turkey muscles. Muscles are made up of long, ...

Scarring can lead to rough texture in the skin, an increase of collagen bundles, and a lack of hair follicles or sweat glands. However, it may also create discoloration within the skin, with damaged tissue taking on a darker shade. This is known as post-inflammatory hyperpigmentation and it ranks among the most common effects of hypertrophic, keloid, and atrophic scarring.. What is Post-Inflammatory Hyperpigmentation?. Post-inflammatory hyperpigmentation occurs when a bodys cellular process is changed. Damaged tissue is detected and several chemical responses occur, with a sudden increase of collagen, protein, and melanin (the property that determines the color of a persons skin, eyes, and hair). These elements are meant to heal the scar. However, they often trigger a sudden darkening of the skin.. Read More: Hyperpigmentation. How Does Post-Inflammatory Hyperpigmentation Affect Scars?. Background: Most data on laser resurfacing have come from studies of people with Fitzpatrick skin types 1 - 3; however, the worlds population is comprised mostly of Fitzpatrick skin types 4 - 6, which are more susceptible to post-inflammatory hyperpigmentation (PIH). Objective: For the purpose of expanding the expertise of plastic surgeons treating patients with darker skin types, this study examined the incidence of PIH in Asians who underwent laser resurfacing, including a histologic arm on fractional ablative resurfacing. Methods & Materials: The clinical study included six subjects of Vietnamese origin who underwent single-depth fractionated CO2 laser resurfacing. The histologic study involved a seventh subject. The MiXto SX® laser with a new scanning handpiece was used, along with magnifying loupes to assess ablative depth after each of three laser passes performed. Photographs were taken at various postoperative intervals. Results: All six clinical subjects showed cosmetic improvement

Climate change and land management are altering forest fire frequency and intensity worldwide. In some Northeast U.S. forests, pitch pine (Pinus rigida Miller) is not suffering from presence but rather a lack of wildfire events. In their absence, prescribed fire is being used to diminish fuel loads, open canopies and reduce competition. Pyrogenic carbon (PyC) produced by the fires may also improve soil moisture retention and plant physiological processes. Where the application of prescribed fire is not feasible due to nearby human populations, we reason prescribed fire PyC could be replaced by anthropogenic PyC product to provide similar soil benefits. We tested this hypothesis with pitch pine seedlings at a site absent overstory planted in submerged tree pots with control and PyC-imbued soils. Investigators found anthropogenic and forest PyC fostered similar growth, soil moisture retention and photosynthetic intrinsic water use efficiency, both significantly higher than unamended soils. We ...
Ok you voted for this post today. Post Inflammatory Hyperpigmentation (PIH) is something I struggle with constantly being darker in skin tone, once my active blemish is gone dealing with PIH is a given which can usually take 3-9months to fade (if left alone to heal).. A study in 2002 evaluating acne in skin of color found that 65.3 percent of African-American (N=239), 52.7 percent of Hispanic (N=55), and 47.4 percent of Asian (N=19) patients developed acne-induced PIH (ref below). Some studies go on to show 80-90% of coloured skin suffering from PIH post-acne. So statistically speaking, if you have coloured skin the chances of you having to deal with PIH are HIGH O HIGH! .. See the above infographic to look into PIH further and we will go through PIE next week. Hope this helps. Reference: Post Inflammatory Hyperpigmentation (Eica et al 2010). ...

South and Southeast Asia and Latin American together comprise 46 countries and are home to approximately 40% of the world population. The sociopolitical and economic heterogeneity, tropical climate, and malady transitions characteristic of the region strongly influence disease behavior and health care delivery. Acute kidney injury epidemiology mirrors these inequalities. In addition to hospital-acquired acute kidney injury in tertiary care centers, these countries face a large preventable burden of community-acquired acute kidney injury secondary to tropical infectious diseases or animal venoms, affecting previously healthy young individuals. This article reviews the epidemiology, clinical picture, prevention, risk factors, and pathophysiology of acute kidney injury associated with tropical diseases (malaria, dengue, leptospirosis, scrub typhus, and yellow fever) and animal venom (snakes, bees, caterpillars, spiders, and scorpions) in tropical regions of Asia and Latin America, and discusses the ...
Although bilimbi fruit is widely consumed in Malaysia, AKI due to Averrhoa bilimbi is rare, and case reports documenting its toxicity are scarce. Its nephrotoxicity is due to oxalate crystal deposition resulting in acute oxalate nephropathy, affecting people with normal kidney function, but more frequently in those with renal dysfunction.2 Bakul et al from India reported a series of cases in Kerala where 10 patients developed AKI following ingestion of bilimbi fruit juice where 7 out of 10 required haemodialysis.3 Nair et al also reported two cases of acute renal injury after both patients took bilimbi fruit juice as a presumed treatment for dyslipidaemia.4 The renal biopsy findings from patients who developed AKI following bilimbi fruit ingestion showed acute tubulointerstitial nephritis and tubular obstruction with calcium oxalate deposits. It is believed that the nephrotoxicity comes from the direct tubular damage from the calcium oxalate crystal obstruction of the tubules. Other mechanisms ...

Niacinamide, like the more familiar niacin, is a form of Vitamin B3. The mainstream medical community uses both niacin and niacinimide in the treatment of diabetes. Niacinamide is also gaining a reputation for its effect on uneven skin tone and its ability to mitigate acne and the red marks it leaves behind. The medical term for this condition is post-inflammatory hyperpigmentation.. Dr. Matthew Draelos, a pioneer in the field of niacin application, says this:. Niacinamide… has shown to be an effective skin-lightening agent, and, due to its anti-inflammatory properties, it is being tested as a possible treatment for acne and rosacea.. POST-INFLAMMATORY HYPERPIGMENTATION. This is a response to injury of the skin and can be the result of acne, burns, friction or aggressive skin treatments. This condition may resolve with time and generally responds to topical products, although it can recur.. WHAT CAUSES PIGMENTATION?. Most of us are born with baby soft, smooth and clear complexion. As we age, however, the melanocytes malfunction or become damaged due to various factors which interfere with the normal functioning of melanin production.. When there is an overproduction of melanin, dark spots and uneven areas of hyperpigmentation surface as a result of the clumping and uneven distribution of melanin in the epidermis.. PIGMENTATION CONTRIBUTING FACTORS. SUN RAYS. Excessive and long-term exposure to sunlight is a major cause of hyperpigmentation in skin (particularly on the face, arms, hands and back) as it is left unprotected from the suns harmful uv rays. Further darkening of already hyperpigmented areas also occurs as melanin ...

Suzan Obagi, MD, Associate Professor of Dermatology and Associate Professor of Plastic Surgery, Director, UPMC Cosmetic Surgery and Skin Health Center University of Pittsburgh Medical Center hears this question a lot:. Many patients ask if they should take a break from using their skincare products during the summer months.? This stems from concerns with sun exposure and increased outdoor activities.? However, I have oftentimes found that patients that need the NuDerm product line most are those with pigmentation issues such as melasma or post-inflammatory hyperpigmentation.? These are conditions that typically flare and worsen during hot, sunny weather.? There are things we become accustomed to in life, morning commutes, waiting in line, messy kids-but bad skin shouldnt be one of them. For Monica, it was. But after a nurse showed her just how dull and discolored her skin had become, she decided to tackle the issue head-on.

I have been a long time double cleansing girl with moisturizers and spot treatments since my college years and I am now in my early 30s. People say that I still have good skin and still look like in my early to mid-20s. Needless to say, I still get the occasional cystic acne about 2x/month and always always on my chin which leaves me with post-inflammatory hyperpigmentation that takes forever to go away. Literally. My nose is bothersome for me because of the pores which I have never been able to fix. I still have, from what it looks, capillaries that have always been visible on my face and I dont know if theres a way to ever make them less visible. My dark undereyes have been there since childhood. Redness around the nose is also an issue, and just basically an overall uneven tone of my skin ...

Ceramides make up approximately 50% of the lipid (oily) component of the intercellularcement which binds epidermal skin cells together, they play a key role in maintaining the moisture level and firmness of the skin. A chronic decline of the ceramide concentration in the skins intercellular cement has been demonstrated to be part of the skin ageing process, as it allows penetration of damaging environmental elements and results in trans-epidermal evaporation of moisture.. Intercellular cement in the skin performs the same function as mortar between the bricks in a wall. As the wall ages, the mortar becomes porous and cracks. Ceramide is the substance in the skin which prevents this from happening to the intercellular cement. The amount of ceramides in the epidermis proportionately influences the skins dryness - fewer ceramides results in drier skin and more crepey wrinkles. Applying ceramides to your skin results in increased moisture retention in the epidermis.. Ceramides are also excellent ...

Hyaluronic acid (HA) is a natural and linear polymer composed of repeating disaccharide units of β-1, 3-N-acetyl glucosamine and β-1, 4-glucuronic acid with a molecular weight up to 6 million Daltons. With excellent viscoelasticity, high moisture retention capacity, and high biocompatibility, HA finds a wide-range of applications in medicine, cosmetics, and nutraceuticals. Traditionally HA was extracted from rooster combs, and now it is mainly produced via streptococcal fermentation. Recently the production of HA via recombinant systems has received increasing interest due to the avoidance of potential toxins. This work summarizes the research history and current commercial market of HA, and then deeply analyzes the current state of microbial production of HA by Streptococcus zooepidemicus and recombinant systems, and finally discusses the challenges facing microbial HA production and proposes several research outlines to meet the challenges.
According to the University of Illinois Extension, soil structure refers to the way inorganic particles, such as sand, silt and clay, combine with decayed organic particles, like humus and compost. Soil with a healthy structure is crumbly to the touch, allowing plenty of room for air, water and energy to move freely.. Adding compost to your garden also helps to neutralize pH and improve the cation exchange capacity (CEC) of soils, increasing their ability to hold nutrients for plant use.. So, why does all of this matter? If youve had trouble getting certain fruits, vegetables or decorative plants to grow in your garden, poor soil structure may be to blame. Think of it this way: If the soil in your garden is hard and clay-like, young roots have to struggle to get through and obtain the nutrients they need for healthy growth. If the soil is sandy, it may be lacking nutrients plants need to thrive.. When used in sufficient quantities, adding compost has both immediate and long-term positive ...
