Cations modulate the substrate specificity of bifunctional class I O-methyltransferase from Ammi majus. (1/4)

Caffeoyl-coenzyme A O-methyltransferase cDNA was cloned from dark-grown Ammi majus L. (Apiaceae) cells treated with a crude fungal elicitor and the open reading frame was expressed in Escherichia coli. The translated polypeptide of 27.1-kDa shared significant identity to other members of this highly conserved class of proteins and was 98.8% identical to the corresponding O-methyltransferase from parsley. For biochemical characterization, the recombinant enzyme could be purified to apparent homogeneity by metal-affinity chromatography, although the recombinant enzyme did not contain any affinity tag. Based on sequence analysis and substrate specificity, the enzyme classifies as a cation-dependent O-methyltransferase with pronounced preference for caffeoyl coenzyme A, when assayed in the presence of Mg2+-ions. Surprisingly, however, the substrate specificity changed dramatically, when Mg2+ was replaced by Mn2+ or Co2+ in the assays. This effect could point to yet unknown functions and substrate specificities in situ and suggests promiscuous roles for the lignin specific cluster of plant O-methyltransferases.  (+info)

Molecular cloning and functional characterization of psoralen synthase, the first committed monooxygenase of furanocoumarin biosynthesis. (2/4)

Ammi majus L. accumulates linear furanocoumarins by cytochrome P450 (CYP)-dependent conversion of 6-prenylumbelliferone via (+)-marmesin to psoralen. Relevant activities, i.e. psoralen synthase, are induced rapidly from negligible background levels upon elicitation of A. majus cultures with transient maxima at 9-10 h and were recovered in labile microsomes. Expressed sequence tags were cloned from elicited Ammi cells by a nested DD-RT-PCR strategy with CYP-specific primers, and full-size cDNAs were generated from those fragments correlated in abundance with the induction profile of furanocoumarin-specific activities. One of these cDNAs representing a transcript of maximal abundance at 4 h of elicitation was assigned CYP71AJ1. Functional expression in Escherichia coli or yeast cells initially failed but was accomplished eventually in yeast cells after swapping the N-terminal membrane anchor domain with that of CYP73A1. The recombinant enzyme was identified as psoralen synthase with narrow substrate specificity for (+)-marmesin. Psoralen synthase catalyzes a unique carbon-chain cleavage reaction concomitantly releasing acetone by syn-elimination. Related plants, i.e. Heracleum mantegazzianum, are known to produce both linear and angular furanocoumarins by analogous conversion of 8-prenylumbelliferone via (+)-columbianetin to angelicin, and it was suggested that angelicin synthase has evolved from psoralen synthase. However, (+)-columbianetin failed as substrate but competitively inhibited psoralen synthase activity. Analogy modeling and docked solutions defined the conditions for high affinity substrate binding and predicted the minimal requirements to accommodate (+)-columbianetin in the active site cavity. The studies suggested that several point mutations are necessary to pave the road toward angelicin synthase evolution.  (+info)

M ultiple environment trials (MET) are generally carried out by plant breeders to select and recommend high yielding and stable genotypes for a set of environments. The analysis of MET data often results in genotype - by - environment interactions which often causes difficulties in the interpretation of results and reduce efficiency in selecting the best genotypes . AMMI and GGE biplot analysis are two recent methods that are widely used to overcome these difficulties in MET data analysis. The objec tive of this study was to compare GGE biplot and AMMI analysis t hat determine the most efficient method for evaluating and describing genotype performance across environments . T en (10) cassava ( Manihot esculenta ) genotypes including two local checks were e valuated across six (6) environments in Southern Ghana . The experimental layout was a randomized complete block design with three replications. The Additive M ain E ffects and M ultiplicative I nteraction (AMMI) analysis of variance ...
Basidiomycetes isolated from rotted roots of alfalfa (Medicago sativa) and khella (Ammi visnaga) near Giza, Egypt, were evaluated for in vitro growth rates, temperature tolerance, pathogenicity, and similarity to isolates of Marasmiellus inoderma, a basidiomycete pathogen of maize and sugarcane in Egypt. All fungi produced similar white, appressed colonies on potato-dextrose, corn-dextrose, and malt agar media. Colony diameters indicated that the alfalfa and khella isolates were not identical; however, they were similar to those of M. inoderma. Growth of the alfalfa and khella isolates was similar to that of the M. inoderma isolates within the temperature range of 25 35 C, but below 20 C and at 38 C, development of the alfalfa and khella isolates was significantly greater than that of either isolate of M. inoderma. In pathogenicity tests, the alfalfa and khella isolates performed similarly but differently from either M. inoderma isolate on all hosts. Characteristics of basidiocarps of the ...
The goal of this research was to assess the stability and yield performance of 150 durum wheat genotypes in multi-environment trials in two locations (Diyarbakir and Kiziltepe), in 2011-2012, and 2012-2013 growing seasons. The trials were designed by Lattice Experimental Design with two replications (incomplete block design). The AMMI (Additive Main Effects and Multiplicative Interaction) and GEI (Genotype×Environment Interaction) analysis were used in the study to estimate GEI effects on grain yield, because of plant breeders great interest in these models for breeding programs. AMMI evaluation indicated that genotypes made the most important contributions to treatments Sum of Squares (59.8%), environments (3.5%), and GEI (36.7%), respectively, suggesting that grain yield had been affected by environment. IPCA 1 and IPCA 2 axes (Principal Component) were significant as P| 0.01 and explained 63.8 and 36.2%, respectively. Results showed that Kiziltepe 2013 was more stable and high yielding, meanwhile
To the best of our knowledge this is the first study regarding the reporting of B chromosomes in this plant species. During the meiotic study of Trachyspermum ammi (L.) Sprague, maximum two B chromosomes were reported in diploid plants which were retained in induced autotetraploid after the treatment of 0.2 % colchicine at seedling stage. In the present study, the meiotic behaviour of B-chromosomes was analyzed in Trachyspermum ammi (L.) Sprague. The presence of B chromosomes and their number in the plant species of higher ploidy level is controversial. The existence of B chromosomes in polyploids varies from species to species. In the present case, no change has been observed in the number of B chromosomes despite the duplication of A chromosomes in autotetraploid plant. The unaffected number of B chromosomes represents their peculiar behaviour of inheritance. The plant species show adaptive mechanism for their retention in population. In the present study, significant changes ( ...

Welcome back to the Lovecraft reread, in which two modern Mythos writers get girl cooties all over old Howards original stories. Today were looking at The Color Out of Space, written in March 1927 and first published in the September 1927 issue of Amazing Stories. You can read the story here. Spoilers ahead.. Summary: A surveyor for the new reservoir tours the area to be inundated. Hes heard the shunned countryside west of Arkham is not good for the imagination. Even he is spooked by the blasted heath: five acres of gray dust like a great spot eaten by acid into the woods and fields where a well releases vapors that stain the sunlight with strange hues.. Arkhamites tell him to ignore Ammi Pierces stories about the heath, so (of course) he seeks Ammi out. The old farmers surprisingly reconciled to a reservoir sinking the familiar landscape-its better underwater since the strange days of 1882-1883.. It began with the meteorite that fell on Nahum Gardners prosperous farm. ...

Private Members Bill C-442 was created to address this disparity between patient experience, scientific evidence, and public policy. Although the Bill received support from the College of Family Physicians of Canada and the Canadian Medical Association (18), it has been publically denounced by AMMI representative Dr. William Bowie, who claimed that it was one of the most egregious anti-science and pseudo-science pieces of legislation Ive ever seen (19). He contended that the Bill, and the preamble in particular, proposes that we ignore decades of solid scientific work and instead buy into a belief system that is largely founded on what I would say is pseudo-science and factoids and misrepresentation of data that does not have any solid scientific support behind it (19). ...

The medicinal plant Ammi visnaga is a valuable source of furanochromones and pyranocoumarins used as vasodilator agents. Its ability to germinate under unfavourable growth conditions, such as saline soil and hypoxia characterizing clay soils and marshes ecosystems, prompted us to qualitatively characterize secondary metabolites in umbels of A. visnaga plants grown under different conditions (in field, hydroponically controlled, and contrasted by salinity and/or hypoxia) by HPLC-ESI/IT/MSn analysis. Subsequently, the quantitative analysis of the bioactive compounds, above all furanochromones and pyranocoumarins, was carried out by HPLC-ESI/QqQ/MS/MS. The results show the influence of growing conditions on the quali-quantitative profile of A. visnaga secondary metabolites and evidence that hydroponic culture leads to increased level of A. visnaga active principles. Furthermore, two furanochromones never reported before were identified and characterized by 1D- and 2D-NMR analysis. ...

Since childhood I have been interested in the world of natural aromatics. This interest gradually developed into our home business White Lotus Aromatics. Keypoints along this aromatic journey were: 1) living on a small farm in India where many tropical fragrant plants were to be found 2) a career in horticulture, highlighted by working on a formal garden estate, Filoli 3) many journeys throughout the length and breadth of India to explore Indias ancient and modern aromatic traditions. Please note that I have an interest in the wonderful world of natural aromatics, but have no therapeutic expertise. gnm is a package for R, which provides facilities for specifying and fitting (by maximum likelihood) a broad class of generalized nonlinear models. These models are like generalized linear models (linear regression, logistic regression, log-linear models, etc.) but may also include one or more nonlinear terms in the predictor function (i.e., the right hand side of the regression equation). Some special cases are models involving multiplicatively structured interactions, such as the UNIDIFF and row-column association models from sociology and the AMMI and GAMMI models from crop science; stereotype models for ordered categorical response; Lee-Carter models for trends in age-specific mortality, and diagonal-reference models for dependence on a square 2-way classification.. The package authors, Heather Turner and David Firth, received the 2007 John M. Chambers Statistical Software Award for their work on gnm, which was supported by the ESRC Professorial Fellowship RES-051-27-0055.. Articles in SCGN ...
A common problem in multienvironment trials are the missing genotype-environmental combinations. Recently, Bergamo proposed a distribution-free multiple imputation method in the interaction matrix. The purpose of this paper is to evaluate the new development and compare it with methodologies that have success in the genotype-environmental trials with missing data, like the alternating least squares (ALS) and the robust estimates, using the Additive Main effects and Multiplicative Interaction Models (AMMI). Was made an simulation study based in real data, doing missed random considering different percentages, imputing the observations and comparing the methodologies through three criteria: the square root of the mean predictive difference, the Procrustes statistic and the Spearmans rank correlation coeficient. Was concluded that the multiple imputation is not better than the imputation based in a additive model without interaction, and the best results for the variance are obtained with robust ...

BACKGROUND: Khellin is a naturally occurring furochromone which, when combined with artificial ultraviolet (UV) A or solar irradiation (KUVA), is reported to repigment vitiligo skin as effectively as PUVA photochemotherapy. The exact mechanism of KUVA-induced repigmentation is unknown. OBJECTIVES: The aim of this study was to test the effect of khellin and KUVA on proliferation and melanogenesis of normal human melanocytes and Mel-1 melanoma cells in vitro. METHODS: Cultured normal human melanocytes, Mel-1 melanoma cells and fibroblasts were treated with khellin, UVA and KUVA and the effect on proliferation determined by cell counting. The effect on melanogenesis was determined by a radiometric melanin formation assay. Changes in gene expression and protein synthesis were determined by Northern blot, reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot analyses. RESULTS: Khellin stimulated proliferation of Mel-1 melanoma cells and melanocytes at concentrations between 1 nmol L-1 and
Background: The emerging trends of multidrug resistance among several groups of microorganisms against different classes of antibiotics led different researchers to develop efficient drugs from plant sources to counter multidrug resistant strains. This study investigated different solvent extracts of Prosopis spicigera (P. Spicigera), Zingiber officinale, and Trachyspermum ammi (T. ammi) to determine their efficacy against multidrug resistant microbes.. Methodology: Successive extractions of these plants were performed using a Soxhlet apparatus, using solvents with increasing polarities. Preliminary phytochemical analysis was also performed .Minimum inhibitory concentration was determined by a two-fold serial dilution method followed by determination of minimum bactericidal/fungicidal concentration. Multidrug resistant (MDR) strains of Candida albicans, Candida krusei, Candida tropicalis, Candida glabrata, Escherichia coli and reference strains of Streptococcus mutans and Streptococcus bovis ...

The studies of phenotypic adaptability and stability are important to breeding programs. They allow for the identification of genotypes with the capacity to take advantage of the stimulus of the environment, with predictable behavior patterns which respond to the environmental variations, be they under specific or broad conditions (Gauch, 1992Gauch, H.G. 1992. Statistical Analysis of Regional Yield Trials: AMMI Analysis of Factorial Designs. Elsevier, Amsterdam, The Netherlands.), or even those ampler and general genotypes with little sensitivity to environmental variation. However, when a substantial number of genotypes are evaluated, the interpretation of the parameters of adaptability and stability becomes laborious, requiring considerable time on the part of the breeder and can lead to misunderstandings. In this case, automation of genotype classification by fuzzy logic is a viable option (Carneiro et al., 2018Carneiro, V.Q.; Prado, A.L.; Cruz, C.D.; Carneiro, P.C.S.; Nascimento, M.; ...

Psoralen synthase catalyzes a pivotal reaction in this pathway and the reaction mechanism has been elucidated and shown to differ from that previously proposed by Birch et al.95 Cell culture systems inducible for the accumulation of coumarins provided a major breakthrough for the investigation of the enzymology, and these systems will certainly be helpful in tackling the still controversial course of ortho-hydroxylation and/or cyclization reactions of cinnamate and 4-coumarate as well as the classification of the enzyme introducing the 8-hydroxyl function in furanocoumarins. Values are given as mean ± SD of six experiments in each group. However, as in the case of benzoic acids, heating in quinoline in the presence of Cu or its salts led in general to better results.
