Biochemical properties of polysaccharides from black pepper. (1/13)

The purified polysaccharides from Piper nigrum were prepared as follows: a hot water extract of pepper seeds was fractionated by ultrafiltration with a 5-kDa-membrane cartridge. A fraction with 5 kDa or bigger molecules was successively purified by open column chromatography on DEAE-Toyopearl 650C and Bio-gel P-60 with each active fraction, resulting in PN-Ib and PN-IIa, purified anti-complementary polysaccharides. None of the anti-complementary activity of any polysaccharide was changed by pronase digestion or polymyxin B treatment, but they were decreased by periodate oxidation. Analysis of component sugar and molecular mass determination of the anti-complementary polysaccharides indicated that PN-Ib with an average molecular mass of 21 kDa contained 88.5% glucose and other negligible minor monosaccharides, while PN-IIa showed a different monosaccharide composition, which contained a significant proportion of galactose, arabinose, galacturonic acid and rhamnose. The molar ratio of galactose and arabinose of PN-IIa (48 kDa) was 1.93:1. PN-1 did not react with beta-glucosyl Yariv reagent, however, PN-IIa did react, which indicated that PN-IIa might be an arabinogalactan. Based upon these results, the usefulness of purified anti-complementary polysaccharides from Piper nigrum is suggested as a supplement for immune enhancement.  (+info)

Screening of antagonistic bacteria for biological control of nursery wilt of black pepper (Piper nigrum). (2/13)

Bacterial antagonists of Phytophthora capsici were isolated from underground shoot portions of rooted cuttings of black pepper. Initially isolates were screened by dual culture on potato dextrose agar and carrot agar. Further, a screening was done on black pepper shoots for supression of lesion caused by the pathogen. Most of the antagonists showed varying levels of antagonism in the dual culture and the shoot assay. Isolate PN-026, showing the highest suppression of lesion development in the shoot assay was found to be the most efficient antagonist in reducing Phytophthora capsici induced nursery wilt of black pepper. This screening involving the host, pathogen, and the antagonist, performed on black pepper shoot (the planting material for this vegetatively propagated crop), could be used as a rapid and reliable method for the isolation of efficient bacterial antagonists of P. capsici.  (+info)

Effects of piperine, the pungent component of black pepper, at the human vanilloid receptor (TRPV1). (3/13)

1. We have characterised the effects of piperine, a pungent alkaloid found in black pepper, on the human vanilloid receptor TRPV1 using whole-cell patch-clamp electrophysiology. 2. Piperine produced a clear agonist activity at the human TRPV1 receptor yielding rapidly activating whole-cell currents that were antagonised by the competitive TRPV1 antagonist capsazepine and the non-competitive TRPV1 blocker ruthenium red. 3. The current-voltage relationship of piperine-activated currents showed pronounced outward rectification (25+/-4-fold between -70 and +70 mV) and a reversal potential of 0.0+/-0.4 mV, which was indistinguishable from that of the prototypical TRPV1 agonist capsaicin. 4. Although piperine was a less potent agonist (EC50=37.9+/-1.9 microM) than capsaicin (EC50=0.29+/-0.05 microM), it demonstrated a much greater efficacy (approximately two-fold) at TRPV1. 5. This difference in efficacy did not appear to be related to the proton-mediated regulation of the receptor since a similar degree of potentiation was observed for responses evoked by piperine (230+/-20%, n=11) or capsaicin (284+/-32%, n=8) upon acidification to pH 6.5. 6. The effects of piperine upon receptor desensitisation were also unable to explain this effect since piperine resulted in more pronounced macroscopic desensitisation (t(1/2)=9.9+/-0.7 s) than capsaicin (t(1/2)>20 s) and also caused greater tachyphylaxis in response to repetitive agonist applications. 7. Overall, our data suggest that the effects of piperine at human TRPV1 are similar to those of capsaicin except for its propensity to induce greater receptor desensitisation and, rather remarkably, exhibit a greater efficacy than capsaicin itself. These results may provide insight into the TRPV1-mediated effects of piperine on gastrointestinal function.  (+info)

Qualitative analysis of MDR-reversing Anastasia Black (Russian black sweet pepper, Capsicum annuum, Solanaceae) extracts and fractions by HPLC and LC-MS-MS methods. (4/13)

In earlier experiments, the MDR (multidrug resistance)-reversal activities of Anastasia Black (Russian black sweet pepper) extracts had been analysed. Recently, the most effective MDR reversing extracts and fractions have been separated by HPLC (high-performance liquid chromatography, for carotenoids) and LC-MS-MS (HPLC combined with mass spectrometry, for phenolic compounds) methods. As a result of the analytical studies, the following flavonoids had been identified: feruloyl glucopyranoside, quercetin rhamnopyranoside glucopyranoside, luteolin glucopyranoside arabinopyranoside, apigenin glucopyranoside arabinopyranoside, quercetin rhamnopyranoside, luteolin arabinopyranoside diglucopy-ranoside, hesperidine and luteolin glucuronide. According to the literature, the aglycones of these phenolic compounds exhibit MDR-reversal activity in vitro, and the connection between the phenolic content of Anastasia Black and MDR-reversal action was therefore studied by different analytical methods. The results of this study revealed that the identified flavonoids of Anastasia Black may be only partially responsible for the modulation of the MDR of mouse lymphoma cells. Other lipophilic compounds, most probably carotenoids, present in Russian black sweet pepper may act as inhibitors of MDR reversal.  (+info)

A preliminary assessment of genetic relationships among agronomically important cultivars of black pepper. (5/13)

BACKGROUND: The impact of diseases such as Phytophthora foot rot and the replacement of unproductive cultivars by high yielding ones has brought about the disappearance of varieties in Piper species, like any other crop. Black pepper (King of spices), is a major spice crop consumed throughout the world. It is widely cultivated across various parts of the world apart from India. The different cultivars may be genetically related and could be a source of valuable genes for disease resistance and an increase in quantity and quality. Even though Western Ghats in India is believed to be the site of origin of this crop, numerous accessions from the NBPGR have not yet been evaluated. Our study aims to investigate the genetic relatedness in major cultivars of black pepper using Amplified Fragment Length Polymorphism. RESULTS: Amplified Fragment Length Polymorphic (AFLP) DNA analysis was performed in thirty popular cultivars of black pepper from National Bureau of Plant Genetic Resources (NBPGR), India. Fingerprint profiles were generated initially with, five different primer combinations, from which three primer pair combinations (EAGC/MCAA, EAGG/MCTA and EAGC/MCTG) gave consistent and scorable banding patterns. From 173 scorable markers, 158(> 90%) were polymorphic which shows there is considerable variation in the available germplasm. The dendrogram derived by unweighted pair group method analysis (UPGMA) grouped the accessions into three major clusters and four diverse cultivars with only 30% similarity. Karimunda, a widely grown and popular cultivar was unique in the fingerprint profiles obtained. CONCLUSION: There are currently few fingerprinting studies using the valuable spice crop black pepper. We found considerable genetic variability among cultivars of black pepper. Fingerprinting analysis with AFLP proved to be an ideal tool for cultivar identification and phylogenetic studies. It shows the high level of polymorphism and the unique characterization of the major cultivars. An extensive range of similarity value between the cultivars was noted (6.01 to 98.13). Further screening of more cultivars will provide valuable information for current breeding programmes.  (+info)

Olfactory stimulation using black pepper oil facilitates oral feeding in pediatric patients receiving long-term enteral nutrition. (6/13)

Patients with severe neurological disorders often require enteral nutrition (EN). Since long-term EN can cause multiple complications, reinstating the oral intake of food is beneficial. Olfactory stimulation using black pepper oil (BPO), a strong appetite stimulant, was reported to facilitate swallowing in older people. Therefore, the effects of olfactory stimulation with BPO were investigated in pediatric patients receiving long-term EN due to neurological disorders. The effects of scenting with BPO for 1 min immediately before every meal were evaluated in ten patients: 4 boys and 6 girls, aged 19-97 months (51 +/- 26 months). The neurological disorders included periventricular leukomalacia (3 patients), hypoxic ischemic encephalopathy (3), Costello syndrome (1), Russell-Silver syndrome (1), Miller-Dieker syndrome (1), and cerebral palsy of unknown etiology (1). In eight of these patients, BPO intervention was continued for 3 months. Five of these eight patients showed increases in the amount of oral intake with desirable effects including facilitated swallowing movement, although complete elimination of the need for EN was not achieved. In the other three patients, BPO intervention was not effective; severe cerebral tissue loss, profound malformation or intractable seizures seemed to reduce the efficacy of BPO. In two cases, BPO intervention was discontinued due to cough or because the odor of BPO was unbearable to the family. In conclusion, olfactory stimulation with BPO facilitated oral intake in a subset of patients on long-term EN. BPO stimulation may be useful for facilitating oral intake when used in combination with conventional methods.  (+info)

Activation of TRPV1 and TRPA1 by black pepper components. (7/13)

We searched in this study for novel agonists of transient receptor potential cation channel, subfamily V, member 1 (TRPV1) and transient receptor potential cation channel, subfamily A, member 1 (TRPA1) in pepper, focusing attention on 19 compounds contained in black pepper. Almost all the compounds in HEK cells heterogeneously expressed TRPV1 or TRPA1, increased the intracellular Ca(2+) concentration ([Ca(2+)](i)) in a concentration-dependent manner. Among these, piperine, isopiperine, isochavicine, piperanine, pipernonaline, dehydropipernonaline, retrofractamide C, piperolein A, and piperolein B relatively strongly activated TRPV1. The EC(50) values of these compounds for TRPV1 were 0.6-128 microM. Piperine, isopiperine, isochavicine, piperanine, piperolein A, piperolein B, and N-isobutyl-(2E,4E)-tetradeca-2,4-diamide also relatively strongly activated TRPA1, the EC(50) values of these compounds for TRPA1 were 7.8-148 microM. The Ca(2+) responses of these compounds for TRPV1 and TRPA1 were significantly suppressed by co-applying each antagonist. We identified in this study new transient receptor potential (TRP) agonists present in black pepper and found that piperine, isopiperine, isochavicine, piperanine, piperolein A, and piperolein B activated both TRPV1 and TRPA1.  (+info)

Adiposity suppression effect in mice due to black pepper and its main pungent component, piperine. (8/13)

We investigated energy metabolism enhancement by pepper by examining suppression of body fat accumulation in mice due to piperine (PIP) and black pepper (BP) intake. To induce adiposity, mice were fed a high-fat, high-sucrose (HFS) diet as a control diet for 4 weeks. Visceral fat weights decreased significantly in the mice fed diets of 0.03% and of 0.05% PIP. Body weight in the 0.05% PIP group also decreased significantly. In the mice fed a diet of 1.0% BP, body weight and visceral fat weights decreased significantly. For all parameters tested, the 1.0% BP group tended to show values slightly lower than those of the 0.03% PIP group. Expression of thermogenic protein uncoupling protein 1 tended to increase in the mice on the 1.0% BP diet. These results indicate that BP suppresses the effect of body fat accumulation mainly through the action of PIP.  (+info)

We try to keep things simple here in Bertie County. For example, we have only three traffic lights in town, and probably could make do with only one! Another example of keeping things simple was Papa Jacks love of grilled steaks seasoned only with salt and pepper. And, since we like to carry on tradition here at Bertie County Peanuts we thought it might be interesting to try seasoning some of our Blister Fried Peanuts with a mixture of salt and pepper. Well, needless to say, that uncomplicated combination of tastes was a big hit with our local taste testers. And, after a few months of recipe tweaks we are proud to introduce Sea Salt & Black Pepper Peanuts. We think youll love the simple combination of flavorings. We do!OU Kosher Certified

One thing I do remember from my original review was how delightfully soft and supple the wrapper was, though that no longer seems to be the case. It is still fairly soft and a bit oily, but not nearly as much as before. It has firmed up a bit as well, with only a slight bit of give and certainly closer to what I would call firm as opposed to the soft side. Light hay and nuts still emanate from the wrapper, though the cold draw still brings a rich, pastry-like profile made up of hay, toasted almonds, powdered sugar and vanilla.. Starting into the first third Im reminded again why I enjoyed the initial profile so much, with sweet fruits, smoky oak, almonds, hints of vanilla and just a touch of black pepper. About an inch in and both the black pepper and smoky oak notes grow, becoming much more dominant and bringing the sweetness of the profile down a notch. Moving into the second third sees the black pepper go from a dominant flavor to an overbearing note, drowning out the sweet fruits and subtle ...
I love turmeric! I especially love that this is organic! Many herbs have additives or metals in them so its important to me to buy organic for full benefits. However, turmeric is only absorbable with black pepper or a healthy fat like coconut oil. So be sure to take this supplement with one or the other to get the full benefits. Turmeric is outstanding for inflammation. It helps my dads gout and my mothers arthritis tremendously. Turmeric helps them so much that they dont need pharmaceuticals to control their inflammation. That being said, they like to take a supplement that already has black pepper in it because its hard for them to remember to take black pepper after this supplement. However, still giving 5 stars because this supplement is pure and Organic! Sometimes that is hard to find! ...
Prior to the European discovery of the New World, long pepper was an important and well-known spice. The ancient history of black pepper is often interlinked with (and confused with) that of long pepper. The Romans knew of both and often referred to either as just piper; many ancient botanists erroneously believed dried black pepper and long pepper came from the same plant. Only after the discovery of the New World and of chile peppers did the popularity of long pepper decline. Chile peppers, some of which, when dried, are similar in shape and taste to long pepper, were easier to grow in a variety of locations more convenient to Europe ...

Chillies made it to Europe with Christopher Columbus, returning from the New World after his 1492 voyage. Columbus was searching for a shorter route to India, source of the valuable spice black pepper. By the time of Columbuss arrival in Cuba, there were probably hundreds of types of chilli peppers being raised. In 1492 Columbus saw some plants cultivated in Hispaniola (the second largest island in the Caribbean) by the Arawak Indians, (# 4.) who called them "axi," which became "aji." Chillies are a completely different botanical subclass, order and family than black pepper (see below). Nonetheless, Columbus, thinking he was in India, called them "red peppers." He also misnamed the inhabitants "Indians.". All chillies are of the genus Capsicum, generally used only for taxonomic discussion. (taxonomy: "a classification of organisms into groups based on similarities of structure or origin etc" - WordNetWeb) The common name comes from the Nahuatl (Aztec) "chil" or "chili". Eating good food is crucial to good health. Plain and simple. No one can argue with that. But we cant stop there. We have to make sure were creating the right environment for the body to soak up all the amazing vitamins and minerals in our food. Unfortunately, because of issues with food allergies, inflammatory bowel disease, intestinal infections, pancreatic problems, thyroid and other glandular dysfunctions, some people may suffer from poor nutrient absorption. Obviously not everyone suffers from those issues.. But a lot of people suffer from everyday stress and poor habits such as not working out or rushing through meals. All of those things can have a negative impact. So how do we encourage our gastrointestinal tract to perform like a lean, mean, digestion machine? Well, one very simple way is to add some spice to your life - literally. Black pepper has been frequently used in Ayurveda to improve digestion. Black pepper contains piperine which helps nutrients penetrate the intestinal wall. ...
The researchers found that the piperine appeared to make the curcumin more effective. This is an especially significant finding because the cancer-preventive drugs tamoxifen or raloxifene, which can have highly toxic side effects, do not affect estrogen-independent tumors ...

I like to experiment with recipes, change ingredients around and swap stuff in or out to make it healthier. This past Sunday I modified a stew recipe that I found on Mind Body Green and it turned out amazing. So good, in fact, that I have to share.. This Turmeric, Lentil, and Vegetable stew takes about 45 minutes to cook and is a nutritional powerhouse. Heres just a few reasons why.... Turmeric, one of the main spices in this stew, has been shown to be effective against arthritis, coughs, cancer, and chronic diseases stemming from inflammation. It helps to block a protein that promotes tumor growth and causes inflammation. Research suggests that it may help reduce the risk of Alzheimers disease. The Papaya (Carica papaya) is an edible fruit of the citrus family, originally discovered in to southern Mexico and Central America and spread to the rest of world by the Spanish Conquistadors. It can now be found growing in every tropical and subtropical country. There are two varieties of papayas, the Hawaiian and Mexican. The Hawaiian varieties are those most commonly found in supermarkets. The pear-shaped fruit weigh approximately 0.5 kg, with a yellow skin when ripe. Depending on the variety, the flesh is either bright orange or pinkish, with a cluster of small black seeds in the center. A fully ripe papaya is juicy, with a sweetish and somewhat musky taste, comparable to that of a musk melon. Both the fruit and leaves contains papain, which assists digestion and is often used to tenderize meat. The edible seeds have a spicy flavor somewhat reminiscent of black pepper, and these seeds are often used, especially in India, as an adulterant for whole black pepper. It is also commonly referred ...

I remember it so clearly-my friend and I sat on the couch enjoying the last rays of the Colorado sunshine pouring into the living room. That night we had plans to eat some Indian food, watch some Netflix and relax with some edible candies recently purchased at a dispensary in Telluride. The edibles were made with a hybrid strain and the label proclaimed "10 mg of THC per candy." The gelatinous, fruit-flavored squares had an undeniably skunky aroma.. I had had bad experiences with edibles before. This form of ingestion is wonderful for people who dont want to smoke or vaporize. But, it tends to take a little more time and the effects can come on unexpectedly. 10 milligrams is definitely enough to feel some sort of way; I knew my friend did not have as much experience with edibles. She usually preferred the effects of inhalation, which are almost immediate. As we unscrewed the child-proof lid to the candy, I mentioned that we should probably start by splitting a candy. We would see how we felt ...

Here in Argentina those are very popular, and a typical recipe. The recipe for chipás is almost the same you used there, with two major differences. First, we use mandioca flour (I dont know what the name would be in english, sorry), wich makes the puffs more... well, puffy, without the need of having so much trouble with the mixing and quantities. Second, the cheese. I use the same amount of cheese than flour - they have to be cheesy! If you can, get 3 types of cheese and mix them. You can also use cream cheese (is it called like that? the cheese that is soft and not like the parmesano and the others?) and a little bit of other hard cheese. With two eggs you are good, add some milk (just some, to make the preparation soft and easy to handle, not so sticky), more salt and voilà! Just in case anyone misunderstood me, I wrote the things that I do differently from you ...
My kids loved the Skinny Pop. Lydia really liked the Black Pepper popcorn. Its not as spicy as I was expecting, and has just a hint of black pepper flavor. The Lightly Salted Skinny Pop is my favorite. This popcorn proves that you can have a rich butter flavor without the popcorn being drenched in butter. It has the perfect mouth-feel and just the right amount of butter flavor. My daughter did say that, if you like it saltier, just turn the bag upside down and shake before you open it. I, personally, thought it had enough salt.. Of course, Skinny Pop features what the kids love best - great taste and easily portable for school lunches. However, its also got what I care about - good nutritional value and no artificial anything. Skinny Pop has zero trans fat and no cholesterol. The Skinny Pack bags are my personal favorite. They only come in the Lightly Salted flavor, but thats OK by me, because its my favorite. Each single serve bag only has 100 calories, 2 grams of fiber and 2 grams of ...

I am searching for the authentic pfeffernusse recipe. Anyone have one or suggest a source? I have googled and found a couple but I dont know which is more authentic. Some have almonds for the nuts part and some recipes don call for nuts at all. The spices called for are generally the same. Then there is the debate over black pepper and white pepper. My neighbor is a WWII vet harking back to a childhood memory of this Christmas cookies. He says the ones he had were round, hard and dusted with powered sugar.

By Dean Robinson. Science has already solved one great historical mystery--the fate of Russias last czar, Nicholas II,whose bones were unearthed with those of his family outside Ekaterinberg, Russia, and authenticated by genetic testing. Now the same method will help sort through the muddle of people claiming links to Nicholass youngest children, Alexei and Anastasia Romanov, believed by many to have escaped their familys 1918 massacre by the Bolsheviks. The stakes are huge: definitive identification of some of historys most romantic figures; a bit part in Russian politics; benefits from a possible media bonanza; and, most intriguingly, a potential claim on a hoard of gold rumored to have been stashed by Nicholas. Most aspiring Romanovs are either charlatans or deluded. But the Ekaterinberg find has touched off a high-tech Cinderella search, attracting many offers of "royal" DNA for comparison to the genetic material in the bones. Efforts are already under way in several cases, including that ...
The Twilight Time Blu-ray of Anastasia looks great. Its also a welcome relief, for the new HD transfer does not have the cold bluish-teal cast imposed over several classic Fox releases of the last few years, notably a Desk Set Blu-ray, and a King and I edition that could have been re-titled "Rhapsody in Blue". This new image is not the same scan as streams from the web: its sharper and more detailed and the colors are dark and rich. The optical transitions have only a slightly higher granularity. The cinematography flatters the excellent costumes, which are perhaps better than what would have been created for Ingrid Bergman back in Hollywood. I did notice quite a few shots with the CinemaScope mumps, as well as some with heavily distorted fields - people at the right of the screen are often half as wide as they should be.. The multi-track audio is fine as well, and Twilight Time gives us its Isolated Score Track option for fans that want to sample Alfred Newmans lush, Romanoff-flavored music. ...

Anastasia was from Rome. She was the daughter of Praepextatus and Fausta and was beautiful in appearance, virtuous and from a wealthy family. Her mother instructed her in the faith of Christ. After her mothers death, she was given in marriage to a pagan named Publius Patricius. He subjected his wife to beatings and would keep her slaved in the house. She endured these torments for a short time since Publius drowned early in their marriage. As a young widow, she never remarried and secretely dedicated her time to the poor, those in prisons and served their needs daily. She would wash their wounds and especially console them during their anguish. Through her intercessions and prayers, she healed many from the effects of potions, poisons, spells and other harmful substances. This is how she received the honour of Deliverer of Potions. Her deeds and miracles spread throughout the area and her fame brought about her arrest under Diocletians persecutions. She was tortured and endured many torments ...
