Behavioural disturbances associated with hyperdopaminergia in dopamine-transporter knockout mice. (25/1163)

Mice lacking the dopamine transporter (DAT-/-) are characterized by high extracellular dopamine levels and spontaneous hyperlocomotion. We performed a detailed analysis of the behavioural phenotype of DAT-/- mice in order to identify other behavioural impairments associated with the hyperdopaminergic tone of these mutant mice. In particular, we investigated locomotor activity, exploration, and social and maternal behaviours, which are known to be regulated by dopamine. DAT-/- mice were easily aroused by novelty and always responded with hyperlocomotion, which interfered with habituation to the testing environment, exploratory behaviour in an open field and the coping response to forced swimming stress. Social behaviours such as interaction with an unknown congener or aggressiveness were not modified in DAT-/- mice compared with DAT+/- and DAT+/+ mice, although the maternal behaviour of mutant females was severely disturbed. Haloperidol and clozapine reversed the hyperactivity in DAT-/- mice, with a rightward shift of the dose-response curve compared with control animals, suggesting a dopamine-mediated effect. These results emphasize the role of dopamine regulation in locomotion, exploration and maternal behaviours and suggest that mice with a genetic deletion of DAT may represent a useful model to elucidate the altered behavioural processes accompanying pathological conditions associated with hyperdopaminergic function.  (+info)

Down-regulation of hypothalamic corticotropin-releasing hormone messenger ribonucleic acid (mRNA) precedes early-life experience-induced changes in hippocampal glucocorticoid receptor mRNA. (26/1163)

Early-life experiences, including maternal interaction, profoundly influence hormonal stress responses during adulthood. In rats, daily handling during a critical neonatal period leads to a significant and permanent modulation of key molecules that govern hormonal secretion in response to stress. Thus, hippocampal glucocorticoid receptor (GR) expression is increased, whereas hypothalamic CRH-messenger RNA (mRNA) levels and stress-induced glucocorticoid release are reduced in adult rats handled early in life. Recent studies have highlighted the role of augmented maternal sensory input to handled rats as a key determinant of these changes. However, the molecular mechanisms, and particularly the critical, early events leading from enhanced sensory experience to long-lasting modulation of GR and CRH gene expression, remain largely unresolved. To elucidate the critical primary genes governing this molecular cascade, we determined the sequence of changes in GR-mRNA levels and in hypothalamic and amygdala CRH-mRNA expression at three developmental ages, and the temporal relationship between each of these changes and the emergence of reduced hormonal stress-responses. Down-regulation of hypothalamic CRH-mRNA levels in daily-handled rats was evident already by postnatal day 9, and was sustained through postnatal days 23 and 45, i.e. beyond puberty. In contrast, handling-related up-regulation of hippocampal GR-mRNA expression emerged subsequent to the 23rd postnatal day, i.e. much later than changes in hypothalamic CRH expression. The hormonal stress response of handled rats was reduced starting before postnatal day 23. These findings indicate that early, rapid, and persistent changes of hypothalamic CRH gene expression may play a critical role in the mechanism(s) by which early-life experience influences the hormonal stress-response long-term.  (+info)

Problem-solving abilities among mothers of infants with failure to thrive. (27/1163)

OBJECTIVE: Maternal problem-solving abilities, as they related to specific child-rearing situations, were examined and compared among mothers of infants with failure to thrive (FTT) and a matched group of comparison mothers. METHODS: Participants were 37 mothers of children diagnosed with FTT and 37 mothers with normally growing children matched on three child variables and five maternal variables. Participants were administered a means-ends problem-solving measure specific to parents of young children and measures of language ability, depression, negative affect, and stressful life events. RESULTS: Multivariate analysis of covariance results supported our main study hypotheses that mothers of infants with FTT would generate fewer problem-solving strategies that would be judged of poorer quality (i.e., less likely to result in positive outcomes) than mothers of healthy, normally growing infants. There were no significant associations obtained among problem-solving variables and individual difference variables (e.g., depression, negative affect, and stressful life events). CONCLUSIONS: Limited maternal problem-solving abilities may contribute to FTT by interfering directly with the quality of nurturance, feeling, and caloric intake the child receives. Recommendations are made for future research and interventions with mothers of children with FTT.  (+info)

Increased proliferative activity and programmed cellular death in the turkey hen pituitary gland following interruption of incubation behavior. (28/1163)

Incubation behavior or broodiness in turkey hens is characterized by ovarian regression, hyperprolactinemia, and persistent nesting. Nest-deprivation of incubating turkey hens results in disruption of broodiness accompanied by a precipitous decline in plasma prolactin (PRL) concentrations. The objective of the present study is to examine cellular changes in the pituitary gland associated with nest-deprivation for 0, 1, 2, 3, 4, or 7 days. Bromodeoxyuridine (BrdU) was administered prior to kill to study proliferative activity. Pituitary tissue sections were immunostained using turkey growth hormone (GH) antibody, and/or chicken PRL peptide antibody, and BrdU antibody. Plasma PRL concentrations declined significantly following nest-deprivation for 1 or more days. The midsagittal pituitary area immunoreactive (ir) to GH was significantly increased while that of PRL was significantly decreased following nest-deprivation for 2 or more days. Terminal deoxy-UTP nick end labeling and PRL-immunostaining revealed an abundance of apoptotic nuclei in both cephalic and caudal lobes of the anterior pituitary gland, suggestive of programmed cellular death of lactotrophs in the pituitary gland of hens nest-deprived for 2 or more days. Mammosomatotrophs were abundant in hens nest-deprived on Day 0 but were absent in hens nest-deprived for 1 or more days. Proliferating (BrdU-ir) cells were significantly abundant in the pituitary cephalic and caudal lobes following nest-deprivation for 1 or more days but were absent on Day 0 or in laying hens. Dual-labeling studies indicated that most of the BrdU-ir nuclei in the caudal lobe were not colocalized in somatotrophs in hens nest-deprived for 1-4 days but did colocalize with GH following 7 days of nest-deprivation. In conclusion, nest-deprivation of incubating turkey hens results in 1) a precipitous decline in plasma PRL concentration, 2) programmed cell death of lactotrophs, 3) disappearance of mammosomatotrophs, 4) increased proliferative activity of pituitary cells, and 5) recruitment of somatotrophs arising primarily from mitosis of nonsomatotrophic cells.  (+info)

Effects of repeated cross-fosterings on preweaning behavior and growth performance of piglets and on maternal behavior of sows. (29/1163)

Extensive cross-fostering is widely used in early-weaning units in order to standardize and improve body weight at weaning. However, young suckling piglets develop teat fidelity and repeated cross-fosterings could go against this behavior. This experiment was therefore conducted to compare the behavior and growth of 13 control and 14 fostered litters. Once every 3 d (from d 1 to 16 of lactation), all piglets were weighed and three piglets were switched between two fostered litters. Their unfostered littermates were called residents. Behavior was recorded for 2 h after weighing and(or) adoption and during one nursing period 24 h later. Fights were more frequent (P < 0.05) in treated than in control litters during and between nursings at all ages of adoption except d 1. Most fights occurred between resident and fostered piglets (P < 0.001), except at d 1. Fights during nursing were still more frequent in treated than in control litters 24 h after adoption (P < 0.02), except at d 1 and 16. More piglets had skin lacerations in treated than in control litters except at d 1 (P < 0.05), and scratches were more frequent in fostered than in resident piglets at d 1 (P = 0.07), 7, 13, and 16 (P < 0.01). At all ages except d 1, failed nursings and snaps at piglets were more frequent in fostered than in control litters (P < 0.05), most snaps being directed at adopted piglets (P < 0.001). In fostered litters, sows spent 15 to 30% less time lying on their sides at d 4, 7, 13, and 16 (P < 0.05). Finally, adopted piglets were 13% lighter than controls at weaning (P < 0.001), whereas residents were heavier than adopted piglets (P < 0.05) but lighter than controls (P = 0.1). These data demonstrate that cross-fostering done repeatedly during lactation is stressful for piglets and sows and does not improve body weight at weaning.  (+info)

Nesting behavior of the swallow-tailed hummingbird, Eupetomena macroura (Trochilidae, Aves). (30/1163)

An August or winter nestling of Eupetomena macroura was fed only every 40-50 min for at least 24 days in the nest, with fewer feedings at midday. As in other hummingbirds, it was brooded only the first week or two, and left alone even on cool nights after 12 days, probably due to the small nest size. The female attacked birds of many non-nectarivore species near the nest, in part probably to avoid predation. Botfly parasitism was extremely high, as in some other forest-edge birds.  (+info)

Parental care in the buffy-tufted-ear marmoset (Callithrix aurita) in wild and captive groups. (31/1163)

Studies on cooperative care of offspring in callitrichid primates are biased in favor of observations in captivity. In the wild, however, individuals have to deal with environmental pressures, which may influence their social behavior. We compared the individual effort attributed to parental care offered by members of a wild group (couple, plus a subadult helper) and two captive groups (A: couple, plus an subadult helper, B: couple, plus four adult helpers) of the buffy-tufted-ear marmoset, Callithrix aurita, from weeks 1-12 after the infants' birth. The carrier (breeding male and female or helper) and the infant's feeding (food sharing and foraging for food) were recorded. Up to week four, while the wild breeding pair shared infant carrying at similar proportions, the male from captive group A carried 100% of the time. Adult helpers from group B were the main carriers. Carrying behavior extended up to week 12 only in the wild group. Food provisioning to the infant was observed earlier in the groups wild and A, but general proportion of feeding records was lower in the wild than in captivity. Energetic cost of travelling and searching for food may be associated with equal division of carrying behavior by the wild breeding pair. Higher proportions of carrying in the groups wild and B may have delayed the development of the infants' motor skills required in foraging. Our data agree with previous studies: the father's lower investment in carrying when adult helpers are present and lower contribution of subadult non-reproductive members.  (+info)

Egg colour matching in an African cuckoo, as revealed by ultraviolet-visible reflectance spectrophotometry. (32/1163)

Despite major differences between human and avian colour vision, previous studies of cuckoo egg mimicry have used human colour vision (or standards based thereon) to assess colour matching. Using ultraviolet-visible reflectance spectrophotometry (300-700 nm), we measured museum collections of eggs of the red-chested cuckoo and its hosts. The first three principal components explained more than 99% of the variance in spectra, and measures of cuckoo host egg similarity derived from these transformations were compared with measures of cuckoo host egg similarity estimated by human observers unaware of the hypotheses we were testing. Monte Carlo methods were used to simulate laying of cuckoo eggs at random in nests. Results showed that host and cuckoo eggs were very highly matched for an ultraviolet versus greenness component, which was not detected by humans. Furthermore, whereas cuckoo and host were dissimilar in achromatic brightness, humans did not detect this difference. Our study thus reveals aspects of cuckoo-host egg colour matching which have hitherto not been described. These results suggest subtleties and complexities in the evolution of host-cuckoo egg mimicry that were not previously suspected. Our results also have the potential to explain the longstanding paradox that some host species accept cuckoo eggs that are non-mimetic to the human eye.  (+info)
