Identification of schistosome-infected snails by detecting schistosomal antigens and DNA sequences. (9/36)

Cercarial shedding tests do not provide species identification of the schistosomes concerned and cannot detect prepatent schistosomal infections. We have demonstrated that both immunodetection by ELISA of schistosomal antigens in snail hemolymph, and dot hybridization of snail extracts by a DNA probe representing highly repeated sequences, proved suitable for detecting infected snails during prepatency as well as patency. A group-specific monoclonal antibody was found to be suitable for detecting Schistosoma mansoni infection in Biomphalaria sp., but not for positive identification of S. haematobium in Bulinus sp. Comparative evaluation of the diagnostic qualities, and technical aspects and cost of these tests, point to the superiority of the immunodetection approach for large scale detection of snails prepatently infected with S. mansoni. This approach is potentially useful for providing extended information on schistosome-snail epidemiology that may facilitate rapid evaluation of the danger of post-control reinfection, and help make decisions on the time and place of supplementary control measures. In this context the potential usefulness of the immunodetection or DNA probing approach for facilitating catalytic model representation of schistosome-snail epidemiology warrants further evaluation. Specific identification of S. haematobium in Bulinus by either of these approaches may be possible depending on the development of suitable antibodies or DNA probes.  (+info)

Ecological studies on the intermediate host snails and the relevance to schistosomiasis control. (10/36)

A detailed knowledge of distribution patterns of schistosome intermediate hosts and their population dynamics and factors affecting these patterns will provide useful information about the possibilities and desirability of conducting snail control measures in various transmission situations. On the basis of various case studies the association between the occurrence of human water contacts and the presence of schistosome intermediate hosts or infections in the intermediate hosts is illustrated. Other parameters affecting snail distribution patterns and density fluctuations are discussed. It is concluded that ecological studies on the intermediate hosts are extremely relevant, either to optimally apply existing control measures or to develop alternative measures of snail control, such as ecological or biological control.  (+info)

Spatial patterns of urinary schistosomiasis infection in a highly endemic area of coastal Kenya. (11/36)

Urinary schistosomiasis remains a major contributor to the disease burden along the southern coast of Kenya. Selective identification of transmission hot spots offers the potential for more effective, highly-focal snail control and human chemotherapy to reduce Schistosoma haematobium transmission. In the present study, a geographic information system was used to integrate demographic, parasitologic, and household location data for an endemic village and neighboring households with the biotic, abiotic, and location data for snail collection/water contact sites. A global spatial statistic was used to detect area-wide trends of clustering for human infection at the household level. Local spatial statistics were then applied to detect specific household clusters of infection, and, as a focal spatial statistic, to evaluate clustering of infection around a putative transmission site. High infection intensities were clustered significantly around a water contact site with high numbers of snails shedding S. haematobium cercariae. When age was considered, clustering was found to be significant at different distances for different age groups.  (+info)

Distribution patterns and cercarial shedding of Bulinus nasutus and other snails in the Msambweni area, Coast Province, Kenya. (12/36)

In the Msambweni area of the Kwale District in Kenya, an area endemic for Schistosoma haematobium, potential intermediate-host snails were systematically surveyed in water bodies associated with human contact that were previously surveyed in the 1980s. Bulinus (africanus) nasutus, which accounted for 67% of the snails collected, was the only snail shedding S. haematobium cercariae. Lanistes purpureus was the second most common snail (25%); lower numbers of Bulinus forskalii and Melanoides tuberculata were also recovered. Infection with non-S. haematobium trematodes was found among all snail species. Rainfall was significantly associated with the temporal distribution of all snail species: high numbers of Bulinus nasutus developed after extensive rainfall, followed, in turn, by increased S. haematobium shedding. Spatial distribution of snails was significantly clustered over a range of up to 1 km, with peak clustering observed at a distance of 400 meters. Water lily (Nymphaea spp.) and several aquatic grass species appeared necessary for local colonization by B. nasutus or L. purpureus.  (+info)

Environmental control of schistosomiasis through community participation in a Moroccan oasis. (13/36)

Summary Akka oasis, in the province of Tata, southern Morocco, is one of the oldest foci of urinary schistosomiasis in Morocco where transmission is still taking place. We report the results of two studies: a cross-sectional snail survey investigated the distribution of Bulinus truncatus in relation to habitat factors in the Akka traditional irrigation system. The presence of aquatic vegetation, especially Potamogeton sp. was identified as a key factor determining snail occurrence and abundance in canals, impoundments and isolated small puddles and streamlets in the Akka riverbed. In a participatory rapid appraisal, the community identified snail control as a way to reduce transmission of schistosomiasis. Without any further outside incentives, the local irrigation committee implemented repeated cleaning and vegetation removal in canals. A longitudinal study evaluated the effect of these measures on populations of B. truncatus. Snail and egg mass densities showed significant reductions after repeated vegetation clearing in the study sites. The participatory approach led to low-cost, environment-friendly schistosomiasis control measures that were effective and sustainable.  (+info)

Large-scale, polymerase chain reaction-based surveillance of Schistosoma haematobium DNA in snails from transmission sites in coastal Kenya: a new tool for studying the dynamics of snail infection. (14/36)

Levels of prepatent Schistosoma haematobium infection were monitored in intermediate host snails (Bulinus nasutus) collected from transmission sites in coastal Kenya, using a polymerase chain reaction (PCR) assay amplifying the Dra I repeated sequence of S. haematobium. The timing and number of prepatent and patent infections were determined for each site and, where the time of first appearance was clear, the minimal prepatent period was estimated to be five weeks. High, persistent, prepatency rates (range = 28-54%), indicated a significant degree of repeated area contamination with parasite ova. In contrast, rates of cercarial shedding proved locally variable, and were either low (range = 0.14-3.4%) or altogether absent, indicating that only a small proportion of infected snails reach the stage of cercarial shedding. Given the apparently strong focal effects of environmental conditions, implications of these new data are discussed regarding the estimation of local force of transmission and the design of control activities.  (+info)

Studies on vesical schistosomiasis among rural Ezza farmers in the southwestern border of Ebonyi State, Nigeria. (15/36)

Studies on vesical schistosomiasis and its snail vectors were carried out between October 2001-May 2002 among rural Ezza farmers inhabiting the southwestern border of Ebonyi State, Nigeria. The people are predominantly farmers. Of the 2,104 urine specimens examined in 10 communities, 466 (22.1%) comprising 305 (23.7%) men and 161 (19.7%) women were infected with visible haematuria as the predominant presenting symptom. Ezza people associate bloody urine with sexually transmitted diseases. There were no significant differences in the prevalence rates amongst various villages and sexes (p > 0.05). There was a gradual increase in the disease prevalence as the subjects' age increases. About 78.3% of the infected persons are aged 0-20 years. Statistical analysis revealed that the prevalence, intensity and visible haematuria were significantly more ( p < 0.05) in subjects under the age of 20 than subjects above 20. Among the infected population, 183 (39.3%) and 283 (60.7%) were excreting 50 eggs/10 ml urine and above 50 eggs/10 ml urine respectively. Lack of visible haematuria is a more valid indicator of the absence of vesical schistosomiasis. Of the various snails collected during malacological survey, mainly B. globosus were infected. Spectroscopic analysis of benzylidene anabaseine complexes with acetylcholine binding proteins as models for ligand-nicotinic receptor interactions. (16/36)

The discovery of the acetylcholine binding proteins (AChBPs) has provided critical soluble surrogates for examining structure and ligand interactions with nicotinic receptors and related pentameric ligand-gated ion channels. The multiple marine and freshwater sources of AChBP constitute a protein family with substantial sequence divergence and selectivity in ligand recognition for analyzing structure-activity relationships. The purification of AChBP in substantial quantities in the absence of a detergent enables one to conduct spectroscopic studies of the ligand-AChBP complexes. To this end, we have examined the interaction of a congeneric series of benzylidene-ring substituted anabaseines with AChBPs from Lymnaea, Aplysia, and Bulinus species and correlated their binding energetics with spectroscopic changes associated with ligand binding. The anabaseines display agonist activity on the alpha7 nicotinic receptor, a homomeric receptor with sequences similar to those of the AChBPs. Substituted anabaseines show absorbance and fluorescence properties sensitive to the protonation state, relative permittivity (dielectric constant), and the polarizability of the surrounding solvent or the proximal residues in the binding site. Absorbance difference spectra reveal that a single protonation state of the ligand binds to AChBP and that the bound ligand experiences a solvent environment with a high degree of polarizability. Changes in the fluorescence quantum yield of the bound ligand reflect the rigidification of the ring system of the bound ligand. Hence, the spectral properties of the bound ligand allow a description of the electronic character of the bound state of the ligand within its aromatic binding pocket and provide information complementary to that of crystal structures in defining the determinants of interaction.  (+info)
