An update on safety studies of SAD B19 rabies virus vaccine in target and non-target species. (9/638)

SAD B19 is an attenuated vaccine virus for oral vaccination of carnivores against rabies. The safety of SAD B19 was investigated in 16 animal species by different routes of administration. During the observation period all animals given the vaccine virus, irrespective of the route of administration, did not show any clinical signs of rabies, with the exception of certain rodent species. In these animals a low residual pathogenicity was observed, however transmission of the vaccine virus to control animals was not demonstrable. No vaccine virus could be detected in the saliva of the six mammal species examined. Furthermore, the genetical stability was shown for SAD B19 through passaging in neural tissue of dogs, foxes and mice. From the results presented here on innocuity and stability, it can be concluded that SAD B19 rabies vaccine is suitable for oral vaccination campaigns for carnivores against rabies.  (+info)

Study of restriction fragment length polymorphism analysis and spoligotyping for epidemiological investigation of Mycobacterium bovis infection. (10/638)

Restriction fragment length polymorphism (RFLP) analysis with probes derived from the insertion element IS6110, the direct repeat sequence, and the polymorphic GC-rich sequence (PGRS) and a PCR-based typing method called spacer oligonucleotide typing (spoligotyping) were used to strain type Mycobacterium bovis isolates from the Republic of Ireland. Results were assessed for 452 isolates which were obtained from 233 cattle, 173 badgers, 33 deer, 7 pigs, 5 sheep, and 1 goat. Eighty-five strains were identified by RFLP analysis, and 20 strains were identified by spoligotyping. Twenty percent of the isolates were the most prevalent RFLP type, while 52% of the isolates were the most prevalent spoligotype. Both the prevalent RFLP type and the prevalent spoligotype were identified in isolates from all animal species tested and had a wide geographic distribution. Isolates of some RFLP types and some spoligotypes were clustered in regions consisting of groups of adjoining counties. The PGRS probe gave better differentiation of strains than the IS6110 or DR probes. The majority of isolates from all species carried a single IS6110 copy. In four RFLP types IS6110 polymorphism was associated with deletion of fragments equivalent in size to one or two direct variable repeat sequences. The same range and geographic distribution of strains were found for the majority of isolates from cattle, badgers, and deer. This suggests that transmission of infection between these species is a factor in the epidemiology of M. bovis infection in Ireland.  (+info)

Association of seasonal reproductive patterns with changing food availability in an equatorial carnivore, the spotted hyaena (Crocuta crocuta). (11/638)

Reproductive seasonality was examined in an equatorial population of free-living spotted hyaenas (Crocuta crocuta) in Kenya. The study population was observed continuously for 10 years, during which time the dates of all births, conceptions, weanings, and cub deaths were recorded. Local prey abundance was estimated two to four times per month, and rainfall was recorded daily throughout the study period. Births occurred during every month of the year, but a distinct trough in births occurred from February to May. This trough occurred approximately one gestation period after the phase of the annual cycle during which prey animals were least abundant in the home range of the hyaenas, and conceptions occurred most frequently when food abundance was greatest. Neither rainfall nor cub mortality were correlated with births or conceptions. Thus, although spotted hyaenas are capable of breeding throughout the year, they exhibit a moderate degree of seasonality that most likely reflects responses to seasonal variation in energy availability.  (+info)

Seroepidemiology of Bartonella vinsonii subsp. berkhoffii infection in California coyotes, 1994-1998. (12/638)

The prevalence of antibodies to Bartonella vinsonii subsp. berkhoffii in coyotes (Canis latrans) in California ranged from 51% in central to 34% in southern and 7% in northern California. Seropositive coyotes were more likely to be from coastal than inland counties (p clustered distribution of Bartonella seropositivity in coyotes suggests that B. vinsonii subsp. berkhoffii infection is vectorborne. Further investigation is warranted to evaluate which arthropods are vectors and what the mode of transmission is from wildlife to domestic dogs and possibly humans.  (+info)

Parasitological survey on wild carnivora in north-western Tohoku, Japan. (13/638)

In the winter of 1997-1998, we collected parasitological data from 60 wild carnivora in the north-western part of Tohoku region, Japan. These included 7 foxes (Vulpes vulpes japonica), 20 raccoon dogs (Nyctereutes procyonoides viverrinus), 29 martens (Martes melampus melampus), 3 weasels (two Mustela sibirica itatsi and one M. nivalis namiyei), and one Japanese badger (Meles meles anakuma). Roundworms (Toxocara canis in foxes and Toxocara tanuki in raccoon dogs), hookworms (Ancylostoma kusimaense and Arthrostoma miyazakiense) and Molineus sp. in the small intestine were the most prevalent in foxes and raccoon dogs. In martens, Aonchotheca putorii in the stomach, Concinnum ten in the pancreatic duct, Molineus sp. and Euryhelmis costaricensis in the small intestine were the most prevalent. Collected parasites include some new helminth species for this region or Japan; the strobilar stage of Taenia polyacantha from foxes, Pygidliopsis summa from a raccoon dog, Eucoleus aerophilus, A. putorii, and Soholiphyme baturini from martens.  (+info)

Molecular characterization of rabies virus isolates from Mexico: implications for transmission dynamics and human risk. (14/638)

Twenty-eight samples from humans and domestic and wild animals collected in Mexico between 1990 and 1995 were characterized by using anti-nucleoprotein monoclonal antibodies and limited sequence analysis of the nucleoprotein gene. The variants of rabies viruses identified in these samples were compared with other isolates from Mexico and the rest of the Americas to establish epidemiologic links between cases and outbreaks and to increase the understanding of rabies epidemiology in the Western Hemisphere. Antigenic and genetic diversity was found in all samples from dogs and dog-related cases, suggesting a long-term endemic situation with multiple, independent cycles of virus transmission. Two isolates from bobcats were antigenically and genetically homologous to the rabies variant circulating in the Arizona gray fox population, indicating a wider distribution of this variant than previously reported. Rabies isolates from skunks were unrelated to any variant analyzed in this study and represent a previously unrecognized cycle of rabies transmission in skunks in Baja California Sur. Two antigenic and genetic variants co-circulating in southern and eastern Mexico were found in viruses obtained from cases epidemiologically related to vampire bats. These results serve as a baseline for the better understanding of the molecular epidemiology of rabies in Mexico.  (+info)

Molecular cytogenetic dissection of human chromosomes 3 and 21 evolution. (15/638)

Chromosome painting in placental mammalians illustrates that genome evolution is marked by chromosomal synteny conservation and that the association of chromosomes 3 and 21 may be the largest widely conserved syntenic block known for mammals. We studied intrachromosomal rearrangements of the syntenic block 3/21 by using probes derived from chromosomal subregions with a resolution of up to 10-15 Mbp. We demonstrate that the rearrangements visualized by chromosome painting, mostly translocations, are only a fraction of the actual chromosomal changes that have occurred during evolution. The ancestral segment order for both primates and carnivores is still found in some species in both orders. From the ancestral primate/carnivore condition an inversion is needed to derive the pig homolog, and a fission of chromosome 21 and a pericentric inversion is needed to derive the Bornean orangutan condition. Two overlapping inversions in the chromosome 3 homolog then would lead to the chromosome form found in humans and African apes. This reconstruction of the origin of human chromosome 3 contrasts with the generally accepted scenario derived from chromosome banding in which it was proposed that only one pericentric inversion was needed. From the ancestral form for Old World primates (now found in the Bornean orangutan) a pericentric inversion and centromere shift leads to the chromosome ancestral for all Old World monkeys. Intrachromosomal rearrangements, as shown here, make up a set of potentially plentiful and informative markers that can be used for phylogenetic reconstruction and a more refined comparative mapping of the genome.  (+info)

Anthropogenic extinction of top carnivores and interspecific animal behaviour: implications of the rapid decoupling of a web involving wolves, bears, moose and ravens. (16/638)

The recent extinction of grizzly bears (Ursus arctos) and wolves (Canis lupus) by humans from 95-99% of the contiguous USA and Mexico in less than 100 years has resulted in dramatically altered and expanded prey communities. Such rampant ecological change and putative ecological instability has not occurred in North American northern boreal zones. This geographical variation in the loss of large carnivores as a consequence of anthropogenic disturbance offers opportunities for examining the potential consequences of extinction on subtle but important ecological patterns involving behaviour and interspecific ecological interactions. In Alaska, where scavengers and large carnivores are associated with carcasses, field experiments involving sound playback simulations have demonstrated that at least one prey species, moose (Alces alces), is sensitive to the vocalizations of ravens (Corvus corax) and may rely on their cues to avoid predation. However, a similar relationship is absent on a predator-free island in Alaska's Cook Inlet and at two sites in the Jackson Hole region of the Rocky Mountains (USA) where grizzly bears and wolves have been extinct for 50-70 years. While prior study of birds and mammals has demonstrated that prey may retain predator recognition capabilities for thousands of years even after predation as a selective force has been relaxed, the results presented here establish that a desensitization in interspecific responsiveness can also occur in less than ten generations. These results affirm (i) a rapid decoupling in behaviour involving prey and scavengers as a consequence of anthropogenic-caused predator-prey disequilibriums, and (ii) subtle, community-level modifications in terrestrial ecosystems where large carnivores no longer exist. If knowledge about ecological and behavioural processes in extant systems is to be enhanced, the potential effects of recently extinct carnivores must be incorporated into current programmes.  (+info)
